WEHI-3 Syngeneic Model Service for Leukemia

Fig 1: WEHI-3 Syngeneic Model for Leukemia preclinical research.

The WEHI-3 syngeneic model of leukemia propagates a mineral oil–induced murine myelomonocytic leukemia in fully immunocompetent BALB/c hosts, pairing a five-decade publication pedigree with dissemination formats that read out as organomegaly, survival and innate immune activity. Alfa Cytology delivers the complete WEHI-3 workflow—authenticated cell banking, intraperitoneal, intravenous or subcutaneous inoculation, balanced assignment, scheduled dosing and organ- and survival-based endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of WEHI-3 Syngeneic Model for Leukemia

Few cell lines carry as much history per passage as WEHI-3. In 1969, Warner, Moore and Metcalf at the Walter and Eliza Hall Institute reported a transplantable myelomonocytic leukemia that had arisen in a BALB/c mouse undergoing mineral oil injections—the regimen meant to provoke plasmacytomas instead produced a tumor of mixed monocytic and granulocytic cells with a chloroma’s greenish cast. Transplantation resolved the tumor into four sublines, two retaining the chloroma phenotype (one diploid, one tetraploid) and two non-chloroma variants (one with a hypodiploid 39-chromosome stemline), and muramidase poured out of affected animals at concentrations frequently exceeding 200 µg/mL in serum and urine. The line’s second act was molecular: WEHI-3 turned out to produce interleukin-3 constitutively through activation of the IL-3 gene by a retroviral-like intracisternal A-particle insertion, making it the founding source of murine IL-3—its conditioned medium still feeds IL-3-dependent lines such as 32D and BaF3—and its cells served as the canonical target in the 1982 bioassay that led to tumor necrosis factor’s purification. MHC-restricted antigen presentation by WEHI-3 was demonstrated as early as 1981, and the derivative WEHI-3B remains a reference for G-CSF-induced myeloid differentiation.

Time-course organ burden in the WEHI-3B syngeneic model — macroscopic spleen and liver images at day 31 across five groups, spleen weight rising from ~0.08 g to ~0.45 g (group 3) and liver weight changes, quantifying disease progression from day 5 to day 31 after WEHI-3B inoculation. (Nagarajan, Senthil, et al., 2021)

As an in vivo model, WEHI-3 is refreshingly direct: introduced into BALB/c mice by the intraperitoneal or intravenous route, the cells establish a disseminated leukemia marked by progressive splenomegaly and hepatomegaly, and the efficacy readouts are the classic ones—body, spleen and liver weights, survival, histopathology of spleen, liver and bone marrow, peripheral-blood blast burden and natural-killer-cell activity, the last of which proved decisive in the thymoquinone studies that documented reduced organ weights alongside boosted NK function. A subcutaneous solid-tumor format exists for sponsors wanting caliper-based dosing windows. The cells themselves are macrophage-like, grow semi-adherent with a doubling time of roughly 18–23 hours, express Fc and complement receptors, lysozyme and G-CSF activity, and carry a validated mouse STR profile plus SNP-array strain verification. Caveats deserve respect: a 1969-era line brings subline divergence (WEHI-3 versus WEHI-3B differ), its autocrine IL-3 can confound cytokine-addition experiments, semi-adherent harvesting demands consistency, and the model is BALB/c-bound. Within those boundaries, it remains an economical, immunocompetent mainstay for preclinical leukemia research.

Cell Line Information: WEHI-3

Key characteristics of WEHI-3 and its documented derivatives are summarized below.

Item Details
Cell line name WEHI-3 (WEHI3); ATCC TIB-68; Cellosaurus CVCL_3622; sibling derivative WEHI-3B at DSMZ ACC 26
Species / strain background Mus musculus, BALB/c; syngeneic host: BALB/c
Origin Myelomonocytic leukemia arising in a male (6-month-old) BALB/c mouse receiving mineral oil (paraffin) injections intended to induce plasmacytomas; peripheral blood origin; established 1969 (Warner, Moore & Metcalf, J Natl Cancer Inst)
Tumor type Transplantable murine myelomonocytic leukemia; chloroma-forming; mixed monocytic and granulocytic populations arising from a bipotent neoplastic stem cell
Historical sublines Four transplantation sublines: diploid and tetraploid chloroma lines, and two non-chloroma lines (one with a hypodiploid 39-chromosome stemline); serum/urine muramidase frequently >200 µg/mL
IL-3 hallmark Constitutive IL-3 production driven by an intracisternal A-particle (IAP) insertion activating the IL-3 gene; the founding source of murine IL-3 — conditioned medium supports IL-3-dependent lines (32D, BaF3); also produces lysozyme and G-CSF activity
Other historical roles Canonical target line in the 1982 bioassay that guided tumor necrosis factor purification; MHC-restricted antigen presentation demonstrated in 1981; WEHI-3B responsive to G-CSF-induced differentiation
Morphology / growth Macrophage-like, semi-adherent (adherent plus suspension); doubling time ~18–23 hours; expresses Fc and complement (C3) receptors; weak ADCC effector
Culture conditions IMDM + 10% FBS + 0.05 mM 2-mercaptoethanol (ATCC formulation) or RPMI-1640 + 10% FBS; 37 °C, 5% CO2; medium renewal every 2–3 days; biosafety level 1
Authentication resources Validated mouse STR profile (interlaboratory study, 2019); SNP-array strain-of-origin profile published
Leukemia model behavior Intraperitoneal or intravenous inoculation into BALB/c → disseminated leukemia with progressive splenomegaly and hepatomegaly; readouts: body/spleen/liver weights, survival, histopathology, peripheral-blood blasts, NK-cell activity (documented in thymoquinone studies); subcutaneous solid-tumor format available
Considerations 1969-era line with documented subline divergence (WEHI-3 vs WEHI-3B); autocrine IL-3 can confound cytokine-addition designs; semi-adherent growth requires consistent harvest; BALB/c-restricted
Representative applications Anti-leukemic drug screening, differentiation-therapy research (G-CSF responsiveness), NK-cell and innate-immunity studies, IL-3/cytokine biology, hematopoietic dissemination models, combination chemotherapy evaluation

Workflow of WEHI-3 Syngeneic Model Construction

Construction of the WEHI-3 syngeneic model is an inoculation workflow whose quality rests on verified cells, vigorous BALB/c recipients, route-matched technique and disciplined clinical monitoring. A standard engagement proceeds as follows:

  1. Source and qualify cells: WEHI-3 stocks are cleared by mycoplasma PCR and authenticated against the published mouse STR profile; constitutive IL-3 production is documented as a functional identity check, and luciferase labeling is titrated in vitro where imaging readouts are required.
  2. Prepare recipients: BALB/c mice (typically 6–8 weeks old) complete acclimation and health screening and are individually marked before inoculation.
  3. Prepare the inoculum: log-phase cells are harvested with consistent semi-adherent technique, washed (three washes where cytokine-free designs require removal of autocrine IL-3), counted and checked for viability before resuspension in PBS.
  4. Inoculate: cells are delivered intraperitoneally or intravenously (tail vein) for disseminated leukemia, or subcutaneously into the flank where a caliper-tracked solid format is preferred, using consistent technique for each route.
  5. Assign groups: leukemia-format animals are randomized by body weight at inoculation; flank-format animals are grouped when tumors become palpable, balanced by tumor volume and body weight.
  6. Dose and follow: test articles are administered on schedule; body weight and clinical status are recorded two to three times weekly (daily near endpoints), with survival as the primary leukemia readout and caliper measurements for flank tumors; peripheral-blood smears or CBC can be added on a defined schedule.
  7. Terminate and analyze: spleen, liver and body weights are recorded; spleen, liver and bone marrow undergo histopathology; peripheral blood is scored for blast burden; splenic and circulating populations are profiled by flow cytometry (CD11b/Gr-1 lineage, NK markers); NK-cell activity assays and cytokine panels complete the package.
