4T1 Syngeneic Model Service for Breast Cancer

Fig 1: 4T1 Syngeneic Model for Breast Cancer preclinical research.

The 4T1 syngeneic model of breast cancer is a murine mammary carcinoma grown in fully immunocompetent BALB/c hosts, combining rapid local growth with aggressive, multi-organ metastasis that closely mirrors advanced human disease. Alfa Cytology delivers the complete 4T1 workflow—authenticated cell banking, subcutaneous or mammary fat-pad implantation, volume-balanced randomization, scheduled dosing and metastasis-inclusive endpoint analysis—giving sponsors decision-ready preclinical datasets tailored to their program, strictly within a preclinical scope.

Overview of 4T1 Syngeneic Model for Breast Cancer

4T1 was established from a spontaneously arising mammary tumor in a BALB/c-background mouse and is distributed by ATCC as CRL-2539; over three decades it has become arguably the most heavily used syngeneic breast cancer model in existence. The line behaves like an orthodox triple-negative-like mammary adenocarcinoma: poorly differentiated, fast-growing in vitro and in vivo, and—decisively for metastasis research—spontaneously disseminates from a primary mammary or flank site to lung, liver, bone (femur and tibia), draining lymph nodes and, in some designs, brain. A quirky genetic trait underpins a classic analytical method: the cells are naturally resistant to 6-thioguanine, so excised lungs can be digested and plated under selective pressure, and every metastatic colony that grows is a colony that arose in vivo—the quantitative clonogenic metastasis assay of Aslakson and Miller that made 4T1 famous in the early 1990s.

Immunologically, 4T1 is a demanding tumor. It is weakly immunogenic, recruits abundant myeloid-derived suppressor cells and regulatory T cells, and responds modestly at best to single-agent checkpoint blockade—which is precisely why the model dominates the preclinical literature on combination immunotherapy, radiation-plus-immunotherapy, nanoparticle and oncolytic approaches, and myeloid-targeting strategies. Practical performance is equally well documented: one million cells in the flank or mammary fat pad produce measurable tumors within about a week and lethal metastatic burden within four to six weeks without intervention, while luciferase-tagged 4T1-luc variants add whole-body bioluminescence tracking of primary growth and organ colonization. The trade-offs are brisk kinetics that compress dosing windows, a suppressive microenvironment that can blunt single-agent immunotherapy, and murine mammary biology that requires appropriate interpretation—all manageable with disciplined design. Within those boundaries, no syngeneic model offers a deeper publication history or a wider methodological toolkit for preclinical breast cancer research.

Cell Line Information: 4T1

Key characteristics of 4T1 and its commonly used derivatives are summarized below.

Item Details
Cell line name 4T1 (murine mammary carcinoma); ATCC CRL-2539
Species / strain background Mus musculus; syngeneic host: BALB/c (line originates from a spontaneous mammary tumor in a BALB/c-background mouse)
Tumor type Poorly differentiated murine mammary adenocarcinoma with a triple-negative-like phenotype
Origin Spontaneous mammary tumor in a BALB/c-background mouse
Morphology Adherent epithelial cells
Culture conditions RPMI-1640 + 10% FBS + 1% penicillin/streptomycin; 37 °C, 5% CO2, humidified incubator
Growth kinetics Rapid in vitro expansion; 1 × 106 cells implanted in flank or mammary fat pad yield measurable tumors within ~1 week; lethal metastatic burden in 4–6 weeks without intervention
Metastatic pattern Spontaneous spread to lung, liver, bone (femur/tibia), draining lymph nodes and, in dedicated designs, brain; tail-vein inoculation models rapid multi-organ colonization
6-thioguanine resistance Naturally 6-TG resistant — the basis of the quantitative clonogenic lung-metastasis assay (plate digested lungs under 6-TG selection and count colonies)
Immune profile Weakly immunogenic with abundant MDSC and Treg infiltration; modest response to single-agent checkpoint blockade — a reference model for combination immunotherapy
Labeled variants 4T1-luc (firefly luciferase) for whole-body bioluminescence tracking; GFP-expressing derivatives in routine use
Typical inoculum Subcutaneous or mammary fat pad: 1 × 106 cells (range 1 × 104–1 × 106, tuned to study length); tail-vein: 2.5 × 105–5 × 105 cells
Considerations Brisk growth compresses treatment windows; suppressive microenvironment can blunt monotherapy; murine mammary biology requires careful interpretation
Representative applications Combination immunotherapy, radiation-plus-immunotherapy, nanoparticle and oncolytic platforms, myeloid-targeting strategies, anti-metastatic screening, bone- and brain-metastasis formats, triple-negative breast cancer-oriented research

Our Services

With in vivo oncology specialists guiding each stage and pathology, bioluminescence imaging and immune-profiling capabilities in-house, Alfa Cytology adapts every 4T1 engagement to the sponsor’s hypothesis—calibrating inoculum size, implantation site, dosing cadence and metastatic readouts so the model’s famous aggressiveness resolves into clean, decision-grade preclinical data.

Workflow of 4T1 Syngeneic Model Construction

Construction of the 4T1 syngeneic model is an implantation workflow whose quality depends on verified cells, vigorous recipients, standardized inoculation and a disciplined measurement cadence. A standard engagement runs as follows:

  1. Source and qualify cells: 4T1 or 4T1-luc stocks are cleared by mycoplasma PCR and identity verification; luciferase expression is titrated in vitro with D-luciferin before expansion.
  2. Prepare recipients: BALB/c mice (typically 6–8 weeks old, females for mammary-context studies) complete acclimation and health screening and are individually marked before implantation.
  3. Prepare the inoculum: log-phase cells are harvested, washed, counted and resuspended in PBS (commonly 1 × 107/mL for a 100 µL dose).
  4. Implant: 1 × 106 cells are injected subcutaneously into the flank, or orthotopically into the mammary fat pad where organ context is required, using consistent anatomical positioning for reproducible caliper geometry.
  5. Randomize: when tumors reach palpability (around day 6–8), animals are assigned to groups balanced by tumor volume and body weight.
  6. Dose and follow: test articles are administered on schedule; tumors are measured with calipers two to three times weekly (volume typically π/6 × length × width × height), with bioluminescence substituted or added for 4T1-luc cohorts; body weight and clinical status are logged in parallel.
  7. Terminate and analyze: tumors are resected and weighed; lungs, liver, lymph nodes and bones are inspected for metastasis, with digested lungs plated under 6-thioguanine selection for clonogenic colony counting; tissues then enter histopathology, immunohistochemistry, flow-cytometric immune profiling and molecular analysis.

Case Study-4T1 Syngeneic Model Development

Alfa Cytology keeps an active file of preclinical case studies on the 4T1 platform: establishment-phase optimization of inoculum and implant site, in vivo growth tracking by caliper or bioluminescence, combination immunotherapy and myeloid-targeting trials, and multi-organ metastasis quantification by ex vivo imaging, histology and 6-thioguanine clonogenic counting. Representative growth curves, imaging series and endpoint summaries can be shared upon request and will be updated as new programs wrap up.
