RPMI 8226 / RPMI 8226-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: RPMI 8226 / RPMI 8226-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The RPMI 8226 / RPMI 8226-luc orthotopic mouse model establishes the founding human multiple myeloma line — established in 1966, lambda-light-chain secreting, and the ancestral substrate of an entire catalogued drug-resistant family — within the bone marrow and tissues of immunodeficient mice. Alfa Cytology delivers this six-decade reference with authenticated banking, subcutaneous or systemic luciferase-imaging configurations, matched parental and multidrug-resistant derivative cohorts, and benchmark arms anchored to the published bortezomib and cellular immunotherapy record, so your cytotoxic, proteasome, exportin, and resistance-overcoming strategies are tested on the line against which the MM pharmacology literature was originally written.

Overview of RPMI 8226 / RPMI 8226-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

RPMI 8226 was established in 1966 by George Moore and colleagues from the peripheral blood of a 61-year-old man with myeloma at diagnosis, and within a year its first publication documented what made it extraordinary: a hematopoietic cell line secreting free lambda immunoglobulin light chains — no gamma, alpha, or mu heavy chains, no intact immunoglobulin — a pure light-chain biology that remains its signature today. The line's phenotype is correspondingly terminal-plasma-cell: EBV negative, CD19 negative, very low CD20, CD28 and CD38 positive, high PCA-1 and Ki-1 (CD30) expression, no HLA class II antigens — and its growth carries an operational quirk that catalogues document plainly: cells grow mostly in suspension but may adhere up to 50%, requiring collection of both fractions at passage. As a member of the LL-100 blood cancer panel, its whole-exome and RNA-sequencing data are publicly available, and its doubling time runs roughly 1.5 days (32–70 hours across curated lots).

Two bodies of validation give the model its depth. The first is the Dalton multidrug-resistant family generated at Moffitt from this parent: 8226/Dox6 and 8226/Dox40 (doxorubicin-selected, MDR-1 upregulated), 8226/LR5 (melphalan-selected), 8226/MR20 (mitoxantrone-selected), and bortezomib-selected 8226/B25 — a multidrug-resistant constellation against which HSP90 inhibitors, selinexor–doxorubicin combinations, and next-generation proteasome strategies have all been benchmarked. The second is the in vivo record: RPMI 8226 is tumorigenic across NOD-SCID, NSG, and NOG hosts with standard subcutaneous growth curves and available bortezomib treatment data, and its luciferase configuration carried the first bioluminescence-imaging myeloma models for graft-versus-myeloma cellular immunotherapy testing (Rozemuller et al., Haematologica, 2008). Orthotopically, systemic delivery models marrow-homing disease while subcutaneous benchmarking anchors efficacy comparisons, and the RPMI 8226-luc variant enables quantitative whole-body bioluminescence in vivo with monoclonal quality control. For programs that need a light-chain-producing parent matched against a library of resistant derivatives, no MM model offers more inherited experimental infrastructure.

Cell Line Information: RPMI 8226 / RPMI 8226-luc

RPMI 8226 (ATCC CCL-155; DSMZ ACC 402; JCRB0034; RRID CVCL_0014) is curated by three international repositories with identity and sequence data on record, and it is a bona fide human myeloma line of the mature plasma-cell phenotype. Growth in vitro is in 90% RPMI-1640 with 10% heat-inactivated fetal bovine serum, seeded at 0.4–0.5 × 106 cells/mL and split 1:2 every three days, collecting both floating and adherent fractions (adherent cells release by tapping, rubber policeman, or brief TNE treatment — trypsin usually unnecessary). The RPMI 8226-luc derivative adds firefly luciferase for quantitative imaging in vivo, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence.

Our Services

The 8226 family's power is its inheritance: a light-chain-secreting parent, five catalogued resistant derivatives, three-repository authentication, and public sequencing — but realizing that inheritance requires partial-adherence-aware handling and parent–derivative pairing discipline. Alfa Cytology runs RPMI 8226 programs with both-fraction collection SOPs (tapping, rubber policeman, or TNE rather than routine trypsin), batch-verified lambda secretion and LL-100 identity, monoclonal luciferase quality control, matched Dox/LR/MR/B25 resistant cohorts where resistance is the question, and endpoint panels that pair tumor kinetics in vivo with serum lambda light-chain quantification — so six decades of accumulated reference data remain usable in your study.

Workflow of RPMI 8226 / RPMI 8226-luc Orthotopic Mouse Model Construction

RPMI 8226 construction follows established human MM xenograft practice, with two refinements: both-fraction culture handling and parent–resistant-derivative pairing. The workflow below covers preparation, the two validated implantation configurations, and light-chain-anchored endpoints.

  1. Both-Fraction Cell Expansion and Quality Control: RPMI 8226 or RPMI 8226-luc cells are expanded in vitro in 90% RPMI-1640 + 10% heat-inactivated FBS, seeded at 0.4–0.5 × 106 cells/mL and split 1:2 every three days, collecting floating and adherent fractions (tapping, rubber policeman, or TNE; trypsin usually unnecessary); batches are confirmed mycoplasma-negative, STR-authenticated, and verified for lambda secretion and CD38 expression, with monoclonal luciferase populations confirmed for tagged lines.
  2. Host Selection and Acclimatization: NOD-SCID, NSG, or NOG mice, typically 6–8 weeks of age, are acclimatized under SPF conditions; disseminated designs may use sub-lethal irradiation to promote marrow engraftment.
  3. Configuration Selection and Inoculation: The study design selects subcutaneous flank implantation (typically 5–10 × 106 cells with Matrigel where required) for localized benchmarking, or systemic intravenous delivery (5–10 × 106 cells in ~200 µL) for marrow-homing disease, with implantation day recorded for the dosing calendar.
  4. Engraftment Verification and Randomization: RPMI 8226-luc cohorts are imaged by bioluminescence in vivo after D-luciferin administration; systemic configurations are confirmed by long-bone and vertebral marrow signal, subcutaneous arms by caliper, and animals are randomized at defined flux or volume thresholds.
  5. Treatment Administration with Parent–Derivative Pairing: Test agents are dosed on study-defined schedules — bortezomib or next-generation proteasome inhibitors, cytotoxics, exportin or HSP90 agents — with matched parental and Dox6/Dox40/LR5/MR20/B25 derivative arms where multidrug-resistance reversal is the question.
  6. Longitudinal Monitoring with Light-Chain Anchoring: Disease burden is tracked weekly by bioluminescence quantification in vivo and by serial serum lambda light-chain quantification — the model's signature non-invasive biomarker correlating with tumor load.
  7. Survival Follow-Up: Animals are followed to humane endpoints with Kaplan–Meier analysis, and disseminated designs monitor for hind-limb impairment from vertebral disease as a clinical signal.
  8. Endpoint Analysis and Reporting: At termination, tumors and femoral/tibial marrow are harvested; human myeloma cells are enumerated by CD38/CD138 or lambda idiotype flow cytometry, bone and soft-tissue infiltration is documented histologically, tissues are processed for H&E, MDR-1 (where derivative arms are included), and cleaved caspase-3; ex vivo bioluminescence maps signal distribution; and imaging, light-chain, flow cytometry, and survival data are compiled into the final report.

Case Study-RPMI 8226 / RPMI 8226-luc Orthotopic Mouse Model Development

RPMI 8226 is the most historically loaded human myeloma line — from the 1967 light-chain discovery through the Dalton resistant-family era to modern proteasome, exportin, and cellular-immunotherapy benchmarking — and the platform has carried a broad range of Alfa Cytology client programs across those same axes. Across these programs, readouts have combined longitudinal bioluminescence in vivo, serum lambda light-chain kinetics, and matched parental–resistant-derivative comparisons with survival analysis. Anonymized case summaries and representative deliverable formats are available on request.
