TC-1 Syngeneic Model Service for Cervical Cancer

Fig 1: TC-1 Syngeneic Model for Cervical Cancer preclinical research.

The TC-1 syngeneic model service for cervical cancer is an immune-competent murine platform in which HPV16 E6/E7- and c-Ha-ras-transformed TC-1 cells are engrafted into syngeneic C57BL/6 mice, providing the reference system for evaluating HPV therapeutic vaccines, checkpoint blockade, and combination immunotherapy against HPV-positive tumors. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated TC-1 / TC-1-Luc expansion and banking, standardized flank implantation, therapeutic-vaccine and checkpoint dosing schedules, E7-specific immune monitoring by tetramer staining, ELISPOT, and intracellular cytokine staining, and full endpoint pathology — all configured around each sponsor’s vaccine platform, regimen, and immunological endpoint.

Overview of TC-1 Syngeneic Model for Cervical Cancer

TC-1 was purpose-built for immunology: derived at the University of Pennsylvania by Lin and colleagues and described in 1996 in Cancer Research, the line was generated by co-transforming primary lung epithelial cells from a C57BL/6 mouse with the HPV16 E6 and E7 oncogenes together with an activated c-Ha-ras oncogene. The design logic was exact — engineer a tumor that stably expresses the E7 oncoprotein, the dominant tumor antigen of HPV-driven cervical cancer, while growing in a fully immunocompetent syngeneic host where antigen presentation, T-cell priming, and effector responses all remain intact. The line is MHC class I positive (H-2Kb, H-2Db), presents well-defined E7 CD8⁺ T-cell epitopes, and retains E6/E7 expression through repeated passage, properties that made it the single most widely used preclinical testbed for HPV therapeutic vaccine development — from peptide and protein vaccines to DNA, dendritic-cell, and bacterial-vectored platforms.

In vivo, TC-1 cells grow briskly in C57BL/6 mice — flank tumors are typically palpable within about a week of implantation — yet remain immunologically tractable: antibody-mediated CD8⁺ T-cell depletion abolishes vaccine-induced control, formally demonstrating the CD8 dependence that the model was built to interrogate. Established tumors respond to therapeutic vaccination with growth inhibition or regression, and the model is where the now-canonical finding that vaccine efficacy is amplified by checkpoint blockade was established; cytokine therapies (IL-12, IL-2), adoptive cell transfer, and bispecific or innate-activating combinations have all been benchmarked on this line. The luciferase-tagged derivative TC-1-Luc, validated for whole-animal imaging, extends the platform to quantitative, longitudinal tumor tracking, while the model’s relevance extends beyond cervix to other HPV-driven malignancies. For sponsors developing E7-directed immunotherapies, TC-1 remains the mandatory, universally accepted first-line syngeneic system.

Cell Line Information: TC-1

The table below consolidates the key biological, cultural, and modeling characteristics of the TC-1 line and its luciferase-labeled derivative TC-1-Luc.

Parameter Details
Cell line designation TC-1 (parental line); TC-1-Luc (luciferase-labeled derivative validated for whole-animal imaging)
Species / genetic background Mouse (Mus musculus), C57BL/6 inbred background; syngeneic in C57BL/6 immunocompetent hosts
Origin Derived by Lin et al. at the University of Pennsylvania (Cancer Research 56:21–26, 1996) from primary lung epithelial cells of a C57BL/6 mouse co-transformed with HPV16 E6, HPV16 E7, and an activated c-Ha-ras oncogene
Depository / catalogue ATCC CRL-2499 (parental TC-1)
Tumor relevance Preclinical model of HPV16-positive cervical cancer; equally applied to other HPV-associated malignancies including head-and-neck and anal cancers
Oncogenes carried HPV16 E6 and E7 (stable expression retained through passage); activated c-Ha-ras
Antigen presentation MHC class I positive (H-2Kb, H-2Db); presents well-characterized HPV16 E7 CD8⁺ T-cell epitopes (including the E7 11–20 epitope), enabling tetramer, ELISPOT, and intracellular-cytokine monitoring of E7-specific immunity
Growth properties Adherent epithelial-like cells growing robustly in standard culture; flank tumors typically palpable within approximately seven days of implantation
Recommended culture conditions RPMI-1640 supplemented with 10% fetal bovine serum with penicillin/streptomycin at 37 °C in a humidified 5% CO₂ atmosphere
Standard implantation practice Published studies implant approximately 2 × 105 to 1 × 106 cells subcutaneously into the flank of C57BL/6 mice; therapeutic treatment typically begins once tumors reach approximately 5 mm (commonly days 7–14 post-implantation), supporting both prophylactic and therapeutic vaccine designs
Documented in vivo responses Therapeutic vaccination inhibits or regresses established tumors; vaccine efficacy is amplified by anti-PD-1/PD-L1 checkpoint blockade; antibody-mediated CD8⁺ depletion abolishes control, formally demonstrating CD8 dependence; responsive to IL-12, IL-2, and adoptive cell therapies
Host strains C57BL/6 immunocompetent mice (syngeneic); selected studies use knockout or humanized variants to dissect specific immune mechanisms
Key research applications HPV therapeutic vaccine development and benchmarking (peptide, protein, DNA, dendritic-cell, and bacterial-vectored platforms); checkpoint blockade single-agent and combination studies; cytokine and innate-activating combinations; adoptive cell therapy evaluation; immune-mechanism studies requiring a defined, E7-positive syngeneic tumor

Our Services

Few models demand as much immunological craft as an E7-positive syngeneic tumor, and Alfa Cytology brings exactly that: authenticated TC-1 / TC-1-Luc banks, standardized C57BL/6 implantation, precisely scheduled vaccine-and-checkpoint regimens, and E7-specific immune analytics alongside endpoint pathology — all run as one accountable preclinical program.

Workflow of TC-1 Syngeneic Model Construction

Construction follows the established flank-engraftment methodology used across the HPV vaccine field: TC-1 cells are expanded under authenticated conditions, implanted subcutaneously into C57BL/6 mice, grown to a standardized baseline, randomized, treated per regimen, and analyzed with E7-specific immune endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: TC-1 or TC-1-Luc cells are STR/identity-verified, confirmed mycoplasma-negative, and expanded into low-passage working banks under standardized RPMI-1640 culture conditions.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at the study-specified dose of approximately 2 × 105 to 1 × 106 cells in 100 μL of PBS or a Matrigel-supported suspension.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane, and TC-1 cells are implanted subcutaneously into the flank using a fine-gauge needle.
  4. Establishment monitoring and randomization: Tumors are monitored by palpation and caliper measurement from approximately day 5; animals are randomized into treatment arms once tumors reach approximately 5 mm, balancing baseline volumes across groups.
  5. Vaccine and therapy administration: Experimental arms — therapeutic or prophylactic vaccine regimens, anti-PD-1/PD-L1/CTLA-4 checkpoint inhibitors, cytokine therapies, adoptive cell transfer, and combinations — are administered per study design with scheduled body-weight and toxicity monitoring.
  6. E7-specific immune monitoring: Peripheral blood, spleen, and tumor-infiltrating lymphocytes are sampled on a study-defined schedule for E7-specific CD8⁺ T-cell quantification by tetramer staining, IFN-γ ELISPOT, and intracellular cytokine staining, alongside tumor-growth tracking.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth curves, survival, and final tumor weight with tumor-infiltrating-lymphocyte profiling by multi-parametric flow cytometry and histopathology (H&E with CD8, Ki67, and marker-specific immunohistochemistry); mechanism studies may include immune-subset depletion arms to establish effector dependence.
