OVCAR-3 / OVCAR-3-luc Orthotopic Mouse Model Service for Ovarian Cancer

Fig 1: OVCAR-3 / OVCAR-3-luc Orthotopic Mouse Model for Ovarian Cancer preclinical research.

The OVCAR-3 / OVCAR-3-luc orthotopic mouse model recapitulates human ovarian carcinoma within its native anatomical niche, enabling more translationally relevant preclinical evaluation of anti-tumor therapies. Alfa Cytology delivers end-to-end construction and study support for this model — from luciferase-tagged cell preparation and surgical orthotopic implantation to longitudinal in vivo imaging, efficacy testing, and histopathological readouts — giving researchers a dependable platform for ovarian cancer drug development.

Overview of OVCAR-3 / OVCAR-3-luc Orthotopic Mouse Model for Ovarian Cancer

OVCAR-3 is a human ovarian adenocarcinoma cell line originally established in 1982 by T.C. Hamilton and colleagues from the malignant ascites of a patient with progressive ovarian adenocarcinoma. When implanted orthotopically — directly into the ovarian bursa of immunodeficient mice — the cells give rise to tumors that grow within the ovarian microenvironment rather than at a subcutaneous site, closely mimicking the peritoneal dissemination pattern, ascites formation, and metastatic spread characteristic of advanced-stage human ovarian cancer. Because the tumor interacts with its tissue of origin, this model preserves key aspects of tumor–stroma crosstalk that are often lost in ectopic models.

Humanized mice bearing peritoneal OVCAR-3 Luc tumors treated with pembrolizumab or PBS, showing longitudinal whole-body BLI at days 4/11/18/25 with group bioluminescence curves, excised tumors and volumes, and immune profiling revealing reduced CD4⁺PD-1⁺ T cells, increased CD45⁺ cell infiltration and differential PD-1⁺ subsets between responding and non-responding mice in the OVCAR-3 Luc humanized model. (Kang, Sung Wan, et al., 2024)

The OVCAR-3-luc variant is engineered to stably express firefly luciferase, allowing tumor growth and spread to be tracked non-invasively by bioluminescence imaging after D-luciferin administration. Tumor burden, response to treatment, and the onset of metastatic lesions can be quantified longitudinally in the same animals, reducing cohort sizes and improving statistical power. Together, the OVCAR-3 / OVCAR-3-luc orthotopic xenograft system is widely applied in in vivo studies of platinum-based chemotherapy, PARP inhibitors, anti-angiogenic agents, hormone-directed therapies, and novel delivery systems, with circulating CA125 serving as an additional pharmacodynamic biomarker in many study designs.

Cell Line Information: OVCAR-3 / OVCAR-3-luc

OVCAR-3 (NIH:OVCAR-3; ATCC HTB-161) is one of the most extensively characterized human ovarian cancer cell lines, widely regarded as a representative model of high-grade serous ovarian carcinoma. The OVCAR-3-luc derivative carries a stably integrated luciferase reporter, combining the parental line's biological features with in vitro and in vivo optical tracking capability. Key characteristics are summarized below.

Our Services

Alfa Cytology builds and executes complete OVCAR-3 / OVCAR-3-luc orthotopic studies under strict preclinical quality standards: authenticated, mycoplasma-tested cells are expanded under controlled conditions, surgically implanted by experienced technicians, and followed by scheduled bioluminescence imaging, body-weight and CA125 monitoring, randomized treatment administration, and terminal necropsy with histopathology — all delivered with a clear study report tailored to your program's decision points.

Workflow of OVCAR-3 / OVCAR-3-luc Orthotopic Mouse Model Construction

Construction of the orthotopic model follows a standardized, quality-controlled sequence. Each stage below is performed with defined acceptance criteria to ensure reproducible tumor establishment and reliable in vivo data.

  1. Cell banking and quality control: OVCAR-3 or OVCAR-3-luc cells are revived from authenticated master stocks, verified by STR profiling and mycoplasma testing, and expanded in RPMI-1640 with 20% FBS plus insulin until the exponential growth phase is reached.
  2. Luciferase verification (OVCAR-3-luc): Luciferase expression is confirmed in vitro by luminescence assay, ensuring uniform reporter signal before implantation.
  3. Animal preparation: Female immunodeficient mice (e.g., BALB/c nude or NOD/SCID, 6–8 weeks old) are acclimated, randomized, and anesthetized for surgery under approved animal ethics protocols.
  4. Orthotopic implantation: Through a small flank or midline abdominal incision, the ovarian fat pad and ovary are exteriorized; a suspension of 1×106–2×106 OVCAR-3 / OVCAR-3-luc cells (optionally mixed with Matrigel) is injected into the ovarian bursa under a dissecting microscope, followed by abdominal closure with sutures or wound clips.
  5. Post-operative recovery and baseline imaging: Mice are monitored daily during recovery; baseline bioluminescence images are acquired after intraperitoneal D-luciferin administration to confirm successful cell deposition.
  6. Longitudinal tumor monitoring: Tumor growth is tracked weekly by bioluminescence imaging on an in vivo imaging system, complemented by body weight measurement, palpation, and optional serial serum CA125 quantification.
  7. Treatment and interim sampling: Once tumors reach predefined signal thresholds, animals are randomized into treatment groups and dosed according to the study design, with interim blood or imaging readouts collected as scheduled.
  8. Endpoint analysis: At study termination, mice are euthanized; primary ovarian tumors, ascites, and metastatic nodules are excised, weighed, and processed for H&E and immunohistochemical or molecular analyses, followed by a full data report.
