MOPC-315 Syngeneic Model Service for Multiple Myeloma (MM)

Fig 1: MOPC-315 Syngeneic Model for Multiple Myeloma (MM) preclinical research.

The MOPC-315 syngeneic model of multiple myeloma propagates a mineral oil–induced murine plasmacytoma with a defined IgAλ2 idiotype in fully immunocompetent BALB/c hosts—the birthplace of idiotype-specific tumor immunology. Alfa Cytology delivers the complete MOPC-315 workflow—density-controlled cell banking, extramedullary or bone-marrow–homing inoculation, M315-protein monitoring, scheduled dosing and marrow- and survival-inclusive endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of MOPC-315 Syngeneic Model for Multiple Myeloma (MM)

MOPC-315 descends from the mineral-oil era of plasma-cell tumor induction: a plasmacytoma was induced in a BALB/c mouse by oil injection, and the tumor’s cells were brought into continuous culture—creating a line that would become one of immunology’s most consequential reagents. The defining product is its immunoglobulin: a monoclonal IgAλ2 myeloma protein, M315, endowed with anti-hapten activity that recognizes dinitrophenyl- and trinitrophenyl-substituted proteins—a chemically defined specificity that made it a workhorse of hapten-binding research. The cells are pharmacologically distinctive too: they are killed by dexamethasone at concentrations as low as 10 nM, and cyclic AMP was shown to induce their apoptosis and inhibit tumor development in vivo. In culture they grow in suspension with an unusual quirk—populations must be maintained between roughly 1 × 105 and 9 × 105 cells per mL, because densities above 1 × 106/mL trigger rapid death—a density-dependence that rewards disciplined passage scheduling.

Tumor growth curves and incidence showing that VEGF-overexpressing MOPC-315 cells (MOPCSVlm) are rejected by immunocompetent BALB/c mice but grow progressively in BALB/c nude mice, demonstrating active tumor immunity in the syngeneic model. (Kim, Byung-Gyu, et al., 2022)

Immunologically, MOPC-315 is where tumor-specific idiotype immunity was born. In 1972, Lynch and colleagues showed that syngeneic BALB/c mice immunized with the purified M315 myeloma protein developed idiotype-specific immunity—rejecting a challenge with the parental MOPC-315 tumor while remaining fully susceptible to unrelated MOPC plasmacytomas, a specificity landmark that launched the anti-idiotype vaccine field; subsequent work showed light-chain immunization alone was protective, and modern descendants include chemokine-idiotype fusion DNA vaccines and an idiotype-specific TCR-transgenic ecosystem (IdL2-315) for mechanistic studies. For myeloma biology proper, the bone-marrow–homing variant MOPC315.BM (Cellosaurus CVCL_VI68) homes to marrow after intravenous injection, produces paraplegia within thirty days and osteolytic disease readable by noninvasive imaging—in the common BALB/c strain, without specialized colonies—with luciferase and DsRed versions for tracking; the intermediate MOPC315.4 and the slower ATCC parental line complete a latency hierarchy. Efficacy reads through serum M315 quantified by ELISA alongside paraplegia and marrow burden. Caveats: density-dependent death demands careful culture; glucocorticoid sensitivity shapes combination designs; and variant choice (extramedullary versus BM-homing) determines the disease course modeled.

Culture characteristics Suspension growth in EMEM (EBSS) + 2 mM glutamine + 1% non-essential amino acids + 10–20% FBS; maintain at 1–9 × 105 cells/mL — rapid death occurs above 1 × 106/mL; 37 °C, 5% CO2; ACDP Hazard Group 2
Idiotype immunology heritage Lynch et al., PNAS 1972: idiotype-specific immunity rejects parental tumor but not unrelated MOPC tumors — founding demonstration for anti-idiotype vaccines; light-chain immunization sufficient for protection; chemokine-idiotype fusion DNA vaccines (Blood, 2007); IdL2-315 idiotype-specific TCR-transgenic ecosystem
Bone-marrow–homing variant MOPC315.BM homes to and expands in bone marrow after intravenous injection; paraplegia within 30 days; osteolytic disease by noninvasive imaging; MOPC315.BM.Luc and MOPC315.BM.DsRed tracking lines; latency hierarchy MOPC315.BM > MOPC315.4 > ATCC parental

Workflow of MOPC-315 Syngeneic Model Construction

Construction of the MOPC-315 syngeneic model is an inoculation workflow whose quality rests on density-controlled cells, strain-matched recipients and disciplined monitoring. A standard engagement proceeds as follows:

  1. Source and qualify cells: MOPC-315 or variant stocks are cleared by mycoplasma PCR and identity-verified, with IgAλ2/M315 production documented; density history is reviewed (cultures kept at 1–9 × 105 cells/mL), and luciferase/DsRed labels are titrated in vitro where imaging readouts are required.
  2. Prepare recipients: BALB/c mice (typically 6–8 weeks old; IdL2-315 TCR-transgenic hosts where idiotype-specific designs call for them) complete acclimation and health screening.
  3. Prepare the inoculum: log-phase cells within the validated density window are washed, counted and resuspended in PBS (subcutaneous dose for extramedullary formats; 5 × 105 MOPC315.BM cells intravenously for marrow homing; irradiation-assisted doses per design).
  4. Inoculate: cells are implanted subcutaneously for the classic extramedullary plasmacytoma, or injected intravenously for bone-marrow–homing disease; irradiation-assisted disseminated formats follow published conditioning practice.
  5. Assign groups: animals are randomized by body weight at inoculation, or at first M315 detection where established-disease designs require.
  6. Dose and follow: test articles are administered on schedule; body weight, clinical status and hindlimb-paralysis onset are recorded, with serum M315 measured by ELISA on a defined schedule; bioluminescence substitutes for labeled cohorts; survival is the primary endpoint.
  7. Terminate and analyze: bone marrow, vertebrae, spleen and liver are harvested; tumor burden is quantified by λ-chain immunostaining and histology; osteolytic disease is assessed by X-ray or micro-CT; M315 kinetics and survival curves complete the dataset.
