UPPL-1541 Syngeneic Model Service for Bladder Cancer

Fig 1: UPPL-1541 Syngeneic Model for Bladder Cancer preclinical research.

The UPPL-154 syngeneic model service for bladder cancer is an immune-competent murine platform in which luminal-like UPPL-154 urothelial tumor cells — derived from the genetically engineered UPPL (Upk3a-CreERT2; Trp53L/L; PtenL/L; Rosa26LSL-Luc) bladder tumor model — are engrafted into syngeneic C57BL/6 hosts, faithfully reproducing the papillary histology, luminal gene-expression program, and immune-cold microenvironment of human luminal bladder cancer. Alfa Cytology delivers this model as a complete preclinical capability — spanning authenticated UPPL-154 expansion and banking, standardized flank or bladder-site implantation, scheduled tumor-growth monitoring, and comprehensive histopathology with tumor-immune microenvironment profiling by flow cytometry and immunohistochemistry — all configured around each sponsor’s immunotherapy, radiation, or combination-therapy hypothesis.

Overview of UPPL-154 Syngeneic Model for Bladder Cancer

The UPPL platform represents a deliberate correction to a long-standing weakness of bladder cancer modeling. Conventional murine lines such as MB49 grow readily in syngeneic hosts but poorly represent the molecular heterogeneity of human disease. To address this, Saito and colleagues engineered the first subtype-specific mouse models of bladder cancer: the UPPL mouse (Upk3a-CreERT2; Trp53L/L; PtenL/L; Rosa26LSL-Luc), which deletes both copies of Trp53 and Pten specifically in urothelial cells to generate luminal-like tumors with papillary histology — the counterpart being basal-like tumors from BBN carcinogen exposure. Gene-expression analyses show that UPPL-derived material maps closely onto human bladder cancer and is far more faithful than MB49. Because primary UPPL tumors form only after a long latency, conditional reprogramming was used to derive transplantable cell lines from these models — including the UPPL tumor-derived line designated UPPL1541 in the literature (rendered in some commercial catalogues as UPPL-154) — and these lines were confirmed to grow in immunocompetent C57BL/6 mice while retaining luminal-like character with high Pparg and Gata3 gene signatures.

What makes the model especially valuable for immunology research is its deliberately inconvenient — and clinically authentic — immune phenotype. Untreated UPPL tumors are strikingly cold: neutrophils dominate the tumor immune microenvironment (a median of about 65% of the CD45-positive compartment), while CD4⁺ and CD8⁺ T cells are scarce (roughly 4.6% and 1.6% respectively) and PD-L1 expression is high on antigen-presenting populations — including about 96% of dendritic cells — yet only modest on tumor cells themselves. Consistent with this biology, anti-PD-L1 monotherapy fails to slow tumor growth and adds nothing to radiation in combination, recapitulating the checkpoint resistance of luminal-dominant human disease; surgical excision followed by flank rechallenge yields unchecked secondary tumors, demonstrating absent treatment-induced immunological memory. At the same time, radiation alone significantly slows growth and reshapes the immune infiltrate, and the model is receptive to modern combination engineering — photodynamic nanoplatforms plus anti-PD-1 have produced abscopal effects in UPPL tumor-bearing mice. For sponsors, this means a single syngeneic line supports cold-to-hot conversion strategies, radiation-plus-checkpoint sequencing, innate-memory interrogation, and recurrence-versus-cure endpoints in vivo within a fully immunocompetent host.

Cell Line Information: UPPL-154

The table below consolidates the key biological, cultural, and modeling characteristics of the UPPL-154 line, cross-referenced with its designation in the primary literature.

Parameter Details
Cell line designation UPPL-154 (UPPL-154 in catalogue usage; the primary literature designates the line UPPL1541, a transplantable cell line derived from UPPL model tumors by conditional reprogramming; sibling line BBN963 derives from BBN basal-like tumors)
Species / genetic background Mouse (Mus musculus), C57BL/6 background; syngeneic in C57BL/6 immunocompetent hosts
Origin UPPL genetically engineered mouse model (Upk3a-CreERT2; Trp53L/L; PtenL/L; Rosa26LSL-Luc) developed by Saito et al. as the first subtype-specific murine model of bladder cancer; the transplantable cell line was generated from UPPL tumors by conditional reprogramming (Liu-lab CRC technique) and confirmed to grow in C57BL/6 mice (Cells 12:1714, 2023)
Molecular subtype Luminal-like — high Pparg and Gata3 gene-signature expression; papillary histology; gene-expression profiles map closely to human luminal bladder cancer and are more faithful to human disease than MB49
Genetic lesions (parental model) Urothelium-specific deletion of both Trp53 and Pten via tamoxifen-inducible Upk3a-CreERT2; Rosa26LSL-Luc cassette supports luciferase-based tracking
Tumor immune microenvironment Immune cold: neutrophils are the most abundant CD45⁺ population (median ≈64.9%); low CD4⁺ T cells (≈4.6%) and CD8⁺ T cells (≈1.6%); high CD4/CD8 ratio; high PD-L1 on antigen-presenting cells (dendritic cells ≈96.3%, plus neutrophils and monocytes) with modest tumor-cell PD-L1 — anti-PD-L1 monotherapy fails to slow growth and shows no synergy with radiation, modeling luminal-type checkpoint resistance
Growth properties Adherent urothelial tumor cells derived under conditional reprogramming and adapted as transplantable lines; robust engraftment in immunocompetent C57BL/6 mice
Recommended culture conditions Standard complete medium per validated laboratory protocol for CRC-derived urothelial lines; conditional-reprogramming medium is used for expansion of conditionally reprogrammed cultures
Standard implantation practice Published studies implant UPPL cells subcutaneously into the flank and randomize animals once tumors reach approximately 0.1 cm³; organ-site (bladder) engraftment and recurrence-rechallenge designs are available per study objectives
Documented in vivo responses Radiation (two 5 Gy fractions 24 hours apart) significantly slows tumor growth and prolongs survival; anti-PD-L1 (250 μg intraperitoneally, four doses) does not slow growth alone or in combination; excision-and-rechallenge shows no treatment-induced immunological memory; photodynamic nanoplatforms plus anti-PD-1 produce abscopal effects
Host strains C57BL/6 immunocompetent mice (syngeneic); immunodeficient hosts only where specified by the study design
Key research applications Luminal-subtype checkpoint resistance and cold-to-hot conversion strategies; radiation-plus-ICI sequencing and memory-generation studies; recurrence-versus-cure and rechallenge endpoints; nanomedicine, photodynamic therapy, and drug-delivery biodistribution studies in bladder tumor-bearing mice

Our Services

Subtype-faithful immunology demands both the right line and the right analytics, and Alfa Cytology provides both: authenticated UPPL-154 banks, standardized engraftment with consistent luminal histology, scheduled growth monitoring, and endpoint panels combining flow cytometry with CK7/CK20/p63 and PD-L1 immunohistochemistry — all run as one accountable preclinical program.

Workflow of UPPL-154 Syngeneic Model Construction

Construction follows the published flank-engraftment methodology used for subtype-specific syngeneic bladder models: UPPL-154 cells are expanded under authenticated conditions, implanted subcutaneously into C57BL/6 hosts, grown to a standardized baseline volume, randomized, treated, and analyzed with immune-microenvironment endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: UPPL-154 cells are confirmed mycoplasma-negative and identity-verified, then expanded into low-passage working banks under validated urothelial culture conditions.
  2. Cell harvest and formulation: Healthy cultures are harvested, counted and viability-checked, then formulated at the study-specified dose in PBS or a Matrigel-supported suspension per the validated protocol.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane, and UPPL-154 cells are implanted subcutaneously into the flank; organ-site bladder engraftment variants follow the same quality steps.
  4. Establishment monitoring: Animals are palpated on a fixed schedule until tumors become measurable; engraftment rate and latency are recorded as primary quality indicators.
  5. Baseline randomization: Tumors are measured and animals are randomized into treatment arms once tumors reach approximately 0.1 cm³, ensuring balanced starting volumes across groups.
  6. Treatment administration: Experimental arms — checkpoint inhibitors, radiation fractions, chemotherapeutics, photodynamic or nanomedicine platforms, and combinations — are administered per study design with scheduled body-weight and toxicity monitoring.
  7. Endpoint and immune-context analysis: Endpoints integrate tumor-growth curves and survival with tumor, spleen, and blood immune profiling by multi-parametric flow cytometry (neutrophils, monocytes, dendritic cells, CD4⁺/CD8⁺ T cells, PD-1/PD-L1 status) and histopathology (H&E with CK7, CK20, p63, Ki67, and PD-L1 immunohistochemistry); rechallenge designs test treatment-induced immunological memory where specified.
