EC9706 Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: EC9706 Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The EC9706 orthotopic mouse model recreates human esophageal squamous cell carcinoma within its native anatomical compartment—the esophageal wall—producing tumors whose growth pattern and local microenvironment more closely resemble the human disease than flank xenografts ever can. Alfa Cytology delivers this model as a complete, sponsor-ready package: from authenticated EC9706 cell preparation and aseptic orthotopic implantation through scheduled in-life monitoring, test article administration, necropsy, and full histopathological workup, giving your program a rigorous, translationally relevant preclinical dataset without the capital investment of an in-house surgical suite.

Overview of EC9706 Orthotopic Mouse Model for Esophageal Cancer

Where a tumor grows shapes how it behaves. In subcutaneous xenografts, ESCC cells expand in loose subcutaneous tissue with an artificial stromal interface, whereas in an orthotopic setting the same cells must contend with the dense muscularis, peristaltic forces, and lymphatic drainage of the esophagus. These constraints drive a different, more disease-like biology: tumors characteristically expand as nodular mucosal lesions that progressively invade from the mucosa through the muscular layers toward the adventitia, and models built on Chinese ESCC backgrounds such as EC9706 have documented lymphovascular permeation and perineural infiltration—hallmarks of clinical ESCC that flank models rarely display. Because the esophagus lies deep in the thoracoabdominal cavity and cannot be calipered, orthotopic studies rely on scheduled endoscopic assessment, ultrasound or micro-CT sizing where available, body weight and survival trajectories, and terminal histology to chart disease course.

The EC9706 background is one of the most intensively studied Chinese ESCC systems. Established in the late 1990s, it has served as a reference platform for investigations spanning apoptosis regulation, angiogenesis inhibition by retinoids, epithelial–mesenchymal transition, microRNA-mediated metastasis suppression, and drug response, with both in vitro findings and in vivo xenograft validations published across these areas. In the orthotopic configuration, the line supports efficacy testing of cytotoxics, targeted agents, and combination regimens, as well as mechanism-of-action studies in which molecular manipulations (knockdown or overexpression) are examined under physiologically faithful growth conditions. For sponsors, the practical appeal of the EC9706 orthotopic model lies in this combination: a literature-rich molecular context, reproducible tumorigenicity in immunodeficient mice, and readouts that translate more credibly into downstream decision-making.

Cell Line Information: EC9706

EC9706 is among the earliest continuously cultured Chinese ESCC lines and remains a cornerstone model in esophageal cancer research. The line was established in 1997 from a resected esophageal squamous cell carcinoma specimen and entered wide circulation after its initial biological characterization. EC9706 cells grow as an adherent monolayer with a reported population generation time of approximately 26 hours, a plate colony-forming efficiency of 91.9%, and the capacity to form clones in soft agar—together indicating strong anchorage-independent growth potential. Comparative genomic hybridization in the original characterization revealed copy number gains at 1p, 1q, 2p, 2q, 5p, 7p, 7q, 11q, 15q and 20q, with losses at 3p, 4, 9p, 14, 18 and Xq. As with all long-cultured lines, STR authentication is regarded as essential before experimental use.

Our Services

An orthotopic esophageal program is unforgiving: a single inconsistent cell lot or a poorly executed implantation can quietly compromise an entire study. Alfa Cytology de-risks the process for sponsors by controlling every variable—STR-authenticated, mycoplasma-negative EC9706 banks; aseptic microsurgical implantation with defined inoculum volumes; standardized in-life observation including endoscopic checks, body weight tracking, and humane endpoints; and terminal necropsy with H&E and immunohistochemistry—so the data you receive reflects your compound, not our technique.

Workflow of EC9706 Orthotopic Mouse Model Construction

Construction follows a defined sequence from cell bank release to terminal analysis, executed under aseptic surgical conditions. Inoculum density, route refinements, and monitoring modalities are finalized with each sponsor according to the experimental objective, with bioluminescent tracking available when a luciferase-labeled EC9706 variant is requested.

  1. Cell bank release: EC9706 vials are revived from the working cell bank, tested negative for mycoplasma, and confirmed by STR profiling against the reference profile before any animal work begins.
  2. Inoculum preparation: Log-phase cells are harvested, counted, and adjusted to the study-specified concentration—typically on the order of 1 × 107 cells in a 25–50 µl carrier volume.
  3. Animal preparation: BALB/c nude mice undergo quarantine and acclimatization, then are randomized into treatment and control cohorts with body-weight stratification.
  4. Anesthesia and exposure: Under general anesthesia with perioperative analgesia, a midline upper abdominal incision exposes the stomach and the distal esophagus.
  5. Orthotopic implantation: Using a fine-gauge needle, the cell suspension is injected slowly into the esophageal wall at the mucosa–submucosa level, and a raised wheal at the injection site verifies correct intramural delivery; the laparotomy is then closed in layers.
  6. Postoperative observation: Animals recover under warming with daily monitoring of weight, feeding, and incision integrity until surgical recovery is complete.
  7. In-life disease monitoring: Tumor development is followed by scheduled endoscopic inspection and/or imaging (bioluminescence for luciferase-labeled variants, or ultrasound/micro-CT where available), alongside body weight and condition scoring.
  8. Terminal analysis: At protocol endpoints, mice are euthanized for gross necropsy; the esophagus, regional lymph nodes, and organs are harvested for tumor measurement, ex vivo imaging, and histopathological processing.

Case Study-EC9706 Orthotopic Mouse Model Development

EC9706 orthotopic studies form an active part of Alfa Cytology’s esophageal cancer model portfolio. In one such preclinical program, our team established reproducible orthotopic tumors from STR-verified EC9706 cells, confirmed intramural tumor location and esophageal wall architecture at necropsy, and supported randomized efficacy arms with scheduled endoscopic monitoring and terminal histopathology. Sponsors have used the resulting datasets to prioritize candidates for downstream development. Further details and representative readouts from EC9706 orthotopic studies are available upon request under confidentiality.
