SW480 Orthotopic Mouse Model Service for Colon Cancer

Fig 1: SW480 Orthotopic Mouse Model for Colon Cancer preclinical research.

The SW480 orthotopic mouse model harnesses a human colorectal adenocarcinoma line isolated from a Dukes' B primary tumor, offering a preclinical window into early-stage disease progression, KRAS-driven signaling, and Wnt/β-catenin pathway dysregulation within an anatomically authentic colon microenvironment. Alfa Cytology engineers this model with authenticated SW480 cell assets, refined cecal wall surgical implantation, and longitudinal tumor monitoring, furnishing investigators with a reliable platform for dissecting oncogenic driver biology and benchmarking therapeutic interventions against KRAS-mutant, MSS colorectal carcinoma.

Overview of SW480 Orthotopic Mouse Model for Colon Cancer

The SW480 cell line was established in 1976 by A. Leibovitz and colleagues at the Scott and White Clinic from a primary colorectal adenocarcinoma classified as Dukes' stage B, resected from a 50-year-old Caucasian male patient. This line carries a homozygous KRAS p.G12V activating mutation that drives constitutive MAPK signaling, alongside a truncating APC mutation (p.Q1338Ter) that abolishes β-catenin degradation and sustains canonical Wnt pathway activation. Additional alterations include heterozygous TP53 missense mutations (p.R273H and p.P309S) and an ATM p.R2461P variant. Genomically, SW480 is microsatellite stable (MSS) and CIMP-negative, with a hypertriploid karyotype. Morphologically, the cells display an epithelial cobblestone growth pattern reminiscent of healthy colonic epithelium, express carcinoembryonic antigen (CEA) and cytokeratin, and exhibit a doubling time of approximately 20–26 hours under standard culture conditions.

When established orthotopically within the murine cecal wall, SW480 tumors faithfully recapitulate the glandular differentiation, stromal architecture, and invasive front dynamics of human Dukes' B colorectal adenocarcinoma. This anatomical placement preserves the luminal microbiome interface, submucosal vascular networks, and mesenteric lymphatic drainage, thereby supporting locoregional progression and occasional distant dissemination. The concurrent KRAS G12V and APC truncation render SW480 intrinsically sensitive to 5-fluorouracil and responsive to dual KRAS/MAPK pathway inhibition strategies, while the MSS background and intact mismatch repair machinery provide a distinct immunological context compared to MSI-H counterparts. These attributes position the SW480 orthotopic platform as an essential substrate for preclinical evaluation of KRAS-directed therapeutics, Wnt pathway modulators, and chemotherapeutic regimens in early-stage colorectal cancer.

Workflow of SW480 Orthotopic Mouse Model Construction

Establishment of the SW480 orthotopic colon cancer model requires meticulous surgical technique, rigorous cell line quality control, and standardized perioperative care to achieve reproducible engraftment and meaningful therapeutic readouts. The protocol outlined below reflects a validated operational framework executed within specific pathogen-free barrier facilities, advancing through the following sequential stages:

  1. Cell Line Authentication and Expansion: SW480 cells are revived from authenticated cryopreserved master stocks and expanded in Leibovitz's L-15 medium supplemented with 10% FBS under standard growth conditions. Prior to surgical use, cells undergo mandatory STR identity confirmation, mycoplasma PCR screening, and viability quantification by trypan blue exclusion to ensure a live-cell fraction exceeding 95%. For studies requiring longitudinal imaging, luciferase-expressing SW480 derivatives are validated for signal linearity in vitro prior to in vivo deployment.
  2. Host Selection and Environmental Acclimation: Female or male athymic nude (nu/nu) or NOD/SCID gamma (NSG) mice aged 6–8 weeks are selected based on study design requirements. Animals are housed in specific pathogen-free vivaria under controlled temperature (22 ± 2°C), humidity (50 ± 10%), and a 12-hour light/dark cycle, with ad libitum access to irradiated chow and acidified water. A minimum five-day acclimation period is observed, during which baseline body weights and clinical observations are documented.
  3. Anesthesia and Aseptic Surgical Preparation: On the day of implantation, mice are anesthetized with 2–3% isoflurane inhalation delivered via nose cone, with anesthetic depth confirmed by absence of pedal withdrawal reflex. The ventral abdominal wall is shaved and sterilized with sequential povidone-iodine and 70% ethanol applications. Sterile surgical drapes are positioned to establish an aseptic operative field, and ophthalmic ointment is applied to prevent corneal desiccation.
  4. Cecal Exposure and Orthotopic Cell Inoculation: A 1.0–1.5 cm midline laparotomy is performed to access the peritoneal cavity. The cecum is gently exteriorized using atraumatic forceps and kept hydrated with warm sterile PBS. A single-cell suspension of 1 × 106 to 2 × 106 viable SW480 cells, resuspended in 25–50 µL of serum-free L-15 medium mixed 1:1 with growth factor-reduced Matrigel, is loaded into a 1 mL syringe fitted with a 30 G needle. The needle is inserted into the subserosal layer of the cecal wall at the anti-mesenteric border, and the cell suspension is slowly delivered to form a discrete submucosal bleb without mucosal perforation.
  5. Hemostasis, Cecal Repositioning, and Wound Closure: The injection site is compressed gently with a sterile cotton swab for approximately 3 minutes to ensure hemostasis and prevent cell leakage into the peritoneal cavity. The cecal wall is inspected for integrity, then gently returned to anatomical position within the abdomen. The abdominal musculature is closed with absorbable 5-0 sutures in a continuous pattern, and the skin is apposed with surgical staples or tissue adhesive. Mice are transferred to a warmed recovery chamber and monitored until full ambulatory recovery.
  6. Postoperative Care and Tumor Surveillance: During the immediate postoperative period, mice receive analgesia according to institutional guidelines and are monitored daily for wound integrity, body weight, and clinical score. Tumor establishment is tracked twice weekly beginning approximately 7–10 days post-implantation. For luciferase-expressing SW480 lines, tumor burden is quantified in vivo via IVIS bioluminescence imaging following intraperitoneal D-luciferin administration. Conventional cohorts are monitored by abdominal palpation and digital caliper measurement of externally palpable masses.
  7. Stratified Randomization and Therapeutic Intervention: Once tumors reach a predetermined volume or bioluminescence signal intensity—typically 100–200 mm³ or 106 to 107 photons/sec—mice are stratified by tumor burden and body weight into treatment and vehicle control cohorts. Investigational agents are administered according to predefined dosing regimens, with routes selected based on pharmacokinetic properties and mechanism of action. Concurrent tumor monitoring continues to capture real-time therapeutic response kinetics.
  8. Necropsy, Triage, and Multi-Platform Endpoint Analysis: At study termination, mice are humanely euthanized under deep anesthesia. The cecum and primary tumor are excised en bloc, photographed, and weighed. Tumor tissue is triaged for formalin fixation and paraffin embedding (H&E, IHC for Ki-67, CD31, phospho-ERK, β-catenin, axin2, cleaved caspase-3), snap-freezing for Western blot or RNA-seq, and dissociation for flow cytometric analysis. Metastatic foci in mesenteric lymph nodes, liver, lung, and peritoneum are enumerated macroscopically and confirmed histologically.

Case Study-SW480 Orthotopic Mouse Model Development

In a recent preclinical investigation, the SW480 orthotopic model was leveraged to assess the anti-tumor efficacy of a novel Wnt/β-catenin pathway inhibitor administered as monotherapy and in combination with 5-fluorouracil in KRAS-mutant, MSS colorectal cancer. Following cecal wall implantation, tumor-bearing mice were stratified by tumor volume and randomized into combination, Wnt inhibitor monotherapy, 5-FU monotherapy, and vehicle control arms. Longitudinal monitoring revealed progressive tumor growth suppression in the combination cohort beginning at day 12 post-treatment initiation, whereas single-agent arms showed only partial response. Terminal histopathological analysis demonstrated markedly reduced nuclear β-catenin accumulation and axin2 expression in combination-treated tumors, alongside attenuated phospho-ERK immunoreactivity and increased apoptotic indices. These observations highlight the capacity of the SW480 orthotopic platform to dissect pathway co-dependency and to generate mechanistic insights into combination strategies targeting the Wnt/KRAS axis in early-stage colorectal adenocarcinoma.
