CE81T/VGH-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: CE81T/VGH-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The CE81T/VGH-luc orthotopic mouse model is a luciferase-tagged esophageal squamous cell carcinoma xenograft system that recapitulates tumor growth in the native esophageal microenvironment and allows real-time, noninvasive tracking of tumor progression. Alfa Cytology delivers this model as a fully managed preclinical service, taking your project from cell line authentication and surgical implantation through longitudinal bioluminescent imaging, efficacy testing, and histopathological endpoints, so your team can focus on interpreting data rather than mastering microsurgery.

Overview of CE81T/VGH-luc Orthotopic Mouse Model for Esophageal Cancer

Orthotopic xenograft models are generated by implanting human tumor cells directly into the organ from which the tumor originated, rather than into a subcutaneous site. For esophageal cancer research, this distinction matters: tumors growing within the esophageal wall are exposed to the same mechanical, stromal, and vascular cues as patient tumors, and they more faithfully reproduce local invasion patterns and metastatic spread. The CE81T/VGH cell line is a well-characterized human esophageal squamous cell carcinoma (ESCC) line originally derived from a 57-year-old male patient of Chinese/Taiwanese origin. It retains an epithelial phenotype, has a reported doubling time of approximately 72 hours, and forms tumors reliably in immunodeficient mice. When CE81T/VGH cells are engineered to stably express firefly luciferase, the resulting CE81T/VGH-luc line emits a photon signal proportional to viable tumor cell number, which converts an anatomically deep, surgically inaccessible tumor into a quantifiable optical readout.

Because the mouse esophagus is a narrow, mobile structure, conventional caliper measurement is impossible, and bioluminescence imaging (BLI) becomes the enabling technology for this model. Luciferase-expressing CE81T/VGH xenografts have been used in published preclinical studies with optical imaging to evaluate anticancer agents, and the parental line has served as a reference model in investigations of apoptosis, DNA repair, and EGFR-related signaling in ESCC. Researchers use the CE81T/VGH-luc orthotopic model for compound efficacy screening, radiosensitizer evaluation, biomarker validation, and mechanistic studies in which tumor growth, regression, and relapse must be followed longitudinally in the same animal. Compared with subcutaneous xenografts, the orthotopic setting yields biologically more relevant dose-response relationships, which is why it is increasingly favored for late-stage preclinical candidate selection.

Cell Line Information: CE81T/VGH-luc

CE81T/VGH-luc is a luciferase reporter derivative of the CE81T/VGH human ESCC cell line. The parental line was established from an esophageal squamous cell carcinoma of a 57-year-old male patient, and stable transfection of the firefly luciferase gene enables optical detection of tumor cells in vivo. The line is maintained under standard adherent culture conditions and is authenticated by short tandem repeat (STR) profiling. Key characteristics are summarized below.

Our Services

Building an orthotopic esophageal xenograft demands specialized surgical skill, a validated luciferase-expressing cell line, and an imaging infrastructure that many laboratories cannot maintain in-house. Alfa Cytology removes that burden: our team handles every stage of the CE81T/VGH-luc orthotopic model, from cell line quality control, mycoplasma testing, and STR verification to microsurgical implantation, scheduled bioluminescent imaging, randomized treatment arms, and full necropsy with histology, delivering study-ready data packages tailored to your protocol.

Workflow of CE81T/VGH-luc Orthotopic Mouse Model Construction

Model construction combines cell line qualification, orthotopic microsurgery, and longitudinal optical monitoring. The procedure below reflects the standard workflow used to establish luciferase-labeled ESCC tumors in the esophageal wall of immunodeficient mice; details such as cell dose and imaging schedule are optimized for each study.

  1. Cell line qualification: CE81T/VGH-luc cells are revived, confirmed negative for mycoplasma, verified by STR profiling, and tested for stable luciferase expression before expansion.
  2. Cell preparation: Exponentially growing cells are harvested, counted, and resuspended at the working concentration in a suitable carrier such as Matrigel to retain cells at the injection site.
  3. Animal preparation: Immunodeficient mice (e.g., BALB/c nude) are acclimatized, randomized into study groups, and anesthetized with adequate analgesia for surgery.
  4. Surgical exposure: A small upper abdominal incision is made, the stomach is gently exteriorized, and the distal esophageal wall is lifted and stabilized with micro-instruments.
  5. Orthotopic implantation: Using a fine-gauge needle, the cell suspension is injected slowly into the esophageal wall, producing a visible bleb that confirms intramural delivery; the incision is then sutured closed.
  6. Postoperative recovery: Mice receive supportive care and are monitored daily for body weight, feeding behavior, and surgical recovery during the establishment phase.
  7. Bioluminescent monitoring: Tumor engraftment and growth are tracked noninvasively at scheduled time points after D-luciferin administration, with signal intensity quantified for each animal.
  8. Endpoint analysis: At study termination, tumors and organs are excised for ex vivo imaging, tumor weight and volume measurement, and histopathological evaluation.

Case Study-CE81T/VGH-luc Orthotopic Mouse Model Development

In a representative preclinical engagement, Alfa Cytology partnered with a client to establish the CE81T/VGH-luc orthotopic model as the efficacy platform for an investigational antitumor agent. Our scientists qualified the reporter cell line, performed orthotopic implantation in immunodeficient mice, and confirmed stable engraftment through serial bioluminescent imaging. Once baseline tumor burden was established, animals were randomized and dosed according to the client’s protocol, with longitudinal imaging revealing treatment-dependent differences in tumor signal, and terminal histopathology confirming tumor location and tissue architecture. The study provided the client with quantitative, image-backed efficacy evidence in a clinically relevant preclinical setting. Additional representative data from our CE81T/VGH-luc orthotopic program can be shared upon request.
