PC3 / PC3-luc Orthotopic Mouse Model Service for Prostate Cancer

Fig 1: PC3 / PC3-luc Orthotopic Mouse Model for Prostate Cancer preclinical research.

The PC3 / PC3-luc orthotopic mouse model for prostate cancer is an androgen-independent, AR-negative human xenograft platform in which highly metastatic PC3 cells are grafted into the prostate of immunodeficient mice, enabling organ-confined tumor growth together with spontaneous spread to regional lymph nodes, lung, and bone. Alfa Cytology supplies this model as an end-to-end preclinical capability — including authenticated PC3 / PC3-luc expansion and luciferase-stable banking, surgical orthotopic implantation or surgical orthotopic implantation of intact tumor fragments, scheduled in vivo bioluminescence surveillance of primary and metastatic lesions, and complete necropsy-based histopathology — all engineered around each sponsor’s compound, schedule, and endpoint blueprint.

Overview of PC3 / PC3-luc Orthotopic Mouse Model for Prostate Cancer

PC3 stands apart from other prostate cancer lines as a model of the fully androgen-independent, AR-null state of advanced disease. The line was established in 1979 by Kaighn and colleagues from a lumbar vertebral bone metastasis of a grade IV prostatic adenocarcinoma resected from a 62-year-old donor, and it has since become the reference line for late-stage, hormone-refractory biology. PC3 carries no detectable androgen receptor or PSA expression, is unresponsive to androgens, glucocorticoids, and EGF/FGF stimulation in vitro, harbors PTEN loss, and is near-triploid with a modal count of 62 chromosomes and no Y chromosome. It is also one of the most aggressively growing prostate lines in culture and in animals, displays high EGFR and TGF-α expression, expresses neuroendocrine markers such as CD44, NSE, and chromogranin A, and shows the highest metastatic potential among the classic prostate cancer panel — exceeding DU145 and far surpassing LNCaP.

(Xiang, Yuzhu, et al., 2013)

Orthotopic grafting of PC3 unlocks this metastatic program: unlike flank xenografts, which rarely disseminate, tumors implanted into the dorsal or dorsolateral prostate lobes invade locally and seed regional lymph nodes and lung at high frequency, and surgical orthotopic implantation (SOI) of intact tumor fragments — which preserves stromal architecture — reproducibly yields the widespread pulmonary and lymphatic metastasis that cell-suspension models often require extensive selection to achieve. The PC3-luc derivative, stably transduced with firefly luciferase, makes every stage of this cascade measurable: tumor take, primary growth, and the emergence of distant lesions can be followed non-invasively by in vivo bioluminescence after luciferin administration, with ex vivo organ imaging at necropsy confirming metastatic distribution. This combination of rapid growth, robust spontaneous metastasis, and quantitative imaging makes the PC3 / PC3-luc orthotopic model a workhorse for anti-metastatic agent evaluation, bone-targeting therapies, neuroendocrine prostate cancer studies, and mechanistic work on invasion and dissemination.

Cell Line Information: PC3 / PC3-luc

The table below consolidates the defining biological and modeling features of the PC3 parental line and its luciferase-labeled derivative PC3-luc. Both are human-derived and require immunodeficient hosts for in vivo work.

Parameter Details
Cell line designation PC3 (parental line); PC3-luc (luciferase-labeled derivative)
Species / donor Human (Homo sapiens), male; isolated from a 62-year-old Caucasian donor with metastatic prostate adenocarcinoma
Origin Established in 1979 by M.E. Kaighn and colleagues from a lumbar vertebral bone metastasis of a grade IV prostatic adenocarcinoma (Kaighn et al., 1979, Investigative Urology 17:16–23)
Depository / catalogue ATCC CRL-1435; Cellosaurus CVCL_0035
Tumor type Poorly differentiated prostatic adenocarcinoma (epithelial origin)
Cytogenetics Near-triploid and markedly aneuploid; modal chromosome number 62, with no Y chromosome detected
Androgen status Androgen independent and AR null: no androgen receptor mRNA or protein, no PSA and no PSMA expression; growth unaffected by androgens or glucocorticoids both in vitro and in vivo
Molecular features PTEN deficient; high EGFR and TGF-α expression; expresses neuroendocrine markers CD44, neuron-specific enolase (NSE), and chromogranin A; transferrin receptor expression supports growth stimulation by bone marrow-derived transferrin
Growth properties Adherent epithelial cells with anchorage-independent growth in soft agar; among the fastest proliferating prostate cancer lines, with population doubling times in the range of roughly 20–35 hours
Recommended culture conditions F-12K medium (Kaighn’s modification) supplemented with 10% fetal bovine serum at 37 °C in a humidified atmosphere (ATCC guidelines); many laboratories alternatively maintain PC3 in RPMI-1640 with 10% FBS
Reporter labeling (PC3-luc) Stable firefly luciferase expression introduced by lentiviral or plasmid transduction; compatible with in vitro reporter assays and whole-animal in vivo bioluminescence imaging
Standard orthotopic inoculum Published protocols range from 5 × 104 to 2 × 106 cells in 10–30 μL of serum-free medium mixed 1:1 with Matrigel or type I collagen, injected into the dorsal or dorsolateral prostate lobe; subcutaneous reference dosing is 1 × 106 cells per site
Surgical orthotopic implantation (SOI) Implantation of histologically intact tumor fragments into the prostate ventrolateral lobes reproducibly produces high-frequency lung and lymph node metastasis without prior in vivo selection
Host strains Athymic nude (BALB/c-nu/nu), SCID, and NSG mice
Metastatic behavior Orthotopic tumors metastasize to locoregional (paraaortic, iliac) lymph nodes and lung at high frequency; skeletal metastases are reproducible via intracardiac injection (1 × 105 cells), and osteolytic bone lesions via intra-tibial inoculation
Key research applications Anti-metastatic and anti-invasion agent evaluation; bone metastasis and osteolytic lesion studies; neuroendocrine prostate cancer (NEPC) biology; angiogenesis research; combination chemotherapy and targeted-therapy efficacy testing in a hormone-refractory setting

Workflow of PC3 / PC3-luc Orthotopic Mouse Model Construction

Construction follows established orthotopic xenograft methodology: luciferase-labeled PC3 cells are expanded under authenticated conditions, formulated in a Matrigel- or collagen-supported suspension (or prepared as intact tumor fragments for SOI), grafted into the prostate of immunodeficient mice, and followed by serial imaging and metastasis-oriented endpoint analysis. The workflow comprises the following stages.

  1. Cell authentication and banking: PC3 or PC3-luc cells are verified by STR profiling, confirmed mycoplasma-free, and expanded into low-passage working banks under standardized F-12K culture conditions.
  2. Cell harvest and formulation: Subconfluent cultures are trypsinized, washed, counted and viability-checked, then resuspended in serum-free medium mixed 1:1 with cold Matrigel or neutralized type I collagen at the study-specified dose (commonly 1 × 105–2 × 106 cells per 10–30 μL) and kept on ice until implantation.
  3. Anesthesia and surgical exposure: Male immunodeficient mice are anesthetized with isoflurane (or ketamine/xylazine where protocol-specified); a lower abdominal incision externalizes the bladder and seminal vesicle, exposing the dorsal or dorsolateral prostate lobe for inoculation.
  4. Orthotopic implantation: The cell suspension is slowly injected into the prostate lobe using a fine-gauge needle (or small intact tumor fragments are grafted for the SOI variant); the needle is held briefly after injection so the matrix partially sets, limiting reflux and equalizing take rates.
  5. Closure and perioperative care: The prostate is repositioned, the abdominal wall and skin are sutured, and animals receive analgesia with monitored recovery according to institutional guidelines.
  6. Engraftment confirmation and serial imaging: Luciferin is administered intraperitoneally and bioluminescence is acquired to confirm tumor take and normalize group allocation; imaging repeats on a weekly cadence to quantify primary growth and flag emerging metastatic signal.
  7. Intervention and endpoint analysis: Experimental arms — chemotherapy, targeted agents, anti-metastatic or bone-targeting compounds, and combination regimens — proceed per study design; endpoints integrate caliper and imaging measurements with necropsy, ex vivo organ bioluminescence to map metastatic distribution, and histopathology (H&E with Ki67 and marker-specific immunohistochemistry) of primary tumor, lymph nodes, lung, liver, and bone.
