TE-7 Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: TE-7 Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The TE-7 orthotopic mouse model grows human esophageal cancer cells within the esophageal wall itself, capturing the tissue architecture, stromal interactions, and invasive behavior that subcutaneous xenografts cannot reproduce, and making it a demanding yet highly informative platform for preclinical esophageal cancer studies. Bringing together authenticated cell resources, skilled microsurgery, and structured study management, Alfa Cytology is well positioned to deliver TE-7 orthotopic models with dependable quality from inoculation through final data package.

Overview of TE-7 Orthotopic Mouse Model for Esophageal Cancer

TE-7 occupies a unique position in the history of esophageal cancer research. Established in Japan by the Nishihira laboratory as part of the pioneering TE series in the late 1970s and early 1980s, the line was for many years one of the very few in vitro models available for esophageal adenocarcinoma and was adopted by laboratories worldwide for studies of p53 inactivation, gene expression profiling, galanin signaling, and polarity gene disruption. Its career took a remarkable turn in 2007, when a multi-method investigation combining xenograft histology, short tandem repeat profiling, mutation analysis, and array-based comparative genomic hybridization demonstrated that TE-7 is in fact an esophageal squamous cell carcinoma line, genetically identical to TE-2, TE-3, TE-12, and TE-13, with its xenografts displaying unequivocal squamous histology. This episode became a textbook case for cell line authentication and reshaped how the field uses the TE panel in vitro.

For animal studies, TE-7 cells form xenografts readily in immunodeficient mice and, like other members of the TE series, are well suited to orthotopic implantation into the esophageal wall. In this configuration, tumors develop in direct contact with esophageal smooth muscle, fibroblasts, and vasculature, allowing investigators to examine wall-layer invasion, tumor–stroma signaling, and the impact of a native biomechanical environment on treatment response in vivo — endpoints that carry far greater translational weight than flank tumor volume alone. Reporter labeling with luciferase or fluorescent proteins further enables non-invasive longitudinal monitoring of tumor burden at this deep anatomical site. With its extensive publication record and now clearly defined squamous identity, TE-7 remains a practical, well-understood option for orthotopic studies of esophageal tumor biology, candidate drug evaluation, and combination regimens, provided the line is handled with the authentication discipline its history demands.

Cell Line Information: TE-7

TE-7 is one of the most cited esophageal cancer cell lines in the literature, and its well-documented identity history makes accurate characterization especially important. The essential facts are compiled below.

Item Description
Cell line name TE-7
Synonyms TE7; TE 7
Species Human (Homo sapiens)
Reported donor Japanese male patient (historically described as esophageal adenocarcinoma)
Disease (per current evidence) Esophageal squamous cell carcinoma; demonstrated to be genetically identical to TE-2, TE-3, TE-12, and TE-13
Origin TE series, Nishihira laboratory, Japan (established late 1970s–early 1980s)
Cellosaurus accession CVCL_9972 (RRID: CVCL_9972)
Authenticity status Flagged as a problematic/misidentified line in international registries; authenticated use requires STR confirmation of its squamous identity
Key identity study Boonstra et al., Cancer Research 67:7996–8001 (2007) — xenograft histology, STR profiling, mutation analysis, and array-CGH proved squamous origin
Morphology Adherent epithelial cells growing as monolayers
Culture conditions Commonly maintained in DMEM or RPMI 1640 supplemented with 10% FBS; 37 °C, 5% CO2
Molecular studies Investigated for p53 inactivation, gene expression signatures, galanin signaling cascade, and disruption of polarity complex genes
Tumorigenicity Readily forms xenografts in immunodeficient nude mice; xenograft histology shows squamous cell carcinoma features
Orthotopic suitability Amenable to intramural implantation into the esophageal wall using standard TE-series orthotopic techniques
Research applications Esophageal tumor biology, signaling pathway studies, anti-cancer drug screening, combination therapy evaluation, and method-development research

Our Services

Aware of the identity history that surrounds TE-7, Alfa Cytology applies an authentication-first workflow — STR profiling before expansion, monitored cell banking, and documented chain of custody — so that every TE-7 orthotopic study begins with cells of verified identity and ends with data you can trust.

Workflow of TE-7 Orthotopic Mouse Model Construction

TE-7 orthotopic tumors are generated by delivering authenticated cells into the wall of the abdominal esophagus through a mini-laparotomy, following the intramural implantation strategy validated for TE-series esophageal cancer lines. The process typically comprises the following stages.

  1. Identity confirmation and cell banking: TE-7 cells are STR-profiled against the documented genotype of the TE series and tested for mycoplasma; verified early-passage stocks are expanded and cryobanked for study use.
  2. Study-batch preparation: Cells are recovered from the authenticated bank, cultured under controlled conditions, harvested in logarithmic phase, counted, and checked for viability before inoculation.
  3. Inoculum formulation: The cell suspension is adjusted — commonly around 1 × 106 cells per mouse — and mixed with an equal or defined volume of ice-cold Matrigel (roughly 20–30 μL total) to anchor the cells at the implantation site.
  4. Laparotomy and esophageal exposure: Anesthetized immunodeficient mice receive a short upper-abdominal incision; gentle elevation of the liver lobe and caudal traction on the stomach bring the distal esophagus into view and place the wall under slight tension.
  5. Intramural implantation: A fine-gauge needle is advanced into the esophageal wall and the cell–Matrigel mixture is slowly deposited within the wall layers; a small pale swelling at the site indicates successful intramural placement without luminal perforation.
  6. Layered closure and recovery: The peritoneum and abdominal wall are closed with continuous 6-0 absorbable suture and the skin with clips or suture; mice receive thermal support and fluids and are monitored daily for weight, feeding, and activity.
  7. In-life monitoring and endpoint work-up: Tumor development is followed by scheduled palpation or imaging (luciferase/fluorescence reporters where applied) together with body-weight records in vivo; at termination, tumors are excised and weighed, and the esophagus plus regional lymph nodes and peritoneum are processed for H&E histology and immunostaining.

Case Study-TE-7 Orthotopic Mouse Model Development

Alfa Cytology has built TE-7 orthotopic modeling experience across preclient preclinical projects involving model establishment, growth monitoring, and intervention testing under authenticated cell handling standards. These efforts have yielded stable implantation outcomes and interpretable histopathology readouts, and project-specific information can be shared as collaborations take shape.

Why Choose Alfa Cytology?

TE-7's well-known identity history makes provider selection unusually consequential — and Alfa Cytology has turned that challenge into a service strength.

  • Authentication-first handling: every TE-7 batch is STR-profiled against the documented TE-series genotype before it enters an animal study.
  • Surgical consistency: standardized intramural implantation technique developed from TE-series orthotopic methodology secures reliable tumor placement.
  • Native-context biology: orthotopic tumors in the esophageal wall deliver stromal interaction and invasion data that flank models cannot provide.
  • Monitoring flexibility: palpation-based schedules or reporter-based imaging can be matched to the budget and readout needs of each program.
  • Transparent reporting: identity documentation, in-life records, and full histopathology accompany every data package.
  • Dedicated preclinical focus: as a research-only CRO, every process is optimized for discovery science rather than stretched across unrelated services.
