MC38-OVA Syngeneic Model Service for Colon Cancer

Fig 1: MC38-OVA Syngeneic Model for Colon Cancer preclinical research.

The MC38-OVA syngeneic model of colon cancer equips the immunogenic MC38 adenocarcinoma with a defined model antigen—chicken ovalbumin—enabling antigen-specific T-cell responses to be tracked with precision in fully immunocompetent mice. Alfa Cytology delivers the complete MC38-OVA workflow—construct-verified cell banking, flank implantation, OT-I/OT-II adoptive-transfer designs, checkpoint and vaccine intervention schedules, and tetramer-based endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of MC38-OVA Syngeneic Model for Colon Cancer

MC38-OVA is the antigen-defined sibling of the classic MC38 line: the mismatch-repair-deficient, hypermutated C57BL/6 colon adenocarcinoma is stably engineered to express full-length chicken ovalbumin, whose two dominant epitopes slot perfectly into the best-characterized T-cell toolkit in immunology—SIINFEKL (OVA257–264) presented on H-2Kb to OT-I CD8 T cells, and ISQAVHAAHAEINEAGR (OVA323–339) on I-Ab to OT-II CD4 T cells. Because OT-I (JAX 003831) and OT-II (JAX 004194) transgenic strains are available on congenic CD45.1 or Thy1.1 backgrounds, cohorts of naive, antigen-specific T cells can be adoptively transferred and followed as they proliferate, differentiate, infiltrate and exhaust inside a real tumor—something no human tumor model permits. The detection toolbox is equally mature: H-2Kb–SIINFEKL tetramers quantify antigen-specific T cells, the 25-D1.16 antibody directly visualizes peptide–MHC complexes on tumor tissue, and the B3Z hybridoma reports presentation in vitro. Constructs vary by laboratory—some versions co-express EGFP at equal copy number, others carry neomycin-selected expression cassettes—so construct verification is a standing prerequisite.

In vivo the model runs on a familiar template: one million cells in the flank, tumors palpable within five to seven days, and—in the classic adoptive-cell-therapy format—two to five million OT-I cells delivered intravenously around day five to seven, after which growth, rejection or equilibrium plays out under the sponsor’s intervention. The system’s publication record is broad and deep: checkpoint blockade has been shown to invigorate OT-I cells within MC38-OVA tumors; regulatory studies of antigen presentation have used it to expose how lncRNA EPIC1, the deubiquitinase USP22 or the oncoprotein SND1 dampen MHC-I display; dendritic-cell and DC-exosome vaccines have been benchmarked against it; DNA vaccines encoding tandem neoantigen strings have used OVA as the reporter epitope; and recent work showed tumor-associated macrophages acquiring and presenting its antigen—cross-dressing—to T cells. Three caveats discipline its use: ovalbumin is a foreign model antigen, so translation to human tumor antigens requires care; expression can drift or be lost under immune pressure, demanding maintenance of selection and pre-implant verification; and its defined antigen makes it more immunogenic than parental MC38, so rejection kinetics run faster and windows are correspondingly shorter.

Cell Line Information: MC38-OVA

Key characteristics of MC38-OVA and its companion reagents are summarized below.

Item Details
Cell line name MC38-OVA; MC38 stably engineered to express full-length chicken ovalbumin (construct varies by laboratory — e.g., EGFP co-expression or neomycin-selected cassettes); a Genentech repository version expresses EGFP plus OVA at equal copy number
Parental line MC38 (murine colon adenocarcinoma, DMH-induced in C57BL/6, 1975; mismatch-repair deficient, MSI-high)
Species / host options Mus musculus; immunocompetent C57BL/6; OT-I / OT-II transgenic C57BL/6; congenic CD45.1 or Thy1.1 hosts for donor tracking; Rag2−/− C57BL/6-background hosts for pure adoptive-cell-therapy designs
Antigen / epitopes Full-length OVA; CD8 epitope SIINFEKL (OVA257–264) on H-2Kb; CD4 epitope ISQAVHAAHAEINEAGR (OVA323–339) on I-Ab
Companion transgenic strains OT-I (C57BL/6-Tg(TcraTcrb)1100Mjb/J, JAX 003831) — OVA-specific CD8 T cells; OT-II (JAX 004194) — OVA-specific CD4 T cells
Detection toolkit H-2Kb–SIINFEKL tetramers; anti-SIINFEKL/H-2Kb antibody clone 25-D1.16; B3Z hybridoma reporter; OVA-specific IFN-γ ELISPOT and intracellular cytokine staining
Standard model format 1 × 106 cells subcutaneously in the flank; tumors palpable within 5–7 days; adoptive transfer of 2–5 × 106 OT-I cells intravenously around day 5–7
Immunogenicity Highly immunogenic by design (defined antigen on an MSI-high background); more susceptible to immune control than parental MC38; PD-L1 upregulation and checkpoint-mediated OT-I invigoration documented
Research uses documented Antigen-presentation regulation (lncRNA EPIC1, USP22, SND1 studies); TCR-avidity and T-cell fate studies; dendritic-cell and DC-exosome vaccines; DNA neoantigen vaccines with OVA reporter epitopes; tumor-associated-macrophage cross-dressing; checkpoint combination designs
Culture conditions As parental MC38 (DMEM or RPMI-1640 + 10% FBS + glutamine + penicillin/streptomycin; 37 °C, 5% CO2), with the appropriate selection antibiotic maintained to stabilize OVA expression
Considerations Ovalbumin is a foreign model antigen requiring translational caution; antigen expression can drift or be lost under immune pressure — maintain selection and verify by Western blot / 25-D1.16 staining before implantation; construct heterogeneity across laboratories requires explicit verification; faster rejection kinetics compress windows
Representative applications Cancer vaccine discovery (peptide, mRNA, DC platforms), adoptive T-cell and TCR-engineering precursor models, checkpoint combinations with antigen-specific tracking, antigen-presentation biology, myeloid cross-dressing, T-cell exhaustion and dysfunction studies

Workflow of MC38-OVA Syngeneic Model Construction

Construction of the MC38-OVA syngeneic model is an implantation-plus-adoptive-transfer workflow whose quality rests on verified antigen expression, well-matched hosts and standardized schedules. A standard engagement proceeds as follows:

  1. Construct verification and cell qualification: MC38-OVA stocks are cleared by mycoplasma PCR and identity checks, with OVA expression confirmed by Western blot and surface SIINFEKL–H-2Kb staining (25-D1.16); the selection antibiotic is maintained through expansion to guard against antigen loss.
  2. Prepare recipients: C57BL/6 mice (6–8 weeks; wild-type, OT-I/OT-II transgenic or congenically marked depending on design; Rag2−/− where pure ACT is required) complete acclimation and health screening.
  3. Prepare the inoculum: log-phase cells are harvested, washed, counted and resuspended in PBS (commonly 1 × 107/mL for a 100 µL dose).
  4. Implant: 1 × 106 MC38-OVA cells are injected subcutaneously into the flank with consistent anatomical positioning for reproducible caliper measurement.
  5. Adoptive transfer where designed: around day 5–7, once tumors are palpable, 2–5 × 106 OT-I (or OT-II) cells are administered intravenously, with optional CFSE or congenic labeling for proliferation and persistence tracking.
  6. Randomize, dose and follow: animals are grouped by tumor volume and body weight; test articles (checkpoint agents, vaccines, adjuvants) are dosed on schedule; tumors are measured two to three times weekly, with optional serial blood draws for tetramer-based T-cell monitoring.
  7. Terminate and analyze: tumors are resected and weighed; tumor-infiltrating lymphocytes are analyzed by flow cytometry with H-2Kb–SIINFEKL tetramers and activation/exhaustion panels; sections are stained with 25-D1.16 and CD3; splenocytes undergo restimulation ICS or ELISPOT; tissues enter histopathology and molecular analysis.
