Pan02 Metastatic Tumor Model Service for Pancreas Cancer

Fig 1: Pan02 Metastatic Tumor Model for Pancreas Cancer preclinical research.

The Pan02 metastatic tumor model service for pancreas cancer provides a fully immunocompetent syngeneic platform for studying pancreatic ductal adenocarcinoma growth, local invasion, and metastatic spread. Alfa Cytology delivers an all-inclusive Pan02 metastatic tumor model service for pancreas cancer programs—from authenticated cell banking and subcutaneous or orthotopic model construction to intrasplenic and tail-vein metastasis formats, longitudinal bioluminescence imaging, and comprehensive endpoint histopathology—empowering sponsors to generate decision-grade in vivo data without standing up their own animal facilities.

Overview of Pan02 Metastatic Tumor Model for Pancreas Cancer

Pan02, also written Panc02 (Cellosaurus CVCL_D627), was established by Corbett and colleagues in 1984 by implanting a 3-methylcholanthrene-soaked suture into the pancreas of a male C57BL/6 mouse and serially transplanting the resulting ductal adenocarcinoma. The adherent, epithelial-like cells give rise to cohesive, poorly differentiated tumors that grow readily in syngeneic C57BL/6 hosts, whose intact immune systems make Pan02 the most widely used murine pancreatic ductal adenocarcinoma model in immuno-oncology. Its relatively slow tumor growth affords an ample treatment window, while its tumor microenvironment—enriched in myeloid-derived suppressor cells, regulatory T cells, macrophages, and fibroblasts with elevated TGF-β, VEGF, and IL-10—faithfully reproduces the immunosuppressive, stroma-rich nature of human pancreatic cancer.

Multi-organ tumor involvement in the Pan02 orthotopic model at days 7/14/21: H&E panels of spleen (dense tumor infiltrate), liver (tumor nodules marked T, liver L), and pancreas (tumor T compressing remnant pancreatic parenchyma P) with high-magnification insets showing tumor-pancreas boundary invasion — documenting the reproducible dissemination pattern of the Pan02 metastatic model. Because Pan02 tumors rarely metastasize from subcutaneous or orthotopic implantation sites, metastatic behavior is typically induced by alternative routes: cells injected into a hemispleen travel through the splenic vein to seed the liver, tail-vein inoculation produces experimental pulmonary metastases, and intraperitoneal injection recapitulates peritoneal dissemination with lymph node involvement. Orthotopic implantation into the pancreatic head, by contrast, models aggressive local growth with invasion of neighboring bowel, stomach, and bile duct. Luciferase-tagged Pan02 derivatives allow whole-body tumor burden to be monitored non-invasively over time, and the line’s defined responsiveness to gemcitabine—the standard-of-care reference agent—together with its well-documented chemoresistance, makes it a rigorous system for evaluating immunostimulatory strategies, anti-metastatic compounds, and combination regimens.

Tumorigenicity Forms tumors in syngeneic C57BL/6 mice (commonly 1 × 106 cells subcutaneously); relatively slow growth provides a wide treatment window
Metastatic Behavior Distant metastasis rare from subcutaneous or orthotopic sites; reliable hepatic metastasis via intrasplenic (hemisplenectomy) injection; pulmonary metastasis via tail-vein inoculation; peritoneal dissemination via intraperitoneal injection
Tumor Microenvironment Myeloid-derived suppressor cells (CD11b+Gr-1+), regulatory T cells, and macrophages dominate infiltrates; TGF-β, VEGF, and IL-10 upregulated; fibrotic stroma
Drug Response Profile Gemcitabine is the reference standard-of-care agent; low-dose gemcitabine selectively depletes intratumoral regulatory T cells; notable intrinsic chemoresistance (e.g., to cisplatin)

Mycoplasma Status Tested negative
Biosafety Level BSL-1

Alfa Cytology brings hands-on expertise in syngeneic pancreatic cancer models to every Pan02 engagement. Sponsors benefit from low-passage cell banking, feasibility-driven route selection, imaging-enabled metastasis tracking, and a complete histopathology and immune-profiling suite—executed under strict quality standards and supported by transparent, milestone-based communication that keeps preclinical timelines firmly on track.

Workflow of Pan02 Metastatic Tumor Model Construction

Developing a Pan02 metastatic tumor model combines authenticated cell preparation, route-specific surgical or systemic inoculation, and systematic endpoint evaluation. The standard workflow proceeds as follows.

  1. Cell Revival and Quality Control: Cryopreserved Pan02 cells are revived under low-passage conditions, with viability testing, mycoplasma screening, and species/identity confirmation completed before expansion.
  2. Study Design and Route Selection: The model format is chosen to match the scientific objective—subcutaneous implantation for primary tumor and immunotherapy studies, orthotopic pancreatic head injection for local invasion, intrasplenic hemisplenectomy for hepatic metastasis, tail-vein injection for lung metastasis, or intraperitoneal injection for peritoneal dissemination.
  3. Cell Preparation: Log-phase cells are harvested, counted, and resuspended in PBS or HBSS; Matrigel admixture is applied for orthotopic injections to minimize peritoneal spillage, and Pan02-luc cells are selected when longitudinal imaging is required.
  4. Tumor Inoculation: Cells are implanted under aseptic conditions via the designated route—for example, 1 × 106 cells injected subcutaneously, or a defined dose delivered into the splenic pulp during hemisplenectomy surgery for liver metastasis studies.
  5. Randomization and Dosing: Animals are randomized into treatment groups once baseline tumor signals are established, and dosing of test articles begins according to the approved study plan.
  6. In-Life Monitoring: Tumor dimensions and body weight are recorded on schedule; tumor burden and metastatic spread are tracked by weekly in vivo bioluminescence imaging, supplemented where needed by ultrasound or MRI.
  7. Endpoint Analysis: At study termination, liver, lung, and peritoneal sites are examined for metastatic nodules; tissues are processed for H&E staining, immunohistochemistry, and flow cytometric analysis of myeloid-derived suppressor cells, regulatory T cells, and macrophage populations.
  8. Data Analysis and Reporting: Growth curves, imaging time courses, metastasis counts, and histology findings are statistically analyzed and consolidated into a comprehensive final report.

Case Study-Pan02 Metastatic Tumor Model Development

In a recent preclinical collaboration, Alfa Cytology assisted a sponsor in profiling candidate therapeutics against Pan02-based pancreatic cancer models. Following format selection and feasibility work, tumor-bearing animals were assigned to randomized treatment arms, disease burden was followed by serial imaging, and multi-organ histopathological and immunological endpoints were assessed. The engagement highlights our capacity to run Pan02 metastatic tumor model studies spanning immunotherapy, anti-metastasis, and combination strategies, with designs adjusted as sponsor needs evolve; representative outcomes and additional case details can be provided on request.
