Neuro-2a (N2a) Syngeneic Model Service for Neuroblastoma

Fig 1: Neuro-2a (N2a) Syngeneic Model for Neuroblastoma preclinical research.

The Neuro-2a (N2a) syngeneic model is the most widely distributed C-1300-lineage neuroblastoma platform in existence, engrafting the World Organisation for Animal Health's rabies-diagnosis reference line into syngeneic A/J hosts where validated vaccine, oncolytic-virotherapy, and intracranial formats meet half a century of neurobiology. Alfa Cytology delivers this uniquely credentialed model as a complete preclinical service — authenticated ATCC banking, subcutaneous and stereotactic intracranial implantation, whole-cell vaccine and oncolytic-HSV benchmarking, and full histopathology — giving sponsors the neuroblastoma system whose virus biology is understood at the regulatory-standard level.

Overview of Neuro-2a (N2a) Syngeneic Model for Neuroblastoma

Neuro-2a is the most famous descendant of the C-1300 tumor — the spontaneous spinal-cord-region neuroblastoma of a strain A albino mouse from which the entire murine neuroblastoma field descends — and its establishment in 1969 by Robert Klebe and Frank Ruddle made history in its own right: the line was conceived as a differentiating stem-cell system, and within a year Olmsted and colleagues had isolated microtubule protein from it for the first time, launching the modern biochemistry of the neuronal cytoskeleton. Deposited as ATCC CCL-131 and now held by repositories on every continent, Neuro-2a produces large amounts of tubulin and acetylcholinesterase, extends neurites on serum withdrawal or retinoic-acid treatment, and carries a distinction no other tumor line in this catalog can claim: the World Organisation for Animal Health designates it for routine rabies diagnosis, making it a global regulatory reference substrate whose virus-handling biology is understood with unusual precision. That virological fluency is directly exploitable — the line is documented as susceptible to herpes simplex virus, vesicular stomatitis virus, and poliovirus, it propagates rabies and prions (the ScN2a scrapie model), and the A/J host strain itself is HSV-sensitive, a pairing that made Neuro-2a the reference system for oncolytic HSV neuroblastoma research.

As an immunocompetent tumor model, Neuro-2a has accumulated a vaccine literature of unusual depth in A/J hosts. Subcutaneously, 106 flank cells grow fast enough to treat within days, and the cytokine-augmented whole-cell vaccine lineage built on the line is among the best quantified in neuroblastoma: IL-2/IL-12 double-transfected Neuro-2a cells abolish tumorigenicity — 83% of vaccinated mice remaining tumor-free beyond 40 days — and three intratumoral doses into palpable established tumors shrink mean day-12 volumes to 36 ± 47 mm3 against 767 ± 423 mm3 in controls, with CD4+ and CD8+ T cells both implicated and rechallenge protection documented; engineered costimulatory variants expressing ICAM-1, CD80, CD86, and 4-1BBL extend the vaccine-design space. The intracranial format is equally distinguished: stereotactic placement of just 104 cells into the right caudate nucleus is uniformly lethal by day 16, yet a premunition prime/boost regimen with M002 — an IL-12-expressing, γ134.5-deleted oncolytic HSV loaded into irradiated tumor cells — produced a significant survival advantage with protection against intracranial rechallenge and exquisite specificity, since the unrelated syngeneic H6 hepatoma grew without restraint in the same protected animals. Two honest caveats frame the design space: the line carries single-copy, non-amplified MYCN and is poorly immunogenic, so programs modeling high-risk MYCN-amplified disease pair it with complementary models. For sponsors in vaccines, oncolytic virotherapy, or CNS-format immunotherapy, Neuro-2a offers a fifty-year track record and a virological pedigree without parallel.

Cell Line Information: Neuro-2a (N2a) Syngeneic Model

Neuro-2a (ATCC CCL-131; Cellosaurus CVCL_0470) is the Klebe and Ruddle 1969 culture line from the C-1300 spontaneous neuroblastoma of a strain A mouse, distributed by the American Type Culture Collection, JCRB, and repositories worldwide. Engineered vaccine transfectants and the prion-adapted ScN2a derivative extend the platform. Core characteristics are compiled below.

Parameter Description
Cell line Neuro-2a (N2a; Neuro 2a; N2A; Nb2a; NB2a); prion-adapted derivative ScN2a; engineered vaccine variants including IL-2/IL-12 transfectants and ICAM-1/CD80/CD86/CD137L costimulatory transfectants
Catalog references ATCC CCL-131; Cellosaurus CVCL_0470; JCRB/IFO50081; BCRJ 0189; RIKEN RCB0492; widely distributed internationally
Species / strain Mus musculus, A/J (syngeneic with A/J hosts; H-2a)
Tissue of origin Spinal cord region; sympathetic nervous system-derived neuroblast
Disease Mouse neuroblastoma
Establishment Established in 1969 by R. J. Klebe and F. H. Ruddle from the C-1300 spontaneous tumor of an albino strain A mouse (Jackson Laboratory), adapted to tissue culture as a differentiating stem cell system (Klebe & Ruddle, J. Cell Biol. 43:69A, 1969); microtubule-protein isolation from the line reported by Olmsted et al., PNAS 65:129–136 (1970)
Lineage heritage One of several clonal derivatives of C-1300 — the first transplantable murine neuroblastoma; Neuro-2a is the most widely distributed member of the family
World-standard identity Designated by the World Organisation for Animal Health (WOAH, formerly OIE) for routine rabies diagnosis — one of the few tumor lines serving as a global regulatory standard; standard substrate for rabies virus titration worldwide
Morphology / growth Neuronal and amoeboid-like morphology; adherent growth; doubling time ~20 hours (JCRB characterization) to 36–48 hours (vendor data); produces large amounts of tubulin and acetylcholinesterase
Virus susceptibility Susceptible to herpes simplex virus, vesicular stomatitis virus, and human poliovirus 1 — the HSV susceptibility of the line, combined with the HSV-sensitivity of the A/J host background, makes it the reference substrate for oncolytic HSV neuroblastoma studies; propagates rabies virus; supports prion replication (ScN2a model)
Oncogene status Single-copy MYCN without amplification — an honest caveat for high-risk MYCN-amplified neuroblastoma questions; poorly immunogenic baseline
Complete growth medium DMEM with 1% non-essential amino acids, 2 mM L-glutamine, 4.5 g/L glucose, and 10% FBS (ATCC/BCRJ formulation); MEM + 10% FBS also standard (JCRB/OriCell); DMEM/F12 + 7% FBS used in the intracranial vaccine literature
Culture conditions 37 °C, humidified atmosphere of 95% air and 5% CO2
Biosafety level BSL-1
Subcutaneous tumorigenicity 106 cells in the posterior flank of A/J mice: fast-growing tumors measurable within ~3 days; validated therapeutic-vaccine platform — IL-2/IL-12-transfected whole-cell vaccines abolish tumorigenicity (83% tumor-free) and retard established tumors (36 ± 47 mm3 versus 767 ± 423 mm3 in controls at day 12, P < 0.001)
Intracranial tumorigenicity Stereotactic implantation of 1 × 104 cells into the right caudate nucleus of A/J mice — uniformly lethal by day 16 in controls; premunition prime/boost vaccination with M002 HSV-IL12-loaded tumor cells confers significant survival advantage (P = 0.00002) with protection against intracranial rechallenge and specificity demonstrated by unrestricted growth of unrelated syngeneic H6 hepatoma
Engineered vaccine lineage Costimulatory transfectants expressing ICAM-1 (CD54), CD80, CD86, and CD137L (4-1BBL) validated as tumor vaccines; IL-2/IL-12 double-transfectant lineage anchors the cytokine-augmented whole-cell vaccine literature
Primary applications Neuroblastoma vaccine and oncolytic virotherapy development, intracranial immunotherapy formats, rabies and neurotropic virus research, prion biology, microtubule and axonal-transport pharmacology, neurotoxicity screening

Our Services

Engaging Alfa Cytology for the Neuro-2a syngeneic model places the most credentialed neuroblastoma line under one accountable team: authenticated ATCC-provenance banks are maintained with mycoplasma-free certification, syngeneic A/J hosts are sourced and acclimated, tumors are established by the subcutaneous route at the validated 106-cell geometry or stereotactically into the caudate nucleus at the documented 104-cell inoculum, and treatment arms — whole-cell vaccine formats, oncolytic HSV regimens, checkpoint combinations, or differentiation-therapy modalities — run through scheduled caliper monitoring or survival analysis to terminal histopathology with neural and immune-marker panels, all under IACUC-compliant conditions and returned as an audit-ready data package.

Workflow of Neuro-2a (N2a) Syngeneic Model Construction

Neuro-2a studies divide into the therapeutic flank format — where vaccine and virotherapy arms are benchmarked against published response curves — and the stereotactic intracranial format, where survival and rechallenge protection are the definitive readouts. A standard program proceeds as follows.

  1. Cell preparation and quality control: Neuro-2a cells are expanded at low passage, confirmed mycoplasma-free, and identity verified against the CCL-131 reference; for vaccine arms, engineered transfectants (IL-2/IL-12, costimulatory variants) are expression-verified before use.
  2. Host selection and acclimation: Female or male A/J mice, typically 5–8 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility.
  3. Implantation: subcutaneous route. 106 cells in 0.1 mL sterile PBS are injected into the posterior flank; the site is compressed briefly, and palpable tumors (~2 × 2 mm) are typically present within 3 days, supporting therapeutic-vaccine timing.
  4. Implantation: stereotactic intracranial route. Under isoflurane anesthesia in a stereotactic frame, 1 × 104 cells in a small volume are infused into the right caudate nucleus at the documented coordinates; the burr hole is sealed and mice recover with analgesia and neurological observation.
  5. Vaccination or treatment administration. Test articles: whole-cell vaccines (irradiated or cytokine-engineered), oncolytic HSV regimens, checkpoint inhibitors, or combination formats — are administered per protocol; published benchmark schedules include intramuscular vaccination at days −7 and +7 (premunition prime/boost) or intratumoral dosing at days 3, 5, and 7 for established flank tumors.
  6. Longitudinal monitoring: Flank tumors are measured by caliper every 2–3 days against the published day-12 benchmark window; intracranial cohorts are monitored daily for survival and neurological signs, with humane endpoints predefined.
  7. Rechallenge and specificity arms where specified: Long-term survivors are rechallenged with Neuro-2a in the contralateral flank or caudate nucleus, and specificity controls receive the unrelated syngeneic H6 hepatoma, following the established rechallenge convention.
  8. Endpoint analysis and reporting: At termination, tumors and brains are processed; tissues undergo H&E with neuroblastoma histology, immunohistochemistry (CD8, CD4, FoxP3, F4/80, PD-L1, neuronal-marker panels), splenocyte cytotoxicity assays against Neuro-2a targets, and tumor-infiltrating-lymphocyte flow cytometry, with Kaplan–Meier survival analysis and all data consolidated into the final report.

Case Study-Neuro-2a (N2a) Syngeneic Model

Alfa Cytology has applied the Neuro-2a syngeneic platform to preclinical programs spanning whole-cell and cytokine-augmented vaccines, oncolytic HSV regimens, checkpoint combinations, and intracranial immunotherapy formats in neuroblastoma. In these engagements, caliper readouts tracked flank tumor kinetics against published benchmark windows, intracranial cohorts were followed for survival with rechallenge protection as the durability readout, and terminal histopathology with neural and immune-marker panels plus splenocyte cytotoxicity assays completed the immunological assessment. Study parameters — implantation format, vaccination schedule, transfectant selection, and endpoint panels — are tailored to each program. Case materials can be shared under confidentiality agreements upon request.
