KPC (derived) / KPC-luc Orthotopic Mouse Model Service for Pancreas Cancer

Fig 1: KPC (derived) / KPC-luc Orthotopic Mouse Model for Pancreas Cancer preclinical research.

The KPC (derived) / KPC-luc orthotopic mouse model recreates human pancreatic ductal adenocarcinoma in its native organ within an immunocompetent host, making it one of the most translationally relevant platforms for preclinical pancreas cancer research. Alfa Cytology is a preclinical-focused oncology CRO experienced in genetically faithful syngeneic modeling. Alfa Cytology offers complete KPC (derived) / KPC-luc orthotopic model services, covering cell line authentication, luciferase labeling verification, surgical implantation, longitudinal in vivo imaging, and in-depth endpoint characterization.

Overview of KPC (derived) / KPC-luc Orthotopic Mouse Model for Pancreas Cancer

The KPC model is a genetically engineered mouse model of pancreatic ductal adenocarcinoma (PDAC) in which an activating KrasG12D mutation and a dominant-negative Trp53R172H mutation are conditionally expressed in the pancreatic epithelium through Pdx-1-driven Cre recombinase. These mice spontaneously progress from pancreatic intraepithelial neoplasia to invasive, metastatic PDAC that closely mirrors the human disease, including dense desmoplastic stroma, prominent macrophage infiltration, and T cell exclusion. From the primary tumors of these animals, low-passage KPC-derived cell lines can be established that retain both driver mutations and the histological and immunobiological features of the parental tumors.

Fig 2: Reference figures for KPC (derived) / KPC-luc cell-related literature.Fig 1. Orthotopic implantation time course showing KPC-Luc total flux peaking at day 9 (~4.5×10⁹ p/s) then declining to background by day 12 while parental KPC tumors grew progressively, with day 9/12 whole-mouse IVIS images, quantified tumor volume (KPC-Luc ~10 mm³ vs KPC ~90 mm³) and metastasis score (P<0.05)—documenting that luciferase expression induces immunogenic rejection of KPC-Luc tumors in immunocompetent mice. (Ferrari, Daniele Pereira, et al., 2024)

When such KPC-derived cells are implanted orthotopically into the pancreas of syngeneic C57BL/6 mice, the resulting tumors grow with rapid, predictable kinetics while preserving the fibrotic stroma and immune architecture characteristic of human PDAC — a decisive advantage over immunodeficient xenograft systems. Because the host immune system remains fully intact, the model is exceptionally well suited to in vivo evaluation of immunotherapies, stroma-targeting agents, KRAS pathway inhibitors, and combination regimens. The luciferase-tagged variant, KPC-luc, enables non-invasive, quantitative in vivo bioluminescence tracking of tumor burden and treatment response, providing sensitivity that caliper measurements cannot achieve in deeply seated pancreatic tumors.

Cell Line Information: KPC (derived) / KPC-luc

Unlike conventional immortalized lines, KPC-derived cells are low-passage tumor cell lines generated directly from PDAC arising in KPC genetically engineered mice. Their defining characteristics are summarized below.

Cell line name KPC (derived) — low-passage PDAC cell lines established from primary pancreatic tumors of KPC mice
Labeled variant KPC-luc — KPC-derived cells engineered to stably express firefly luciferase (often as a Luc-GFP fusion), enabling bioluminescence imaging; reporter-positive populations are typically purified by FACS
Origin model KPC genetically engineered mouse model: LSL-KrasG12D/+; LSL-Trp53R172H/+; Pdx1-Cre, first described in 2005
Species / strain Mouse (Mus musculus), C57BL/6 genetic background (original model on mixed 129Sv/C57BL/6, extensively backcrossed)
Disease Pancreatic ductal adenocarcinoma (PDAC)
Genotype Endogenous KrasG12D activating mutation and Trp53R172H dominant-negative mutation, conditionally activated by Cre-mediated recombination in the pancreatic lineage
Morphology Epithelial-like, adherent monolayer
Growth properties Anchorage-dependent adherent growth at low passage; retains tumorigenicity and parental tumor characteristics
Recommended medium DMEM + 10% FBS with penicillin/streptomycin (RPMI-1640 + 10% FBS is also widely used)
Culture conditions 37 °C, humidified 5% CO2 atmosphere
Key biological features Retains driver mutations of the parental KPC tumors; recapitulates dense fibrosis/desmoplasia, F4/80+ macrophage infiltration, and T cell exclusion after implantation
Immunogenicity Poorly immunogenic; in vivo growth rates comparable in immunodeficient and immunocompetent hosts
Host for implantation Syngeneic C57BL/6 mice (immunocompetent); also grows in immunodeficient strains
Orthotopic inoculum Commonly 0.5 × 104 to 5 × 105 cells in approximately 50 μL saline or PBS, injected into the pancreatic parenchyma by laparotomy; published protocols vary by line and study design
Metastatic pattern Primary pancreatic tumors with spontaneous spread to liver, lung, diaphragm, peritoneum, and lymph nodes
Bioluminescence imaging Intraperitoneal D-luciferin (e.g., 150 mg/kg) followed by acquisition on an IVIS-type in vivo imaging system; imaging 1–2 times per week is typical
Derivation method Primary tumors minced and enzymatically dissociated (collagenase/dispase), plated on collagen-coated dishes, passaged to eliminate fibroblast contamination
Quality control Genotype confirmation of KrasG12D and Trp53R172H, routine mycoplasma testing, and pre-injection viability assessment (typically >90%)
Typical applications Preclinical evaluation of immunotherapies, KRAS pathway inhibitors, stromal-targeting agents, oncolytic approaches, and combination strategies

Workflow of KPC (derived) / KPC-luc Orthotopic Mouse Model Construction

Constructing a KPC (derived) orthotopic model integrates tumor cell biology, microsurgery, and in vivo imaging. The standard workflow proceeds through the following stages.

  1. Cell line verification and expansion: KPC (derived) or KPC-luc cells are expanded at low passage, genotyped to confirm KrasG12D and Trp53R172H, tested for mycoplasma, and assessed for viability; luciferase expression in KPC-luc is validated by an in vitro bioluminescence assay.
  2. Syngeneic host preparation: Immunocompetent C57BL/6 mice, typically 6–8 weeks of age, are acclimatized and randomized under an approved animal ethics protocol.
  3. Inoculum preparation: Log-phase cells are harvested into a single-cell suspension in saline or PBS at the designated dose (commonly in the 0.5 × 104 to 5 × 105 cell range), with viability reconfirmed immediately before surgery.
  4. Orthotopic implantation by laparotomy: Under anesthesia and aseptic conditions, a small left-flank incision exposes the pancreas, and the cell suspension is injected directly into the pancreatic parenchyma; the site is inspected for leakage before closure.
  5. Post-operative monitoring: Analgesia and supportive care are provided, and animals are observed daily until recovery, with body weight and clinical status recorded throughout.
  6. Bioluminescence imaging and randomization (KPC-luc): Beginning shortly after implantation, mice receive intraperitoneal D-luciferin and are imaged to establish baseline signals; subsequent weekly in vivo imaging sessions quantify tumor growth and metastatic spread, and animals are randomized to treatment groups once signals reach predefined thresholds.
  7. Treatment administration (optional): Test articles — including immune checkpoint inhibitors, stromal modulators, KRAS pathway inhibitors, or combinations — are dosed according to the customized study design.
  8. Endpoint analysis: At termination, mice undergo necropsy with measurement of primary pancreatic tumor weight and dimensions, enumeration of metastatic lesions in liver, lung, and lymph nodes, and tissue collection for H&E, Masson trichrome or IHC staining of fibrosis and immune infiltrates, plus optional flow cytometry, followed by statistical analysis and report delivery.
