KYSE-510-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: KYSE-510-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

KYSE-510-luc Orthotopic Mouse Model Service for Esophageal Cancer provides a bioluminescence-readable xenograft system built on one of the few well-differentiated ESCC lines, allowing tumors to grow in the esophageal wall while their burden is quantified non-invasively over time. Alfa Cytology delivers this model as a complete preclinical package—luciferase-verified cell banking, organ-accurate surgical implantation, scheduled imaging sessions, and thorough histopathological endpoints—so researchers gain both visual and quantitative evidence of treatment effect within a single study.

Overview of KYSE-510-luc Orthotopic Mouse Model for Esophageal Cancer

KYSE-510 is a human esophageal squamous cell carcinoma (ESCC) line with a distinctive pedigree: it was established from a well-differentiated cervical-esophageal tumor with moderate adventitial invasion, resected from a 67-year-old female patient who had already received cisplatin and radiotherapy. The line retains a well-differentiated squamous phenotype in culture—epithelioid cells growing as a monolayer with multilayer foci—and forms well-differentiated tumors when implanted in nude mice. The KYSE-510-luc derivative adds stable firefly luciferase expression to this background, permitting tumor establishment and progression to be followed by in vivo bioluminescence imaging after D-luciferin administration.

An orthotopic version of this model places luciferase-labeled cells directly into the esophageal wall, where they develop in the company of the organ’s native stroma, vasculature, and lymphatic drainage rather than on the flank. This matters experimentally: well-differentiated histology is preserved, transmural growth patterns emerge, and longitudinal light output traces each animal’s tumor trajectory with far greater sensitivity than periodic palpation. KYSE-510’s documented biology—TP53 mutation, amplification of MYC and CCND1, microsatellite-stable status, and an expression profile close to that of ECA-109—further equips the model for mechanistic work, from epithelial–mesenchymal transition and invasion studies to evaluations of differentiation-directed or DNA-damage-based therapies.

Cell Line Information: KYSE-510-luc

The parental KYSE-510 line is among the most thoroughly characterized ESCC models, held by the DSMZ (ACC 374) and JCRB collections and profiled in the CCLE/DepMap and COSMIC projects. Its luciferase-tagged derivative extends these credentials into imaging-based preclinical research. The table below consolidates the key documented attributes.

Our Services

Alfa Cytology approaches the KYSE-510-luc orthotopic model with a histology-first mindset: because the line carries a genuinely well-differentiated phenotype, implantation, imaging, and endpoint analysis are all calibrated to preserve that biological identity rather than force it into a generic xenograft workflow. The result is a study in which photon counts, growth curves, and H&E sections tell one coherent story, delivered with the documentation quality your internal review boards expect from preclinical work.

Workflow of KYSE-510-luc Orthotopic Mouse Model Construction

Constructing a luciferase-enabled orthotopic model with KYSE-510 involves parallel preparation of the reporter cells and the surgical program, converging at implantation and continuing through imaging to pathology. A typical engagement unfolds as follows:

  1. Cell authentication and reporter confirmation: KYSE-510 cultures are STR-profiled against the reference database, mycoplasma-tested, and screened for stable luciferase expression by in vitro light output before expansion.
  2. Master and working bank preparation: Qualified vials are prepared at controlled passage numbers so that all animals in a study derive from one characterized cell bank.
  3. Surgical-day cell preparation: On the day of surgery, cells are thawed or harvested from log-phase flasks, counted, viability-checked, and concentrated into a small-volume inoculum (on the order of 1 × 105 cells in a 10 μL volume) for wall-layer implantation.
  4. Anesthesia and esophageal exposure: Immunodeficient mice are anesthetized, and the esophagus is exposed through a brief abdominal or cervical approach with aseptic technique and intraoperative analgesia.
  5. Orthotopic implantation: The KYSE-510-luc suspension is injected into the esophageal wall under magnification, with the needle track controlled to keep the inoculum within the wall layers rather than the lumen or adjacent tissues.
  6. Recovery and imaging schedule: Animals recover under observation, then enter a predefined imaging calendar in which D-luciferin is administered and photon flux is recorded at consistent time points to build longitudinal growth curves.
  7. Randomization and treatment: When tumor signals plateau within the target window, mice are assigned to vehicle or treatment arms and dosed according to the study design, with imaging continuing through the treatment phase.
  8. Terminal histopathology: At endpoint, esophageal tissues are harvested intact for caliper measurement, H&E staining to confirm well-differentiated squamous histology, and immunohistochemical or molecular assays as specified.

Case Study-KYSE-510-luc Orthotopic Mouse Model Development

Alfa Cytology has supported preclinical programs that called for a luciferase-labeled, organ-matched esophageal cancer model with preserved differentiation status, delivering studies that combined surgical orthotopic implantation with serial bioluminescence readouts and histopathological confirmation. These engagements are documented internally and can be discussed under confidentiality, providing prospective clients with practical benchmarks on model behavior, imaging performance, and study duration before committing to a full program.
