EL4 Syngeneic Model Service for Lymphoma

Fig 1: EL4 Syngeneic Model for Lymphoma preclinical research.

The EL4 syngeneic model service for lymphoma is an immune-competent C57BL/6 platform built on the oldest continuously used chemically induced T-cell lymphoma line in existence, supporting subcutaneous, intradermal, and systemic study formats alongside the field-standard EG7-OVA model antigen system. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated EL4 / EG7-OVA expansion with low-passage banking, implantation into C57BL/6 hosts across validated formats, scheduled caliper and in vivo imaging monitoring, and full lymphoma histopathology with T-cell and costimulation immunophenotyping — all configured around each sponsor’s vaccine, adoptive-therapy, costimulation, or CAR-T hypothesis.

Overview of EL4 Syngeneic Model for Lymphoma

EL4 is a genuine relic of the founding era of tumor immunology: established by Gorer in 1950 from a lymphoma induced in a C57BL/6 mouse by the carcinogen 9,10-dimethyl-1,2-benzanthracene (DMBA), it is the oldest continuously cultured murine lymphoma line and one of the oldest cancer cell lines of any kind. ATCC classifies it as a mouse precursor T-cell lymphoblastic lymphoma/leukemia; the cells carry H-2b and Thy-1.2, retain the T-cell character that made them indispensable to T-cell biology, and carry the quirks of their era — low-level expression of Gross murine leukemia virus (type G) antigen, documented interleukin production (IL-4 per ECACC, and classic IL-2 production upon stimulation), sensitivity to phytohemagglutinin, and a striking resistance to cortisol and dexamethasone at 10−4 M that has been exploited in glucocorticoid-response research. A caution documented in the modern literature deserves emphasis: extended passage drives genetic drift and progressively increased immunogenicity in vivo, so low-passage banking is a genuine scientific necessity for this line, not a formality.

In vivo, the parental line is a reliable grower — intradermal implantation of 2 × 106 cells into syngeneic C57BL/6 mice produced tumors in 20 of 20 animals — and it was on this dependability that some of immunology’s most influential experiments were built: the landmark B7 costimulation studies in which EL4 transfected with B7-1 was rejected in 28 of 30 mice while parental tumors grew unchecked, establishing the costimulatory paradigm that underpins modern checkpoint biology. The line’s most consequential derivative is EG7-OVA — EL4 transfected with a complete chicken ovalbumin cDNA — which became the single most widely used model antigen system in cancer immunology, powering DNA- and viral-vector vaccine studies, OT-1 adoptive T-cell therapy experiments, radiation-plus-Th1 cell therapy work, and nanoparticle-engineered cell platforms; published protocols span 2 × 105 to 4 × 106 cells depending on format, with tumors measurable within one to two weeks. Gene-humanized derivatives now extend the platform into CAR-T territory — B-hBCMA-luc EL4, engineered with a chimeric human/mouse BCMA and luciferase, is a documented validation substrate for BCMA-directed cell therapies. Seventy-five years after its derivation, EL4 remains where T-cell lymphoma immunotherapy ideas are first tested.

Cell Line Information: EL4

The table below consolidates the key biological, cultural, and modeling characteristics of the EL4 line and its EG7-OVA and humanized-target derivatives.

Origin Established by Gorer in 1950 (British Journal of Cancer 4:372) from a lymphoma induced in a C57BL/6 mouse by systemic 9,10-dimethyl-1,2-benzanthracene (DMBA); later part of the Tumor Immunology Bank collection transferred from Salk to ATCC in 1981
Depository / catalogue ATCC TIB-39; Cellosaurus CVCL_0255; ECACC 85023105; DSMZ ACC-831; JCRB1350; EG7-OVA at ATCC CRL-2113
Tumor type Mouse precursor T-cell lymphoblastic lymphoma/leukemia (ATCC/NCIt C21925); classical murine thymoma model
Karyotype and growth Karyotype reported at 2n = 39; population doubling time approximately 12–20 hours across catalogue records; suspension lymphoblasts maintained at 3–9 × 105 cells/mL
Immunophenotype H-2b and Thy-1.2 positive; low-level Gross murine leukemia virus (type G) surface antigen reported (Journal of the National Cancer Institute 48:265, 1972); interleukin production documented (IL-4 per ECACC; classic IL-2 production upon stimulation); phytohemagptenin-sensitive; resistant to cortisol and dexamethasone at 10−4 M
Passage caution Extended passage causes genetic drift with progressively increased in vivo immunogenicity — low-passage banking and authentication are essential quality steps for this line
Recommended culture conditions DMEM with 2 mM glutamine and 10% fetal bovine serum (ECACC formulation), or RPMI-1640 with 10% FBS and 0.05 mM 2-mercaptoethanol; maintain at 3–9 × 105 cells/mL with regular medium renewal
Implantation formats Intradermal: 2 × 106 cells per mouse, tumors in all recipients with volumes ≈1,100 mm³ by day 14; subcutaneous: EG7-OVA protocols span 2 × 105 to 4 × 106 cells, measurable tumors in 7–14 days; contralateral EL4/EG7-OVA designs (4 × 106 EG7 plus 2 × 106 EL4) enable antigen-specificity controls in OT-1 adoptive therapy; intravenous and intraperitoneal routes supported for dissemination studies
Documented in vivo responses Parental EL4 grows in 20/20 C57BL/6 recipients while B7-1-transfected EL4 is rejected in 28/30 with 60% tumor-free survivors — the foundational costimulation experiment; DNA-vaccine and viral-vector vaccination regimens protect against EG7-OVA challenge and shrink established tumors; local radiation combined with Th1 cell therapy inhibits growth through tumor-specific CTLs
Engineered derivatives EG7-OVA (ATCC CRL-2113) presents OVA peptide SIINFEKL on H-2Kb for CD8-tetramer, OT-1 TCR-transgenic, and B3Z-reporter studies; B7-1/B7-2 transfectants established the costimulation paradigm; B-hBCMA-luc EL4 carries a chimeric human/mouse BCMA with luciferase for CAR-T validation; membrane-anchored Gaussia luciferase versions support bioluminescent tracking

Key research applications T-cell lymphoma immunotherapy and costimulation studies; vaccine platform benchmarking using the EG7-OVA model antigen; adoptive T-cell therapy and combination radiation-plus-cell-therapy evaluation; checkpoint and glucocorticoid-response research; CAR-T target validation with gene-humanized derivatives; nanoparticle-engineered cell delivery and bioluminescent tracking

Our Services

A line this venerable is only as good as the passage discipline behind it, and Alfa Cytology runs EL4 accordingly: low-passage authenticated banks with documented immunogenicity controls, validated implantation formats for parental and EG7-OVA arms, scheduled caliper and in vivo imaging monitoring, and endpoint pathology tuned to T-cell lymphoma — all under one accountable preclinical operation.

Workflow of EL4 Syngeneic Model Construction

Construction follows the published methodologies for the platform’s deployment formats: EL4 or EG7-OVA cells are expanded under authenticated low-passage conditions, implanted intradermally or subcutaneously into C57BL/6 mice, grown to a standardized baseline, randomized, treated, and analyzed with antigen-specific and histopathological endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: EL4 or EG7-OVA cells are identity-verified, confirmed mycoplasma-negative, documented for low passage number, and re-checked for antigen expression (OVA for EG7 arms), then expanded into working banks under standardized DMEM or RPMI-1640 culture conditions.
  2. Cell harvest and formulation: Log-phase cultures are counted and viability-checked, then resuspended at the study-specified dose of approximately 2 × 105 to 4 × 106 cells in 100 – 200 μL of PBS or a Matrigel-supported suspension per the validated format.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane; cells are implanted intradermally or subcutaneously into the flank, with contralateral EL4/EG7-OVA designs used where antigen-specificity controls are required.
  4. Establishment monitoring and randomization: Tumors are measured by caliper from first palpability (typically 7–14 days); luciferase-arm studies include in vivo imaging sessions; animals are randomized once tumors reach protocol-specified baselines.
  5. Treatment administration: Experimental arms — vaccines, adoptive OT-1 or transgenic T-cell transfer, checkpoint and costimulation reagents, radiation fractions, CAR-T against humanized targets, and combinations — proceed per study design with body-weight and toxicity monitoring.
  6. Antigen-specific immune monitoring: Draining-lymph-node, splenic, and tumor-infiltrating populations are sampled on a study-defined schedule for OVA-tetramer⁺ CD8⁺ T-cell quantification, cytotoxicity assays against EL4 versus EG7 targets, and cytokine profiling by ELISPOT and intracellular staining.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition and survival with histopathology (H&E confirming T-lymphoblastic morphology, plus CD3, CD8, Ki67, and marker-specific immunohistochemistry), ex vivo antigen-specificity confirmation in reporter arms, and terminal immune-context profiling.
