KP (KP1 / KP4) Syngeneic Model Service for Lung Cancer

Fig 1: KP (KP1 / KP4) Syngeneic Model for Lung Cancer preclinical research.

The KP (KP1 / KP4) syngeneic model service for lung cancer is an immune-competent C57BL/6 platform built on tumor-derived lines from the KrasG12D;Trp53 conditionally null (KP) genetically engineered mouse model — the gold-standard lung adenocarcinoma genotype — supporting flank-efficacy, orthotopic, and checkpoint-combination study designs. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated KP1 / KP4 expansion and banking, subcutaneous or intrapulmonary implantation, scheduled caliper or micro-CT monitoring, and full adenocarcinoma histopathology with tumor-microenvironment profiling — all configured around each sponsor’s Kras-pathway, checkpoint, or combination-therapy hypothesis.

Overview of KP (KP1 / KP4) Syngeneic Model for Lung Cancer

The KP designation refers to the most consequential genotype in lung adenocarcinoma modeling: KrasG12D activation combined with Trp53 deletion, engineered conditionally in C57BL/6 mice (Jackson Laboratory strain 032435) and induced in the lung epithelium by intratracheal or intranasal adenoviral Cre delivery — a system that recapitulates the stepwise progression from adenoma to invasive adenocarcinoma and remains the reference genetic framework for KRAS-driven human lung cancer. From these autochthonous tumors, cell lines can be established that retain the engineered genotype and grow in fully immunocompetent syngeneic C57BL/6 hosts; the lines designated KP1 and KP4 in the preclinical CRO literature (also listed as KP1-lung and KP4-lung) belong to this family, as do published relatives such as the independently characterized KP1–3 lines and KPL86. Documented characterization of KP-family lines confirms the expected KrasG12D recombination and Trp53 loss, anchorage-independent growth, and a molecular posture of high HMGA2 with low NKX2.1 that tracks with aggressive in vivo behavior.

What distinguishes the KP syngeneic system is its translational wiring. Because the driver lesion is the same KrasG12D found in human lung adenocarcinoma, the platform has served the targeted-therapy era directly: autochthonous KP tumors — which activate CDK4 signaling in their p53-null state — respond to the CDK4/6 inhibitor palbociclib, and concomitant CDK4 plus MAPK-pathway blockade (palbociclib with the MEK inhibitor AZD6244) augments response in Kras-mutant tumors, establishing the therapeutic logic now pursued in the clinic. As a transplantable syngeneic system, KP lines support both flank grafts for rapid efficacy screening and intrapulmonary orthotopic implantation — the format that best preserves lung microenvironmental context for tumor-host interaction and immunotherapy studies, with interferon-γ inducible PD-L1 expression providing a mechanistic bridge to checkpoint sensitivity. For sponsors, KP1/KP4 thus offer the rare combination of a bona fide oncogene-defined lung adenocarcinoma genotype, immune-competent C57BL/6 hosting, orthotopic capability, and a benchmarked targeted-therapy rationale in a single platform.

Cell Line Information: KP (KP1 / KP4)

The table below consolidates the key biological, genetic, and modeling characteristics of the KP-derived syngeneic lines, cross-referenced with the parental genetically engineered model.

Standard implantation practice Flank: approximately 5 × 105 cells in a 50% HBSS/50% Matrigel suspension for syngeneic efficacy studies; orthotopic: approximately 2.5 × 105 cells in 40 μL into the left lung through a surgical approach, with micro-CT monitoring documented for response assessment
Documented in vivo therapeutic responses Autochthonous KrasG12D;Trp53-null lung tumors respond to CDK4/6 inhibition (palbociclib), with augmented response when MAPK-pathway blockade (AZD6244) is added concomitantly — the combination rationale benchmarked on the KP genotype; orthotopic C57BL/6 platforms of this class support anti-PD-1 sensitivity profiling via interferon-γ-inducible PD-L1
Host strains C57BL/6 immunocompetent mice (syngeneic); knockout and reporter variants used for mechanism studies
Key research applications KrasG12D-targeted therapy and MEK/CDK4-combination evaluation; checkpoint immunotherapy in an oncogene-defined lung adenocarcinoma; orthotopic-versus-flank microenvironment comparison; tumor-immune microenvironment dissection; imaging-tracked (micro-CT, bioluminescence) response studies; GEMM-to-syngeneic translational bridging

Our Services

An oncogene-defined lung platform earns its keep only when genotype authentication and microenvironment fidelity are both enforced, and Alfa Cytology runs the KP (KP1 / KP4) program to that standard: conditional-allele recombination verified by genomic PCR, validated flank and intrapulmonary formats, scheduled caliper or micro-CT monitoring, and endpoint pathology with Kras-pathway pharmacodynamics — all under one accountable preclinical operation.

Workflow of KP (KP1 / KP4) Syngeneic Model Construction

Construction follows the published C57BL/6 orthotopic-line methodology: KP1 / KP4 cells are expanded under authenticated conditions, implanted subcutaneously or intrapulmonarily into C57BL/6 mice, grown to a standardized baseline, randomized, treated, and analyzed with efficacy and microenvironment endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: KP1 / KP4 cells are mycoplasma-tested, identity-verified, and confirmed for KrasG12D;Trp53 recombination by genomic PCR — the defining quality step for the platform — then expanded into low-passage working banks.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at approximately 5 × 105 cells (flank, in 50% HBSS/50% Matrigel) or 2.5 × 105 cells in 40 μL (orthotopic) per validated protocol.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane; cells are implanted subcutaneously into the flank, or injected into the left lung through a brief surgical exposure with a fine-gauge needle, per the study arm.
  4. Establishment monitoring and randomization: Flank tumors are measured by caliper from first palpability; orthotopic burden is tracked by scheduled micro-CT where available; animals are randomized once tumors reach the protocol-specified baseline, balancing groups.
  5. Treatment administration: Experimental arms — MEK and CDK4/6 inhibitors alone and combined, Kras-pathway agents, checkpoint inhibitors, and radiotherapy or combination regimens — proceed per study design with body-weight and toxicity monitoring.
  6. Imaging and interim measurements: Caliper measurements or serial micro-CT sessions track tumor kinetics; reporter-arm studies support bioluminescence-based burden quantification on a scheduled cadence.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition and survival with histopathology (H&E confirming lung adenocarcinoma histology, plus Ki67, phospho-ERK/phospho-RB pathway staining, CD8, and PD-L1 immunohistochemistry) and tumor-immune-microenvironment profiling by flow cytometry or spatial methods where designs require it.
