B16-OVA / B16-OVA-luc Orthotopic Mouse Model Service for Melanoma

Fig 1: B16-OVA / B16-OVA-luc Orthotopic Mouse Model for Melanoma preclinical research.

The B16-OVA / B16-OVA-luc orthotopic mouse model of melanoma overlays a defined model antigen on the aggressive, poorly immunogenic B16 background, creating a pigmented syngeneic tumor in which antigen-specific CD8+ T-cell responses — priming, homing, effector control, and escape — can be quantified longitudinally by luciferase bioluminescence in fully immunocompetent mice. Alfa Cytology delivers this platform as an integrated preclinical service: verified OVA line generation or qualification, luciferase tagging, intradermal, flank, or lung-seeding engraftment in C57BL/6 recipients, OT-I/OT-II companion-cell workflows, checkpoint and adoptive-therapy dosing, and endpoints spanning tumor metrics, antigen-presentation status, immune profiling, and histopathology.

Overview of B16-OVA / B16-OVA-luc Orthotopic Mouse Model for Melanoma

B16-OVA is a B16 melanoma lineage engineered to express chicken ovalbumin, and several documented construction routes lead to the same immunological destination: the earliest B16-Ova line (clone Mo5.20.10) was made by transfecting B16-F0 with the pAc-neo-OVA plasmid; modern implementations use either the Sleeping Beauty transposon system — co-transfecting B16-F10 with a pT2 vector encoding OVA, eGFP, and neomycin resistance plus the SB11 transposase, followed by G418 selection at 400 µg/mL, limiting-dilution monoclonalization, and 25-D1.16-antibody confirmation of surface SIINFEKL-H-2Kb — or lentiviral pLVX-EF1a-IRES-PURO delivery with puromycin selection. What the engineering buys is precise: every tumor cell processes and presents the OVA 257-264 peptide (SIINFEKL) on H-2Kb, converting a notoriously cold tumor into an antigen-positive one while leaving its full melanocyte identity — gp100, TRP-1, TRP-2, tyrosinase, melanin — intact. The immunological consequences are dramatic and well documented: OVA-expressing B16 tumors grow measurably slower than parental B16 in immune-competent hosts because of antigen-driven immune rejection pressure, and they respond far more strongly to checkpoint blockade, with anti-PD-1 producing clear antitumor activity in B16-OVA where parental B16 largely resists.

Schematic of the melanoma mouse model comparing wild-type B16-F10 versus soluble OVA-expressing B16F10 versus membrane-bound OVA-expressing B16F10 under immune checkpoint blockade (anti-PD1 or anti-PDL1 + anti-CTLA4), where membrane-localized OVA renders B16F10 tumors immunotherapy-responsive with extended survival, paralleling the clinical TMB + %mAg survival predictor. As an in vivo system, B16-OVA pairs the classic B16 engraftment routes with a uniquely quantitative T-cell toolkit. Subcutaneous and intradermal implantation support standard efficacy cohorts; tail-vein delivery of 5×105 cells seeds the lungs, with metastatic burden verifiable by luciferase signal by day 8 — a design that has supported studies showing intravenous OT-I effectors efficiently eliminate lung-resident OVA-positive melanoma. OT-I CD8+ T cells (and OT-II or SMARTA CD4+ counterparts on CD45.1 congenic backgrounds) can be tracked into tumors, expanded ex vivo, or gene-modified — luciferase-tagged OT-I cells have been shown to home to B16-OVA tumors in vivo, and piggyBac-engineered IL-12-expressing OT-I splenocytes delayed tumor growth and improved survival. The platform has likewise served exosome-based OVA vaccines with TLR-3 stimulation, oncolytic viruses expressing tumor-associated antigen, STING-agonist nanoparticle cross-presentation assays with irradiated B16-OVA, and mechanistic studies of antigen escape under T-cell pressure, where serial co-culture selects surviving OVA-negative variants. Luciferase-tagged B16-OVA derivatives, including an mKate2-SIINFEKL fusion construct (B16-KOVA), extend all of these designs into whole-animal imaging.

Our Services

Because B16-OVA exists in multiple laboratory-built flavors, its quality lives in verification rather than in a catalog number — and that is exactly how Alfa Cytology runs it. Each lot is released only after confirming uniform SIINFEKL-H-2Kb presentation by 25-D1.16 flow cytometry, intact melanocyte antigen expression, and — for luciferase derivatives — uniform in vitro glow. Engraftment routes are matched to the question at hand, from intradermal primary tumors to day-8 lung-metastasis designs, and OT-I/OT-II companion workflows are staged under synchronized schedules. Checkpoint arms carry built-in parental-B16 comparisons, and antigen-status analysis at endpoint distinguishes true immune control from antigen escape. Through the full B16-OVA / B16-OVA-luc orthotopic workflow, Alfa Cytology gives sponsors one accountable partner for this demanding antigen model.

Workflow of B16-OVA / B16-OVA-luc Orthotopic Mouse Model Construction

The B16-OVA-luc orthotopic workflow combines verified antigen engineering with route-matched engraftment and immunology-aware endpoints. The sequence below reflects the standard construction and execution flow.

  1. Antigen and reporter verification: B16-OVA / B16-OVA-luc lots are expanded under appropriate selection (G418 or puromycin), mycoplasma-tested, and released only after 25-D1.16 antibody confirms uniform SIINFEKL-H-2Kb presentation, melanocyte antigens and pigmentation are intact, and luciferase glow is homogeneous in vitro.
  2. Inoculum preparation: Log-phase cultures are harvested, washed, counted, and resuspended in sterile PBS at the study dose (for example, 5×105 cells for lung-seeding designs; 2×106 to 3×106 for flank or intradermal cohorts).
  3. Recipient and donor staging: Syngeneic C57BL/6 (or CD45.1 congenic) mice are acclimatized and stratified; OT-I, OT-II, or SMARTA donor mice are staged in parallel for adoptive-cell arms.
  4. Route-specific engraftment: Cells are delivered intradermally, subcutaneously, or via tail vein (5×105 cells; lung signal confirmed by day 8 in metastatic designs), with baseline bioluminescence after D-luciferin documenting engraftment.
  5. Randomization and treatment allocation: Cohorts are randomized at defined tumor size or signal into arms — anti-PD-1 or other checkpoint antibodies, OVA vaccines or exosomes, oncolytic viruses, adoptive OT-I (naive, activated, or gene-modified) — with parental B16 comparison arms where antigen dependence is in question.
  6. Longitudinal monitoring: Caliper and bioluminescence readouts track primary tumors and metastatic burden on synchronized schedules, body weight and clinical signs are scored throughout, and donor-cell persistence is tracked in congenic designs by flow cytometry.
  7. Endpoint and antigen-status analysis: At termination, tumors and lungs are excised, weighed, and imaged ex vivo; antigen presentation status (25-D1.16 staining) and OVA expression are assessed on surviving tumor; tumor-infiltrating OT-I or endogenous CD8+ cells are quantified by SIINFEKL-tetramer flow cytometry; and histopathology with melanin confirmation closes the study.

Case Study-B16-OVA / B16-OVA-luc Orthotopic Mouse Model Development

Alfa Cytology has implemented the B16-OVA orthotopic platform in preclinical programs spanning checkpoint-sensitization studies, CAR-T and TCR-T proof-of-concept against OVA, vaccine and exosome adjuvant platforms, and oncolytic viroimmunotherapy. In these studies, caliper and bioluminescence readouts tracked primary and lung-metastatic disease on synchronized schedules, parental-B16 arms controlled for antigen dependence, and OT-I adoptive-cell designs — naive, ex vivo activated, or gene-modified — were executed with donor tracking in congenic recipients. Endpoint analyses combined tumor metrics with SIINFEKL-tetramer quantification of tumor-infiltrating T cells and 25-D1.16 antigen-status assessment to distinguish immune control from antigen escape. Program-specific designs and results are retained as sponsor-confidential deliverables, and representative outlines can be furnished upon request.
