Fig 1: MDA-MB-231 Metastatic Tumor Model for Breast Cancer preclinical research.

The MDA-MB-231 Metastatic Tumor Model Service for Breast Cancer delivers the world’s most widely used human triple-negative breast cancer xenograft platform, engineered for both primary-tumor efficacy work and multi-organ metastasis research. Alfa Cytology provides this model as an integrated preclinical service — authenticated cell expansion, immunodeficient host preparation, subcutaneous or orthotopic implantation, experimental and spontaneous metastasis induction, and full histological endpoint analysis — so breast cancer programs can move from compound to conclusion with confidence.

Overview of MDA-MB-231 Metastatic Tumor Model for Breast Cancer

MDA-MB-231 is a human breast adenocarcinoma cell line established in the 1970s by the group of R. Cailleau at M.D. Anderson Hospital from the pleural effusion of a 51-year-old patient with advanced metastatic disease. The line became the archetypal model of triple-negative breast cancer (TNBC): it lacks estrogen receptor, progesterone receptor, and HER2 amplification, displays a mesenchymal, highly invasive phenotype, and harbors the characteristic genomic lesions of the disease — TP53 mutation (R280K), KRAS G13D, BRAF alteration, homozygous CDKN2A/B deletion, and a near-triploid aneuploid karyotype with a modal chromosome number of 64. These molecular signatures, together with a CD44⁺/CD24⁻ cancer stem cell compartment and intrinsic multidrug resistance, make MDA-MB-231 the standard in vitro and in vivo benchmark for studying the biology and therapy of aggressive, hormone-refractory breast cancer.

Establishment and validation of caudal artery injection for the MDA-MB-231 bone metastasis model: anatomy of the caudal vessels, real-time fluorescence imaging confirming rapid arterial dissemination versus tail-vein pulmonary trapping, and BLI/ex vivo quantification showing selective hindlimb bone localization after MDA-MB-231 injection. (Kuchimaru, Takahiro, et al., 2018)

Because it is of human origin, MDA-MB-231 grows only in immunodeficient hosts. In athymic nude mice the cells form poorly differentiated grade III adenocarcinomas and micrometastasize to lung and lymph nodes, while in NOD/SCID and particularly NSG mice — where residual NK activity is absent — both primary growth and spontaneous metastasis to lung, liver, bone, brain, and lymph nodes are markedly enhanced, closely reproducing the organotropism of human disease. The model supports every major inoculation format used in metastasis research: subcutaneous or orthotopic mammary fat pad implantation (about 1 × 106 cells, commonly with Matrigel) for spontaneous dissemination, tail vein injection for experimental lung colonization, and intracardiac injection for bone and brain tropism. A rich ecosystem of derivatives — luciferase/GFP reporter lines and organotropic sublines selected in vivo for lung (LM2-4, 4175), bone (BoM-1833), brain (BrM2), and leptomeningeal (LeptoM) homing — further allows mechanistic dissection of each step of the metastatic cascade under bioluminescence imaging in vivo.

Our Services

Alfa Cytology maintains authenticated, mycoplasma-free MDA-MB-231 stocks together with luciferase-tagged and organotropic subline resources, and operates established nude, NOD/SCID, and NSG xenograft colonies for this model. Study configurations range from rapid subcutaneous efficacy screens to orthotopic and experimental metastasis programs with serial bioluminescence imaging and multi-organ endpoint analysis — all executed under strict randomization and quality control.

Workflow of MDA-MB-231 Metastatic Tumor Model Construction

Constructing an MDA-MB-231 metastatic study involves coordinated cell, animal, and imaging workstreams. The sequence below describes how the model is built and evaluated:

  1. Cell Line Preparation: MDA-MB-231 cells are authenticated by STR profiling, confirmed mycoplasma-free, and expanded at controlled passage; reporter and organotropic sublines are verified before implantation.
  2. Host Selection and Acclimatization: Female immunodeficient mice (athymic nude, NOD/SCID, or NSG, depending on the metastatic endpoint) are acclimatized under controlled conditions and randomly allocated to groups.
  3. Tumor Cell Implantation: Cells are delivered by the planned route: about 2 × 106 cells with Matrigel subcutaneously, 1 × 106 cells into the fourth mammary fat pad orthotopically, 1 × 106 cells via tail vein, or 2 × 105 cells intracardiacally.
  4. Primary Tumor Establishment: Implant sites are monitored until measurable tumors form; caliper-based volume tracking and body-weight recording proceed on a fixed schedule.
  5. Randomization and Dosing: Animals are re-randomized once tumors reach the target size (e.g., approximately 100 mm3), and treatment with vehicle, reference agents, or test articles begins on the designated regimen.
  6. Longitudinal Metastasis Monitoring: Luciferase-labeled cohorts undergo serial bioluminescence imaging in vivo to quantify primary and metastatic burden non-invasively; health status is tracked throughout.
  7. Terminal Multi-Organ Analysis: At necropsy, primary tumors are excised and weighed; lungs, bones, brain, liver, and lymph nodes are examined macroscopically, processed for hematoxylin and eosin histology, and evaluated for human tumor cells by species-specific markers.
  8. Data Analysis and Reporting: Tumor-growth curves, imaging signals, metastatic incidence and burden, and survival data are statistically analyzed and consolidated into a final study report.

Case Study-MDA-MB-231 Metastatic Tumor Model Development

In a recent preclinical collaboration, Alfa Cytology implemented the MDA-MB-231 metastatic model for a sponsor pursuing a therapeutic candidate in triple-negative breast cancer. The scope included expansion and implantation of luciferase-labeled MDA-MB-231 cells in immunodeficient hosts, allocation of vehicle and treatment arms, scheduled tumor and body-weight monitoring with serial bioluminescence imaging, and terminal examination of primary tumors and distant organs with histological confirmation of metastases — producing a coherent evidence package that supported the client’s preclinical development decisions. Program formats can be arranged around primary-tumor efficacy, spontaneous or experimental metastasis quantification, or combination-therapy objectives.
