MGC-803 / MGC-803-luc Orthotopic Mouse Model Service for Gastric Cancer

Fig 1: MGC-803 / MGC-803-luc Orthotopic Mouse Model for Gastric Cancer preclinical research.

The MGC-803 / MGC-803-luc orthotopic mouse model for gastric cancer enables human tumor cells to colonize the stomach wall of immunodeficient mice, where they establish invasive primary tumors that can be followed over time without invasive procedures. Specializing exclusively in preclinical oncology research, Alfa Cytology supports this model end to end: luciferase-labeled cell preparation, microsurgical implantation into the gastric wall, scheduled in vivo bioluminescence imaging, and full endpoint histopathology.

Overview of MGC-803 / MGC-803-luc Orthotopic Mouse Model for Gastric Cancer

MGC-803 is a human gastric cancer cell line originally reported in 1980 after isolation from the poorly differentiated mucinous adenocarcinoma of a 53-year-old male patient. The line grows as an adherent, epithelial-like monolayer with a population doubling time of roughly 22 hours, carries a chromosome count in the 58–61 range, and displays vigorous tumorigenicity when introduced into immunocompromised animals. Researchers should be aware that STR-based authentication analyses have since flagged MGC-803 as a suspected hybrid lineage sharing genetic material with HeLa, a caveat that informed study design should acknowledge; in practice, the line remains one of the most frequently employed tools in gastric cancer research, particularly within the Asian research community, and reproducible behavior has been documented across a wide range of in vitro and in vivo applications.

Bioluminescence imaging of an MGC803-Luc orthotopic gastric cancer model established by implantation of luciferase-positive tumor fragments onto the gastric serosa, enabling evaluation of primary tumor burden and metastatic dissemination. (Guan, Xiaoqing, et al., 2024)

In orthotopic xenograft work, MGC-803 cells labeled with firefly luciferase (MGC-803-luc) are inoculated beneath the serosa of the mouse stomach, most often in immunodeficient strains such as BALB/c nude or NOD-SCID mice. The resulting primary tumor expands locally and progressively breaches the muscular layers of the gastric wall, a process that luciferase tagging renders visible through repeated whole-animal imaging after D-luciferin administration. Because the bioluminescent signal scales with viable tumor mass, the MGC-803-luc orthotopic model supports quantitative tracking of tumor growth kinetics, treatment-induced regression or regrowth, and the onset of abdominal spread, making it a practical platform for evaluating antitumor and anti-invasive candidate agents. Figure 1 provides a visual summary of how this orthotopic system is assembled and monitored.

Cell Line Information: MGC-803 / MGC-803-luc

Karyotype Hyperdiploid; chromosome number reported in the 58–61 range
Authentication Note STR-profiled and registered with Cellosaurus; listed in the ICLAC Register of Misidentified Cell Lines as a suspected HeLa hybrid lineage — researchers are advised to run in-house STR verification
Molecular Features c-Myc pathway activation and hTERT expression reported; responsiveness to ROS-inducing agents and modulation of epithelial-mesenchymal transition (EMT) documented in the literature
MGC-803-luc Derivative Stably expresses firefly luciferase, permitting non-invasive in vivo bioluminescence imaging with signal intensity proportional to viable tumor burden
Orthotopic Behavior Subserosal implantation in immunodeficient mice yields locally invasive primary gastric tumors suitable for growth and invasion monitoring
Research Applications Orthotopic tumor growth and invasion studies, antitumor and anti-invasive drug efficacy screening, real-time therapy response monitoring, gastric cancer stem cell and tumor microenvironment research

Workflow of MGC-803 / MGC-803-luc Orthotopic Mouse Model Construction

Establishing an MGC-803 or MGC-803-luc orthotopic gastric tumor is a staged process that moves from authenticated cell material through subserosal inoculation and serial imaging to structured endpoint analysis. The stages below describe the standard execution path used for this platform.

  1. Cell Bank Verification and Expansion: MGC-803 or MGC-803-luc stocks are authenticated in-house by STR profiling and confirmed mycoplasma-free, then expanded in vitro under the appropriate serum-supplemented medium.
  2. Luciferase Performance Check (MGC-803-luc Only): For luciferase-tagged stocks, signal output per cell is calibrated so that downstream in vivo photon flux can be converted into relative tumor burden.
  3. Mouse Conditioning and Randomization: Immunodeficient recipients, typically BALB/c nude or NOD-SCID mice, are acclimatized and distributed into groups by body weight, with anesthesia and analgesia administered per approved protocols.
  4. Gastric Wall Exposure: Through a small midline laparotomy performed under aseptic conditions, the stomach is gently externalized and the serosal surface is prepared for cell delivery.
  5. Subserosal Cell Inoculation: A single-cell suspension of MGC-803 or MGC-803-luc cells (commonly around 106 cells in a low-volume carrier) is deposited slowly into the subserosal plane of the gastric wall, where the cells anchor and proliferate.
  6. Closure and Postoperative Recovery: The abdominal layers are sutured and animals are returned to warmed cages, with daily health and body weight checks until complete recovery.
  7. Serial Bioluminescence Monitoring: Following intraperitoneal D-luciferin injection, whole-animal imaging sessions are scheduled at defined intervals to chart primary tumor expansion and detect abdominal dissemination in real time.
  8. Endpoint Necropsy and Histology: At the scheduled sacrifice point, the primary gastric tumor is resected and weighed, the stomach is processed for H&E and immunohistochemical assessment of invasion depth, and abdominal organs are surveyed for metastatic foci.
