Fig 1: Mia-PaCa 2 / Mia-PaCa 2-luc Orthotopic Mouse Model for Pancreas Cancer preclinical research.

The Mia-PaCa 2 / Mia-PaCa 2-luc orthotopic mouse model places one of the most aggressive human pancreatic ductal adenocarcinoma cell lines into its organ of origin, where tumors invade locally, spread along nerves, and seed distant metastases much as they do in patients. Alfa Cytology is a preclinical oncology CRO with demonstrated proficiency in orthotopic pancreas cancer modeling. Alfa Cytology offers complete Mia-PaCa 2 / Mia-PaCa 2-luc orthotopic model services, spanning authenticated cell expansion, luciferase reporter verification, surgical or ultrasound-guided implantation, longitudinal in vivo imaging, and thorough endpoint characterization.

Overview of Mia-PaCa 2 / Mia-PaCa 2-luc Orthotopic Mouse Model for Pancreas Cancer

MIA PaCa-2 is one of the most widely used human pancreatic ductal adenocarcinoma (PDAC) cell lines. It was established in 1975 by A. Yunis and colleagues from the tumor of a 65-year-old Caucasian man and grows as an adherent, epithelial-like monolayer with a doubling time of roughly 26 hours. The line is genetically distinctive: it carries a homozygous activating KRASG12C mutation — a codon 12 substitution found in only a minority of pancreatic tumors — together with the gain-of-function TP53R248W mutation and a homozygous deletion of CDKN2A/p16INK4A, while SMAD4 remains wild-type. This mutational profile, combined with rapid proliferation, strong soft-agar clonogenicity, and elevated PI3K/AKT pathway activity, has made MIA PaCa-2 a workhorse for pancreatic cancer biology and drug screening, and a model of particular interest for testing KRAS G12C-directed inhibitors.

An orthotopic model was established in BALB/c nude mice by injecting MIA PaCa-2 cells (resuspended in Matrigel) into the pancreatic parenchyma; probe specificity was subsequently verified using IVIS after confirming pancreatic tumor growth. (Ren, Han, et al., 2022)

Orthotopically implanted into the pancreas of immunodeficient mice, MIA PaCa-2 cells engraft with essentially 100% efficiency and generate poorly differentiated, highly infiltrative tumors that reproduce hallmark features of human PDAC, including perineural invasion that reaches 100% within eight weeks and dissemination to regional and distant sites. Luciferase-labeled Mia-PaCa 2-luc cells permit whole-body, non-invasive quantification of tumor burden by in vivo bioluminescence imaging, an essential capability for deeply seated pancreatic tumors that calipers cannot measure. Taken together, the Mia-PaCa 2 / Mia-PaCa 2-luc orthotopic model offers a faithful and technically tractable platform for preclinical evaluation of cytotoxic agents, targeted therapies, stromal modulators, and delivery technologies in pancreatic cancer research.

Workflow of Mia-PaCa 2 / Mia-PaCa 2-luc Orthotopic Mouse Model Construction

Establishing a Mia-PaCa 2 / Mia-PaCa 2-luc orthotopic model integrates validated cell preparation, precise implantation technique, and quantitative longitudinal imaging. The standard process unfolds as follows.

  1. Cell line authentication and expansion: MIA PaCa-2 or Mia-PaCa 2-luc cells are expanded in DMEM-based medium, confirmed by STR profiling and mycoplasma testing, and checked for viability; luciferase expression in Mia-PaCa 2-luc is validated by an in vitro bioluminescence assay.
  2. Host preparation: Immunodeficient mice (e.g., athymic nude, NOD-SCID, or NSG), typically 4–8 weeks of age, are acclimatized and randomized in compliance with an approved animal ethics protocol.
  3. Inoculum preparation: Log-phase cells are harvested into a sterile single-cell suspension in PBS or saline, commonly 2 × 105 cells in 40 μL per mouse, with viability confirmed immediately before implantation.
  4. Orthotopic implantation: The pancreatic parenchyma is accessed by laparotomy, or by minimally invasive ultrasound-guided injection, and the cell suspension is delivered directly into the pancreas; the entry site is checked for reflux to prevent peritoneal seeding artifacts.
  5. Post-operative care and recovery: Analgesia is provided and animals are monitored daily until fully recovered, with body weight and clinical condition recorded.
  6. Bioluminescence imaging and randomization (Mia-PaCa 2-luc): Following an initial engraftment window, mice receive intraperitoneal D-luciferin and are imaged weekly by in vivo bioluminescence imaging; animals are randomized to treatment arms once signals reach predefined thresholds.
  7. Treatment administration (optional): Test compounds, targeted agents (including KRAS G12C inhibitors), biologics, or combination regimens are dosed according to the customized study design.
  8. Endpoint analysis: At study termination, necropsy is performed to weigh the primary pancreatic tumor, score metastatic nodules and perineural invasion, collect ascites where present, and process tissues for H&E and immunohistochemistry, followed by statistical analysis and a consolidated final report.
