JC Syngeneic Model Service for Breast Cancer

Fig 1: JC Syngeneic Model for Breast Cancer preclinical research.

The JC syngeneic model is the deliberately weakly immunogenic breast cancer platform, engrafting a spontaneously arisen murine mammary adenocarcinoma — rigorously verified as virus-particle-free at establishment — into syngeneic BALB/c hosts where concomitant immunity, not rejection, defines the immune readout. Alfa Cytology delivers this distinctive model as a complete preclinical service — authenticated cell banking, subcutaneous, intraperitoneal, or intravenous implantation at modest inocula, immunization–challenge study formats, and full histopathology — giving sponsors a window into immunologically silent breast tumors that highly immunogenic models cannot provide.

Overview of JC Syngeneic Model for Breast Cancer

JC was established by T. Y. Chao and colleagues in 1989 from something every breast cancer researcher hopes for and rarely gets: a primary adenocarcinoma that developed spontaneously in the mammary gland of an aged virgin female BALB/c mouse — no carcinogen, no transgene, no engineered oncogene, simply the disease arriving on its own in an immunologically normal host. The characterization paper that introduced the line is a model of rigor rarely seen in modern cell-line reports: isoenzyme analyses of glucose-6-phosphate dehydrogenase, lactate dehydrogenase, and peptidase proved the cells genuinely murine and free of cross-species contamination; karyotyping revealed a broad chromosome range of 26 to 100 with a modal number of 40; and electron microscopy detected tonofilaments and desmosomes confirming epithelial identity, while the negative finding of no type B or C virus particles — with only occasional intracisternal A particles — distinguished JC from the virus-associated mammary models of its era. Tumorigenicity was, in the authors' words, easily established in immunocompetent syngeneic hosts by subcutaneous, intraperitoneal, and intravenous routes alike.

It is the immunology, however, that gives JC its lasting identity. In the classic immunization–challenge assay, vaccination with JC tumor cells never produced rejection — but it significantly prolonged the incubation period before palpable tumors appeared, and concomitant immunity was demonstrably present in challenge experiments. That phenotype — a very weakly immunogenic, highly tumorigenic mammary carcinoma that the immune system can delay but not destroy — models the clinically common "immunologically silent" breast tumor far better than hot, rejection-prone lines, and it made JC a valued substrate for experimental immunotherapy from its first years: cytokine-augmented approaches, adoptive-cell strategies, photodynamic therapy, and hyperthermia combinations have all been benchmarked on this line. The practical profile is attractive for modern programs: small inocula on the order of 2 × 105 cells establish flank tumors, all three classical routes are validated, and the documented absence of B/C-type viral particles removes a virological confound that complicates interpretation in some mammary models. For sponsors studying immune tolerance, therapy-resistant immunophenotypes, or modalities that must work in cold tumors, JC offers something its famous siblings cannot: a genuinely silent tumor that grows anyway.

Cell Line Information: JC Syngeneic Model

JC is the murine mammary adenocarcinoma line established by Chao and colleagues in 1989 from a spontaneous primary tumor of an aged virgin female BALB/c mouse, characterized by isoenzyme, karyotypic, and ultrastructural analysis. The line remains available through the mammary tumor research community. Core characteristics are compiled below.

Parameter Description
Cell line JC (murine mammary adenocarcinoma line JC)
Establishment Established by T. Y. Chao and colleagues (1989) from a spontaneously developed primary adenocarcinoma of an aged virgin female BALB/c mouse; maintained in continuous culture for experimental immunotherapy for more than five years at establishment
Species / strain Mus musculus, BALB/c (syngeneic with BALB/c hosts)
Tissue of origin Mammary gland; primary spontaneous adenocarcinoma
Disease Mammary adenocarcinoma
Origin verification Isoenzyme analyses (glucose-6-phosphate dehydrogenase, lactate dehydrogenase, peptidase) confirmed murine origin with no contamination from other species
Karyotype Chromosome number ranges 26–100 with a modal number of 40
Ultrastructure Electron microscopy detected tonofilaments and desmosomes, confirming epithelial nature; no type B or C virus particles detected, with occasional intracisternal A particles observed
Tumorigenicity Easily established in immunocompetent syngeneic hosts by subcutaneous, intraperitoneal, and intravenous injection of viable JC tumor cells; small inocula (on the order of 2 × 105 cells) suffice for flank tumors
Immunogenicity Very weak immunogenicity demonstrated by immunization–challenge: no rejection observed, but a significant prolongation of the pre-palpable incubation period versus controls; concomitant immunity detected in challenge assays
Complete growth medium RPMI-1640 or DMEM with 10% fetal bovine serum (standard formulations in the literature)
Culture conditions 37 °C, humidified atmosphere of 95% air and 5% CO2
Biosafety level BSL-1
Historic immunotherapy role Used for experimental immunotherapy since establishment — a classic substrate for cytokine, adoptive-cell, photodynamic, and hyperthermia studies in weakly immunogenic tumors
Distinctive niche Absence of B/C-type virus particles distinguishes JC from virus-associated mammary models, simplifying virological confound assessment; the very weak immunogenicity phenotype models the clinically common "immunologically silent" breast tumor
Primary applications Breast cancer immunotherapy in poorly immunogenic settings, tumor-immune tolerance mechanisms, cytokine and adoptive-cell therapy evaluation, photodynamic and hyperthermia combination studies, general mammary carcinoma pharmacology

Our Services

Engaging Alfa Cytology for the JC syngeneic model places a rigorously characterized, virus-particle-free mammary platform under one accountable team: authenticated, low-passage JC banks are maintained with mycoplasma-free certification, syngeneic BALB/c hosts are sourced and acclimated, tumors are established by the subcutaneous, intraperitoneal, or intravenous route at the modest inocula the line tolerates, immunization–challenge and concomitant-immunity formats are executed where immune-mechanism questions require them, and treatment arms from cytokine and adoptive-cell strategies to photothermal and hyperthermia combinations run through scheduled monitoring to terminal mammary and systemic histopathology, all under IACUC-compliant conditions and returned as an audit-ready data package.

Workflow of JC Syngeneic Model Construction

JC studies are defined by route selection and, where immune questions are primary, by the classical immunization–challenge geometry. Both rest on the line's easy tumorigenicity and documented weak immunogenicity. A standard program proceeds as follows.

  1. Cell preparation and quality control: JC cells are expanded at low passage, confirmed mycoplasma-free, and viability verified; suspensions are prepared fresh at modest inocula (on the order of 2 × 105 cells per subcutaneous site).
  2. Host selection and acclimation: Female BALB/c mice, typically 6–10 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility.
  3. Implantation: subcutaneous route. Cells in 0.1 mL sterile PBS are injected into the flank; the site is compressed briefly on needle withdrawal, and palpable tumors establish readily at the documented small inoculum.
  4. Implantation: intraperitoneal or intravenous routes. For systemic or dissemination formats, defined doses are administered i.p. or via the lateral tail vein, exploiting the line's documented easy establishment by both routes.
  5. Immunization–challenge formats where specified: For immune-mechanism studies, cohorts are primed with irradiated or viable JC cells and challenged after a defined interval; latency-to-palpable-tumor and concomitant-immunity readouts follow the 1989 convention.
  6. Randomization and treatment. Tumor-bearing mice are randomized by volume into arms, and test articles: cytokines, adoptive-cell products, photodynamic or photothermal agents, hyperthermia combinations — are administered per protocol with body weight recorded.
  7. Longitudinal monitoring: Tumors are measured by caliper on the protocol cadence; clinical signs are scored daily, and humane endpoints are predefined consistent with the line's vigorous growth.
  8. Endpoint analysis and reporting: At termination, primary tumors and systemic organs are resected and weighed; tissues undergo H&E with mammary histology, immunohistochemistry (CD8, CD4, FoxP3, F4/80, CD31, PD-L1 panels), and where immunization arms are included, delayed-type hypersensitivity and serum-antibody assessment, with all data consolidated into the final report.

Case Study-JC Syngeneic Model

Alfa Cytology has applied the JC syngeneic platform to preclinical programs spanning immunotherapy in weakly immunogenic settings, cytokine and adoptive-cell evaluation, photothermal and hyperthermia combination studies, and immune-tolerance mechanism research in breast cancer. In these engagements, caliper readouts tracked primary tumor kinetics with latency-sensitive designs where immunization arms were included, while terminal mammary histopathology with immune-marker panels completed the pharmacological and immunological assessment. Study parameters — implantation route, inoculum, challenge formats, and endpoint panels — are tailored to each program. Case materials can be shared under confidentiality agreements upon request.
