MOPC-315 / MOPC-315-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: MOPC-315 / MOPC-315-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The MOPC-315 / MOPC-315-luc orthotopic mouse model for multiple myeloma engrafts a mineral oil-induced BALB/c plasmacytoma into the bone marrow compartment of immunocompetent hosts, combining decades of idiotype-immunology pedigree with modern bioluminescence tracking of marrow disease and bone destruction. Dedicated exclusively to preclinical oncology, Alfa Cytology delivers this syngeneic platform end to end: authenticated MOPC-315 / MOPC-315-luc cell supply, bone marrow-homing tail-vein inoculation into BALB/c mice, scheduled imaging paired with idiotype IgA ELISA, and terminal microCT-supported bone analysis.

Overview of MOPC-315 / MOPC-315-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

MOPC-315 stands on the deepest historical foundation of any murine myeloma model. In the 1960s, Michael Potter at the US National Cancer Institute discovered that repeated intraperitoneal injections of mineral oil—or its defined component pristane—induce plasmacytomas in BALB/c mice, a strain rendered susceptible by hypomorphic alleles of p16 and Frap, and the MOPC (Mineral Oil-induced Plasmacytoma of Potter) panel he single-handedly created transformed antibody research. MOPC-315 is one of that panel’s most famous members: a B-cell plasmacytoma that secretes an IgAλ2 myeloma protein remarkable for binding dinitrophenyl and trinitrophenyl haptens, making it an early workhorse of antibody structure and idiotype research. Like most peritoneal plasmacytomas, it carries a Myc-activating Igh-Myc translocation, and it is strikingly glucocorticoid-sensitive—killed by as little as 10 nM dexamethasone. The classical line grows in suspension and requires disciplined density control, since cultures allowed above roughly 106 cells per milliliter die off rapidly.

Subcutaneous MOPC-315 plasmacytoma model comparing empty-vector (MOPCpEF) versus VEGF-overexpressing (MOPCSVm) cells in immune-competent BALB/c versus BALB/c nude mice, showing that MOPCSVm tumors grew faster in BALB/c but were rejected in nude mice via irradiation/adoptive splenocyte transfer experiments, with tumor volume curves tracking growth over 25 days. For faithful multiple myeloma modeling, the bone marrow-homing derivative MOPC-315.BM is the orthotopic instrument of choice: generated by Hofgaard, Bogen, and colleagues through nine generations of serial intravenous autografting of marrow-derived plasma cells, it acquired exquisite bone marrow tropism, genuine osteolytic capacity, and a median survival of about one month in BALB/c recipients. Luciferase-tagged MOPC-315.BM delivered by tail-vein injection at 1 × 105 cells homes to femur, tibia, and spleen, with the first bioluminescent signal detectable by day 9—earlier than the serum M315 idiotype rises above background—and progressive multifocal skeletal growth that histology confirms as tumor in marrow with osteolytic destruction of cortical bone. The line expresses the marrow-homing receptor triad CXCR4, CD44, and α4β1 (VLA-4), the latter two being direct targets of approved myeloma drugs, and it has served melphalan response validation, IL-34 and Notch bone-loss mechanism studies, eosinophil and megakaryocyte niche research, idiotype vaccination, and oncolytic virus programs—all in an immunocompetent host capable of modeling immune-tumor interactions that immunodeficient systems cannot.

Cell Line Information: MOPC-315 / MOPC-315-luc

MOPC-315’s half-century of use has left it documented from hapten-binding biochemistry to whole-animal imaging. The defining attributes are summarized below.

Cell Line Name MOPC-315; bone marrow-homing subline MOPC-315.BM (derived from MOPC-315.4); luciferase-labeled derivative: MOPC-315.BM-luc+ (firefly luciferase, used in the Bogen-laboratory imaging protocols)
Catalog / Accession ECACC 85022106 (parent MOPC-315, deposited via Oxford); Cellosaurus CVCL_VI68 (MOPC-315.BM; parent MOPC-315.4, CVCL_VI67); not an ATCC deposit
Species / Tissue / Disease Mouse (Mus musculus) / plasmacytoma / plasma cell malignancy (extramedullary by origin; rendered bone marrow-homing in the .BM subline)
Derivation Mineral oil-induced plasmacytoma of the MOPC panel generated by Michael Potter (NCI) in BALB/c mice in the 1960s; MOPC-315.BM established by Hofgaard et al. (2012) through nine generations of serial intravenous autografting of bone marrow-derived plasma cells
Secreted Product IgAλ2 myeloma protein binding dinitrophenyl/trinitrophenyl haptens; idiotype (M315) detectable in serum by ELISA as a tumor burden biomarker
Genetic Features Myc-activating Igh-Myc translocation [t(12;15)] typical of peritoneal plasmacytomas; lambda light chain expressed (kappa not expressed)
Drug Sensitivity Killed by 10 nM dexamethasone (ECACC documentation); classical readout for glucocorticoid sensitivity in preclinical screens
Marrow-Homing Receptors CXCR4, CD44, and α4β1 integrin (VLA-4) highly expressed; dynamically regulated in vivo—relevant because both CD44 and VLA-4 are targeted by approved drugs
Morphology / Growth Round suspension cells; maintain at 1–9 × 105 cells/mL and never exceed ~106 cells/mL, at which rapid death occurs; biosafety level 1 (hazard group 2 per ECACC conventions)
Culture Conditions EMEM (EBSS) + 2 mM glutamine + 1% non-essential amino acids + 10–20% fetal bovine serum; 37 °C, 5% CO₂; twice-weekly handling with density discipline
Orthotopic Protocol (Published) Tail-vein injection of 1 × 105 MOPC-315.BM-luc+ cells into BALB/c wild-type mice; first BLI signal by day +9; multifocal growth in skeleton and spleen; osteolytic femoral lesions; paraplegia as terminal endpoint; ~1 month median survival
Imaging and Biomarker Readouts IVIS BLI after D-luciferin (150 mg/kg i.p., 10-minute delay); ex vivo organ imaging confirms localization; serum M315 idiotype-specific IgA ELISA correlates with BLI over 43 days but is less sensitive early
Therapeutic Validation Melphalan (5 mg/kg i.p., 12 days) slowed tumor progression and reduced dissemination relative to untreated controls; dexamethasone sensitivity documented at nanomolar concentrations in vitro
Research Applications Multiple myeloma drug and immunotherapy evaluation, idiotype vaccination and CD4 T-cell response research, myeloma bone disease mechanism studies (IL-34, Notch signaling), BM niche and accessory-cell (eosinophil, megakaryocyte) research, oncolytic virus and adoptive therapy programs, glucocorticoid sensitivity benchmarking

Our Services

A MOPC-315 / MOPC-315-luc engagement with Alfa Cytology is run as one continuous program: density-disciplined cell banking, tail-vein orthotopic inoculation of the marrow-homing subline into immunocompetent BALB/c recipients, IVIS imaging paired with M315 idiotype ELISA, dosing per the client’s regimen, and a terminal package combining skeletal microCT analysis, ex vivo organ imaging, and bone marrow immunohistochemistry.

Workflow of MOPC-315 / MOPC-315-luc Orthotopic Mouse Model Construction

Building a MOPC-315-luc orthotopic myeloma model turns a classical plasmacytoma into a quantitative bone marrow imaging system: density-controlled culture keeps the cells alive and healthy, tail-vein inoculation seeds the skeleton, and dual imaging-plus-idiotype readouts chart the disease. The workflow below reflects this execution.

  1. Cell Bank Verification and Expansion: MOPC-315 or MOPC-315.BM-luc stocks are authenticated and mycoplasma-tested, then expanded in EMEM-based medium with strict density discipline (1–9 × 105 cells/mL), since overgrowth above ~106 cells/mL triggers rapid death.
  2. Luciferase Calibration (MOPC-315-luc Only): Signal output per cell is verified in vitro, and CD138+CD4+ identity of the marrow-homing line is confirmed by flow cytometry before in vivo use.
  3. Mouse Preparation and Randomization: Immunocompetent BALB/c mice are acclimatized and randomized by body weight under approved protocols; no irradiation or immunodeficiency is required for syngeneic engraftment.
  4. Orthotopic Tail-Vein Inoculation: 1 × 105 MOPC-315.BM-luc+ cells in a small sterile volume are delivered via the tail vein so that plasma cells home to femoral, tibial, and shoulder marrow and to the spleen.
  5. Engraftment Confirmation and Longitudinal Imaging: Whole-animal BLI after D-luciferin (150 mg/kg i.p., 10-minute delay) detects the first skeletal foci by day 9, with scheduled re-imaging tracking the emergence of new foci and growth of established ones; serum M315 idiotype IgA is measured in parallel.
  6. Treatment Administration: Test agents—alkylators such as melphalan, glucocorticoids, niche-targeted antibodies, or immunotherapies—are dosed per the client’s schedule once engraftment is confirmed.
  7. Surveillance and Survival Endpoint: Daily monitoring tracks body weight and neurological status; paraplegia, severe weight loss, or welfare criteria define the survival endpoint, typically around one month after inoculation.
  8. Terminal Skeletal and Organ Analysis: At endpoint, femurs, tibiae, and spine are scanned by microCT or processed for H&E to quantify osteolytic lesions; spleen, liver, and other organs are imaged ex vivo, and marrow is profiled by flow cytometry and immunohistochemistry (lambda light chain) to confirm plasma cell identity and burden.
