MAVER-1 / MAVER-1-luc Orthotopic Mouse Model Service for Lymphoma

Fig 1: MAVER-1 / MAVER-1-luc Orthotopic Mouse Model for Lymphoma preclinical research.

The MAVER-1 / MAVER-1-luc orthotopic mouse model disseminates an aggressive, leukemic-variant mantle cell lymphoma — one of the few MCL lines carrying the defining t(11;14) translocation together with MYC rearrangement, ATM deletion, and biallelic TP53 inactivation — through the hematolymphoid system of immunodeficient mice. Alfa Cytology delivers this therapeutically unforgiving platform with authenticated banking, subcutaneous or systemic luciferase-imaging configurations, benchmark arms spanning BTK, BCL-2, and CDK4/6 pathways, and pairing against derived venetoclax-resistant MAVER-1 cohorts, so your mantle cell lymphoma agents and combination strategies are tested against the resistance biology that actually defeats current therapy.

Overview of MAVER-1 / MAVER-1-luc Orthotopic Mouse Model for Lymphoma

MAVER-1 was established in Italy and published in Haematologica in 2006 as a model of leukemic, aggressive mantle cell lymphoma — a disease whose cell lines are notoriously difficult to generate. The line carries the full genetic signature of high-risk MCL: the t(11;14)(q13;q32) translocation with its breakpoint in the major translocation cluster driving cyclin D1 overexpression, a superimposed MYC rearrangement at 8q24, deletion of ATM, deletion of 8p22, and TP53 inactivated by the combination of an exon 8 mutation and 17p13 deletion. Its immunophenotype is textbook MCL — IgM and lambda light chain positive, CD5+, CD10−, CD19+, CD20+, CD23−, CD79a+, cyclin D1+ — with a rearranged immunoglobulin gene in germline configuration for mutational status. This stacked genotype explains both the line's aggressiveness and its documented drug behavior: MAVER-1 is one of the two canonical primarily ibrutinib-resistant MCL lines, standing alongside Z-138 in contrast to the ibrutinib-sensitive JeKo-1, REC-1, and Mino trio.

That primary-resistance phenotype has made MAVER-1 a workhorse for second-generation MCL pharmacology. The HCK/BTK dual inhibitor KIN-8194 inhibits MAVER-1 growth and suppresses AKT-S6 signaling in an HCK-dependent manner; the CDK4/6 inhibitor abemaciclib shows nanomolar activity (IC50 70–952 nM across MCL lines including MAVER-1); and CTPS1 targeting remains effective even against a derived venetoclax-resistant MAVER-1. On the apoptosis axis, the dual BCL-2/BCL-XL inhibitor AZD4320 proved highly efficacious in MAVER-1 models where venetoclax resistance was modeled by BCL-XL upregulation, and in NSG xenografts MAVER-1 develops acquired venetoclax resistance under 100 mg/kg daily dosing — resistance accompanied by BCL-XL, MCL-1, and BCL-2 upregulation with BIM sequestration, which the BCL-XL-selective inhibitor A1155463 overcomes in combination with venetoclax. Orthotopic and subcutaneous luciferase configurations extend these benchmarks to whole-animal kinetics in vivo, making MAVER-1 one of the few single lines in which BTK insensitivity, BCL-2 family rewiring, and cell-cycle targeting can be interrogated in one study.

Cell Line Information: MAVER-1 / MAVER-1-luc

MAVER-1 (ATCC CRL-3003; RRID CVCL_1870) grows in suspension in vitro in RPMI-1640 with 10% fetal bovine serum and is among the most thoroughly characterized of the roughly nine MCL lines described in the literature. Its stacked high-risk genotype — t(11;14) plus MYC rearrangement, ATM deletion, and biallelic TP53 loss — makes it the preferred substrate when a study needs MCL biology that has already escaped first-line targeted therapy. The MAVER-1-luc derivative adds firefly luciferase for quantitative imaging in vivo, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence.

Our Services

MCL programs succeed when the model already embodies the resistance their drug is designed to overcome. Alfa Cytology runs MAVER-1 studies with batch-verified cyclin D1 expression and TP53 status, monoclonal luciferase banks for clean imaging kinetics in vivo, benchmark arms spanning BTK, BCL-2, and CDK4/6 pathways at clinically relevant doses, and derived venetoclax-resistant cohorts where combination durability is the question — all reported against the published MCL line sensitivity panel your reviewers already know.

Workflow of MAVER-1 / MAVER-1-luc Orthotopic Mouse Model Construction

MAVER-1 model construction follows the compact, standardized protocols of the MCL xenograft literature, with study design organized around the line's primary-resistance phenotype. The workflow below details construction, imaging-guided staging, and apoptosis-pathway endpoints.

  1. Cell Expansion and Quality Control: MAVER-1 or MAVER-1-luc cells are expanded in vitro in RPMI-1640 + 10% FBS to the logarithmic phase; batches are confirmed mycoplasma-negative, STR-authenticated, and verified for cyclin D1 expression, CD5/CD20 immunophenotype, and t(11;14) translocation, with monoclonal luciferase expression confirmed for tagged lines.
  2. Host Selection and Acclimatization: NSG mice, typically 6–8 weeks of age, are acclimatized for approximately one week under SPF conditions and health-screened before assignment to study cohorts.
  3. Route Selection and Inoculation: The configuration is chosen — subcutaneous flank implantation (classically 5–10 × 106 cells in ~100 µL PBS, with Matrigel where required) for localized benchmarking, or intravenous/intraperitoneal delivery for disseminated disease designs — with implantation day recorded for the dosing calendar.
  4. Engraftment Verification and Randomization: Systemic disease is confirmed by whole-body bioluminescence imaging in vivo after D-luciferin administration; flank tumors are staged by caliper to a predefined volume, and mice are randomized into vehicle, benchmark, and investigational arms with matched starting burdens.
  5. Treatment Administration: Test agents are dosed on study-defined schedules — BTK inhibitors, venetoclax alone or with BCL-XL partners, CDK4/6 inhibitors, or metabolic-targeting compounds — with body weight and clinical condition recorded throughout.
  6. Longitudinal Monitoring: Tumor burden is tracked twice weekly by caliper (flank) or bioluminescence quantification (systemic designs) in vivo, with optional serial blood sampling for circulating MCL cells by human CD45 flow cytometry.
  7. Acquired-Resistance Cohorts: Where resistance evolution is the question, treatment is continued until progressive disease emerges, capturing BCL-2 family protein shifts — BCL-XL, MCL-1, and BCL-2 upregulation with BIM sequestration — by immunoprecipitation and western blot for mechanistic attribution.
  8. Endpoint Analysis and Reporting: At termination, tumors and spleens are harvested and weighed, disseminated organs are inspected and imaged ex vivo for luciferase studies, and tissues are processed for BCL-2, BCL-XL, MCL-1, BIM, and cleaved caspase-3 immunohistochemistry alongside Ki-67 proliferation and H&E; tumor-growth inhibition, bioluminescence kinetics, apoptosis markers, and survival data are compiled into the final report.
