EC109-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: EC109-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The EC109-luc orthotopic mouse model is a luciferase-engineered xenograft platform in which human esophageal squamous cell carcinoma cells grow within the esophageal wall of immunodeficient mice, permitting serial, noninvasive measurement of tumor burden by bioluminescence imaging. Alfa Cytology provides this model as an end-to-end preclinical solution, covering reporter cell line verification, surgical orthotopic implantation, scheduled in-life imaging sessions, randomized dosing arms, necropsy, and histopathological assessment, so that sponsors receive a complete, analysis-ready data package without building an in-house surgical and imaging program.

Overview of EC109-luc Orthotopic Mouse Model for Esophageal Cancer

Orthotopic implantation places tumor cells in the tissue compartment where the disease naturally arises, and this spatial context has profound consequences for tumor biology. In esophageal cancer, subcutaneous xenografts grow rapidly and are easy to measure, yet they lack the desmoplastic stroma, peristaltic mechanics, and lymphovascular anatomy of the esophagus. Orthotopic models overcome these limitations: tumors developing in the esophageal wall acquire an architecture closer to the human disease, with nodular mucosal growth, mural invasion from mucosa toward adventitia, lymphovascular permeation, and perineural infiltration—features repeatedly documented in luciferase-labeled ESCC orthotopic systems. Because the esophagus cannot be palpated or calipered, stable expression of firefly luciferase in EC109 cells provides the quantitative surrogate: emitted photon flux after D-luciferin injection correlates with viable tumor mass, converting tumor growth into a numeric readout that can be captured weekly in living animals.

The EC109-luc orthotopic model has become a practical workhorse for preclinical esophageal cancer research. Its applications span anti-tumor drug candidate evaluation, radiosensitization studies, gene function investigations—for example, pathway knockdown experiments in which bioluminescence trajectories reveal the in vivo consequence of molecular perturbation—and biomarker or combination-regimen screening. The model is also compatible with resistance research, since the parental EC109 background has been used to derive well-characterized drug-resistant derivatives. Compared with endpoint-only designs, the longitudinal imaging format reduces animal numbers, detects treatment effects earlier, and captures patterns of regression, stable disease, and relapse within individual subjects—attributes that make the platform well suited for late-stage lead optimization and translational decision-making.

Cell Line Information: EC109-luc

EC109-luc is generated by stably transducing the Eca-109 human ESCC line with a firefly luciferase reporter, combining one of the oldest and most widely used Chinese ESCC lines with an optical tracking module suitable for in vivo imaging. The parental line was established in 1973 from a middle-esophageal squamous cell carcinoma of a Chinese female patient and has been deposited with major cell banks. The line grows as adherent, polygonal, epithelial-like cells, expresses cytokeratin and EpCAM consistent with its squamous origin, and forms tumors in BALB/c nude mice. Principal characteristics are listed below.

Our Services

Few laboratories can justify the specialized microsurgery, imaging hardware, and quality-control burden that an orthotopic luciferase program requires. Alfa Cytology consolidates all of it: we maintain authenticated, mycoplasma-free EC109-luc stocks, perform the esophageal implantation procedure under aseptic conditions, execute scheduled bioluminescence reads, administer test articles precisely as specified, and close each study with full necropsy, organ imaging, and histology—freeing your scientists to concentrate on experimental logic and data interpretation.

Workflow of EC109-luc Orthotopic Mouse Model Construction

Construction of the EC109-luc orthotopic model follows a sequence of cell qualification, orthotopic delivery through the gastric subserosal route, and longitudinal optical follow-up. The workflow below is adapted from established luciferase-labeled ESCC orthotopic methods and is adjusted in cell number, volume, and imaging cadence according to each sponsor’s protocol.

  1. Reporter cell line verification: EC109-luc vials are revived, tested for mycoplasma contamination, confirmed by STR profiling, and validated for luciferase signal linearity before use.
  2. Inoculum preparation: Log-phase cells are trypsinized, counted, and mixed with Matrigel at the study-specified concentration, commonly on the order of 2 × 106 cells per µ20 µl.
  3. Mouse conditioning: Female BALB/c nude mice are quarantined, acclimatized, and randomized into groups before the surgical session.
  4. Anesthesia and exposure: Mice are anesthetized and placed supine; a 7–10 mm upper abdominal incision exposes the stomach and lower esophagus.
  5. Orthotopic inoculation: A 31-gauge needle loaded with the cell–Matrigel suspension is inserted through the anterior gastric wall and advanced subserosally across the esophagogastric junction, depositing cells slowly into the abdominal esophageal wall.
  6. Closure and recovery: The abdominal wall is sutured, analgesia is provided, and animals are monitored daily until full recovery.
  7. Serial bioluminescence imaging: Following intraperitoneal D-luciferin injection, photon flux is measured at defined intervals to chart engraftment, growth kinetics, and response to treatment.
  8. Study endpoint and tissue analysis: Mice are euthanized at protocol-defined endpoints for ex vivo imaging of the esophagus, regional nodes, and organs, followed by weighing, fixation, sectioning, and histopathological review.

Case Study-EC109-luc Orthotopic Mouse Model Development

In a recent preclinical collaboration, Alfa Cytology was engaged to develop the EC109-luc orthotopic model as the pharmacology platform for a sponsor’s candidate program. Our team completed reporter cell qualification and surgical implantation, confirmed reproducible engraftment by weekly bioluminescence imaging, and supported randomized efficacy arms in which imaging trajectories separated treatment groups well before terminal readouts. End-of-study histopathology verified orthotopic tumor location and preserved esophageal wall architecture. Detailed representative results from this and related EC109-luc studies are available on request under confidentiality.
