MCF-7 Metastatic Tumor Model Service for Breast Cancer

Fig 1: MCF-7 Metastatic Tumor Model for Breast Cancer preclinical research.

The MCF-7 metastatic tumor model brings estrogen receptor-positive, luminal-type breast cancer into immunodeficient hosts, with orthotopic and cardiac-injection formats that capture both hormone-dependent growth and spread to bone and lymph nodes. Alfa Cytology operates the complete MCF-7 workflow—from hormone-supplemented implantation and endocrine dosing to bioluminescence tracking and bone-lesion analysis—so ER-positive breast cancer programs can move from compound profiling to mechanism-informed decisions.

Overview of MCF-7 Metastatic Tumor Model for Breast Cancer

MCF-7 was established in 1973 by Soule and colleagues at the Michigan Cancer Foundation—the source of its name—from the pleural effusion of a 69-year-old woman with metastatic breast adenocarcinoma. It was the first human breast cancer line to survive indefinite serial passage, and it retains several hallmarks of differentiated mammary epithelium, including cytoplasmic estrogen receptors that process estradiol and the ability to form domes in culture. The cells display a luminal A phenotype—estrogen receptor positive, progesterone receptor positive, and without HER2 amplification—carry wild-type TP53, an activating PIK3CA mutation, and a hypertriploid karyotype with a modal chromosome number of 82, and express WNT7B together with insulin-like growth factor binding proteins BP-2, BP-4, and BP-5.

One operational fact defines the MCF-7 model: in immunodeficient mice, xenograft growth is estrogen dependent. Host animals are therefore primed with slow-release 17β-estradiol pellets before tumor cell delivery, and the hormone becomes a tunable variable—pellet dose has been shown to modulate both tumor take and the incidence of metastasis. In the cardiac-injection format, estrogen-supplemented nude mice develop radiographically detectable osteolytic bone lesions with essentially complete incidence within about a month, providing a workable model of ER-positive bone metastasis that most ER-negative lines cannot mimic; in the mammary fat-pad format, tumors grow orthotopically and, particularly in engineered derivatives such as Six1-overexpressing clones, metastasize to regional and distant lymph nodes. Luciferase-tagged MCF-7 variants allow this progression to be tracked non-invasively, and the line’s endocrine sensitivity makes it the standard substrate for tamoxifen, fulvestrant, and CDK4/6 inhibitor combination studies.

Cell Line Information: MCF-7

Reference culture of MCF-7 calls for insulin-containing medium—classically EMEM with 10% fetal bovine serum and 0.01 mg/mL bovine insulin, with DMEM-based formulations and phenol-red-free variants in common use—and the line is a slow, steady grower, typically doubling every 30 to 40 hours. Its documented STR profile, blood group antigens, and isoenzyme pattern allow unambiguous identity confirmation, which matters for a line cited in thousands of publications and circulated for five decades. The cells are large, adherent epithelial cells, around 20 to 25 microns in diameter, and under appropriate conditions they organize into three-dimensional domes—an unusual behavior among breast cancer lines that reflects their retained differentiation.

Our Services

Because hormone context shapes every readout in this model, Alfa Cytology treats estrogen management as a study variable rather than a background detail: pellet dose and timing, withdrawal schedules, and circulating estradiol status are planned with the same care as the treatment arms themselves. Sponsors receive in vitro potency data, in vivo efficacy and survival curves, and histology-ready tissues from a single, coordinated effort.

Workflow of MCF-7 Metastatic Tumor Model Construction

MCF-7 studies follow a distinctive sequence in which hormone priming precedes tumor implantation and imaging cadence follows the metastatic format. The stages below describe the most common configurations, from routine fat-pad efficacy work to cardiac-injection bone metastasis programs.

  1. Cell authentication and expansion: MCF-7 stocks are confirmed by STR profiling, cleared by mycoplasma testing, and in vitro expanded in insulin-containing medium under strict passage discipline.
  2. Hormone priming: Recipient female nude mice are implanted with slow-release 17β-estradiol pellets several days before tumor cell delivery; pellet dose, commonly between 0.18 and 0.72 mg depending on the design, is recorded as a study variable.
  3. Inoculation: Luciferase-labeled or parental MCF-7 cells are injected in a supportive matrix: typically 1 × 106 to 5 × 106 cells into the mammary fat pad or flank, and roughly 1 × 105 cells via the left cardiac ventricle for bone-metastasis formats.
  4. Establishment and imaging baseline: Tumor take is confirmed by palpation, with weekly bioluminescence or radiographic readouts; enrollment is triggered once tumors enter the preset volume window.
  5. Randomization and endocrine dosing: Cohorts are balanced into vehicle, benchmark (such as tamoxifen or fulvestrant), and test-article arms; dosing proceeds per schedule, with estradiol-withdrawal arms included where the design requires them.
  6. In-life monitoring: Caliper or bioluminescence measurements, body weight, and clinical observations are logged at defined intervals, and bone lesion development is scored radiographically in cardiac-injection studies.
  7. Termination and tissue collection: Tumors, lymph nodes, and hind-limb bones are harvested; lesion area and bone mineral density (DXA) are quantified, and histology, IHC, and molecular endpoints are finalized before statistical summarization and report issuance.

Case Study-MCF-7 Metastatic Tumor Model Development

Alfa Cytology has applied the MCF-7 platform to preclinical questions including endocrine-agent benchmarking, CDK4/6 inhibitor combinations designed to delay or reverse resistance, and estrogen-dependent bone-colonization studies with radiographic lesion quantification confirmed in vivo. Programs have also used luciferase-tagged derivatives for non-invasive tracking of orthotopic progression and lymph-node involvement, with readouts spanning tumor volume, survival, histopathology, and hormone-pathway biomarkers; program-specific materials can be shared once confidentiality agreements are in place.

Why Choose Alfa Cytology?

Hormone-dependent models punish imprecise execution: pellet timing, estradiol depletion, and imaging cadence can each decide whether a study succeeds. Alfa Cytology has built its MCF-7 operations around exactly these details.

  • Endocrine expertise: estradiol pellet regimens, withdrawal arms, and hormone-status monitoring are managed by experienced staff.
  • Multiple metastatic formats: orthotopic fat-pad growth, cardiac-injection bone colonization, and lymph-node tracking from one cell source.
  • Imaging breadth: bioluminescence, radiography, and DXA-based bone measurements integrated into study designs.
  • Resistance coverage: parental and endocrine-resistant derivative lines support mechanism-of-resistance programs.
  • Verified starting material: STR-confirmed identity, mycoplasma clearance, and documented low-passage handling.
  • Reporting clarity: milestone updates, raw-data access, and statistics-ready datasets on delivery.
