K7M2.WT Syngeneic Model Service for Sarcoma

Fig 1: K7M2.WT Syngeneic Model for Sarcoma preclinical research.

The K7M2.WT syngeneic model is an immunocompetent preclinical sarcoma system in which murine osteosarcoma cells engraft in BALB/c hosts and progress with spontaneous metastasis to the lung. Alfa Cytology builds this model end to end for sponsors—authenticated cell supply, in vivo inoculation, treatment administration, and metastasis endpoint analysis—so sarcoma programs can generate translational evidence quickly.

Overview of K7M2.WT Syngeneic Model for Sarcoma

K7M2.WT is a murine osteosarcoma cell line that originated from a spontaneous bone tumor in a BALB/c mouse. Osteosarcoma, the most common primary malignant tumor of bone, belongs to the sarcoma family. During its derivation (Khanna et al., 2000), parental K7 cells were injected into the proximal tibia of a BALB/c host; the resulting primary tumor metastasized spontaneously to the lung, and the pulmonary deposits were serially passaged through paraosteal tibial muscle flaps before being dissociated into a single-cell suspension to establish K7M2.WT. The line retains an osteoblastic phenotype—expressing bone sialoprotein, osteopontin, biglycan, decorin, and ezrin—and is highly aggressive: following inoculation into syngeneic BALB/c mice, spontaneous pulmonary metastases arise in over 90% of animals.

Because donor and host share the same genetic background, K7M2.WT tumors grow in animals with intact immunity, which distinguishes the model from human xenografts propagated in immunodeficient rodents. That feature makes it a practical surrogate for studying tumor–immune interactions, the metastatic cascade, and the activity of immunomodulatory agents. The cells can additionally be engineered to express firefly luciferase, enabling non-invasive bioluminescence tracking of primary lesions and lung colonization across the study. Depending on the scientific question, K7M2.WT can be implanted subcutaneously for routine efficacy work, orthotopically into the tibia to mimic the natural bone microenvironment, or injected intravenously to model experimental metastasis.

Cell Line Information: K7M2.WT

K7M2.WT—also recorded as K7M2 wt or simply K7M2—was established by Khanna and colleagues through a deliberate in selection strategy rather than a single isolation event. The parental K7 line, itself derived from a spontaneous BALB/c osteosarcoma, was reintroduced into the proximal tibia; tumor-bearing lungs were then harvested, and lung deposits were cyclically re-implanted into paraosteal tibial muscle flaps of naïve animals until a uniformly metastatic tumor emerged. Dissociation of that tumor yielded K7M2.WT. Its molecular footprint confirms bone-lineage commitment—bone sialoprotein, osteopontin, biglycan, decorin, and ezrin are all expressed—while CD31 positivity and factor VIII production reflect vascular-associated features reported for the line. These traits, together with its reproducible lung-homing behavior, have made K7M2.WT a reference tool for sarcoma biology and immuno-oncology screening.

Our Services

With this immunocompetent platform at its core, Alfa Cytology shapes each K7M2.WT study around a defined program question—whether that is ranking anti-metastatic candidates, stress-testing a combination immunotherapy, or qualifying an imaging biomarker. The team takes projects from cell line engineering and pilot inoculation through full in vivo efficacy runs with histopathology and immune profiling, returning interpretable data on agreed timelines.

Workflow of K7M2.WT Syngeneic Model Construction

Construction of a K7M2.WT syngeneic study follows a logical sequence from cell qualification to data delivery. Although each program is customized, the workflow below captures the standard path that most sarcoma efficacy and metastasis projects take.

  1. Cell authentication and expansion: K7M2.WT cells are verified by STR profiling, confirmed mycoplasma-free, and expanded in vitro under controlled, low-passage conditions.
  2. Study design: The implantation route (subcutaneous flank, orthotopic intra-tibial, or tail-vein lung colonization), group size, randomization, and blinding scheme are fixed according to the endpoint of interest.
  3. Tumor cell inoculation: A single-cell suspension, typically 1 × 106 to 5 × 106 cells in a 100 µL volume depending on the route, is implanted into immunocompetent BALB/c mice, with baseline imaging recorded where applicable.
  4. Tumor establishment monitoring: Subcutaneous and orthotopic lesions are measured by caliper two to three times weekly; luciferase-tagged studies add weekly IVIS sessions for non-invasive burden tracking.
  5. Randomization and treatment: Once tumors reach the predefined volume window (commonly 80–150 mm3), animals are randomized into vehicle, benchmark, and test-article groups, and dosing begins per the approved schedule.
  6. In-life observations: Body weight, clinical signs, and humane endpoints are recorded throughout, with interim imaging to flag early metastatic spread.
  7. Endpoint analyses and reporting: At study termination, tumors are excised and weighed, lungs are scored for surface nodules and processed for H&E, and tumor-immune profiling (IHC and flow cytometry) is performed before statistical analysis and report assembly.

Case Study-K7M2.WT Syngeneic Model Development

Alfa Cytology has applied K7M2.WT-based systems across a range of preclinical sarcoma programs. Supported work has included luciferase-tagged derivatives for whole-body imaging of tumor spread, orthotopic tibial implantation that recreates local bone destruction, and tail-vein experimental metastasis setups for rapid screening. Within these frameworks, sponsors have evaluated checkpoint blockade, combination regimens, targeted small molecules, and novel drug-delivery constructs, with readouts spanning tumor growth curves, metastatic burden, survival, and immune microenvironment profiling. Detailed program data can be shared under confidentiality arrangements as study contexts allow.
