Urethane-Induced Lung Cancer Model Service

Fig 1: Urethane-Induced Lung Cancer Model preclinical research.

The urethane-induced lung cancer model is the reference system for generating Kras-driven pulmonary adenomas and adenocarcinomas in immunocompetent mice through nothing more demanding than a simple intraperitoneal injection of a water-soluble, stable carcinogen. Alfa Cytology delivers this deceptively simple platform with full scientific rigor, managing strain selection, carcinogen administration, longitudinal tumor monitoring, and complete histopathological scoring so that sponsors can generate publication-ready lung tumor data with minimal operational burden.

Overview of Urethane-Induced Lung Cancer Model

Urethane, the common name for ethyl carbamate, is a fermentation by-product that occurs naturally in fermented foods and beverages, yet in the laboratory it has become one of the most reliable and economical carcinogens ever characterized. The compound is inexpensive, water-soluble, chemically stable, and comparatively safe to handle, and a single intraperitoneal injection in a susceptible mouse strain is sufficient to initiate lung tumorigenesis without any additional promoter. Its mechanism has been mapped in detail: hepatic and pulmonary cytochrome P450, chiefly CYP2E1, oxidizes urethane in two steps to vinyl carbamate and then to vinyl carbamate epoxide, a potent electrophile that forms etheno-DNA adducts. Because lung tissue expresses the Kras oncogene at exceptionally high transcriptional levels relative to other organs, pulmonary cells suffer mutagenesis disproportionately, which explains the pronounced lung tropism of the model.

The lesions that emerge reproduce the human disease spectrum with unusual fidelity. On sensitive backgrounds such as A/J, multiple bronchioloalveolar adenomas become visible on the lung surface by about 25 weeks after a single injection, and a subset progress to adenocarcinomas by 40 weeks, histologically echoing the adenocarcinoma subtype of human non-small cell lung cancer. Roughly 80% of the tumors carry an activating Kras Q61 mutation, with Q61L predominating in A/J mice and Q61R in C57BL/6 animals, mirroring one of the most important oncogenic lesions in human lung adenocarcinoma. Resistant strains such as C57BL/6 and FVB/N respond to a multi-dose weekly regimen with near-100% tumor incidence, and environmental modifiers such as high-fat feeding or atherogenic diets measurably shift tumor burden, allowing the model to serve genetic-modifier discovery, chemoprevention trials, inflammation studies, and therapeutic evaluation conducted in vivo.

Principles of Urethane-Induced Lung Cancer Model Construction

The urethane model is conceptually simple but operationally nuanced: tumor output is governed by strain susceptibility, dosing schedule, and the genetic susceptibility of the Kras locus in lung epithelium. The governing elements are summarized below.

Element Agent / Factor Function in Model Construction Representative Conditions
Carcinogen Urethane (ethyl carbamate), intraperitoneal injection A complete carcinogen requiring no separate promoter; CYP2E1 oxidizes it in two steps to vinyl carbamate epoxide, which covalently binds DNA to form etheno adducts (1,N6-ethenodeoxyadenosine and 1,N4-ethenodeoxycytidine) that convert to mutations during replication. 1 g/kg (equivalent to 1 mg/g body weight) as a single injection in susceptible strains; the same dose given weekly for ~10 injections in resistant strains
Genetic driver Kras Q61 activating mutation Approximately 80% of induced lung tumors carry a single driver mutation at Kras codon 61, Q61L predominating in A/J and Q61R in C57BL/6 mice; the lung's unusually high Kras transcription makes this organ the primary mutagenesis target. Mutational status verifiable by sequencing at endpoint
Strain sensitivity Genetic background of the host A/J mice are highly susceptible and develop multiple adenomas after one injection; FVB/N shows intermediate sensitivity; C57BL/6 is resistant to single doses but reaches near-100% incidence under weekly multi-dose regimens. Single-dose protocol for A/J; 10-weekly-injection protocol for C57BL/6J
Lesion spectrum Bronchioloalveolar hyperplasia → adenoma → adenocarcinoma Lesions progress through atypical adenomatous hyperplasia and papillary or solid adenomas toward adenocarcinoma, recapitulating the adenocarcinoma subtype of human NSCLC; multiplicity and nodule diameter are the standard burden metrics. Adenomas grossly visible from ~25 weeks (A/J single dose) or ~14 weeks (FVB/N); adenocarcinomas by ~40 weeks
Environmental modifiers Diet and inflammation High-fat diet increases tumor burden, whereas atherogenic diets can suppress it; NF-κB, IL-1β, IL-6, and STAT3 inflammatory signaling accompanies and promotes tumorigenesis. Optional dietary arms or anti-inflammatory interventions per study design

Our Services

From the first injection to the final tumor count, Alfa Cytology engineers each urethane study around the client's strain of choice and analytical goals, whether that means maximizing multiplicity on A/J mice, running chemoprevention arms on C57BL/6 cohorts, or capturing the adenoma-to-adenocarcinoma transition with serial imaging.

Workflow of Urethane-Induced Lung Cancer Model Construction

A urethane engagement is operationally lightweight up front, with the analytical weight shifting to the long latency phase and the terminal endpoint. A typical study proceeds through the following steps.

  1. Strain and regimen selection: The scientific question determines the host, A/J for single-injection sensitivity, C57BL/6J for compatibility with genetic tools under a 10-weekly-injection schedule, or FVB/N for intermediate kinetics, along with group sizes and any dietary or therapeutic arms.
  2. Animal acclimatization and randomization: Six- to eight-week-old mice undergo a minimum one-week habituation period before being randomized by weight into control and treatment cohorts.
  3. Urethane solution preparation: Urethane is dissolved at the working concentration in sterile saline, prepared fresh or from sealed stock, and kept at room temperature given the compound's excellent aqueous stability.
  4. Carcinogen administration: Animals receive the protocol-defined intraperitoneal injections, a single 1 g/kg dose for susceptible strains or weekly 1 g/kg doses for approximately ten weeks in resistant strains, with injection-site and general-condition checks after each dose.
  5. Latency-phase husbandry and monitoring: For the subsequent 15 to 40 weeks, animals are maintained under standard conditions with regular weighing and welfare scoring; optional dietary interventions, test articles, or candidate therapeutics are administered on the sponsor-defined schedule.
  6. Interim tumor surveillance (optional): Where timelines demand earlier insight, cohorts can be examined by high-resolution micro-CT or processed at staggered interim time points to profile lesion evolution from hyperplasia through adenoma.
  7. Endpoint euthanasia and lung harvest: At the final time point, animals are euthanized, lungs are inflated and excised en bloc, and surface nodules are counted and measured to derive tumor multiplicity and burden.
  8. Histopathology and molecular characterization: Tissues are fixed, sectioned, and graded for hyperplasia, atypical adenomatous hyperplasia, adenoma, and adenocarcinoma, complemented by Ki-67 immunostaining, inflammatory marker panels, and Kras codon 61 mutational analysis.
