KYSE-510 Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: KYSE-510 Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The KYSE-510 orthotopic mouse model grows a well-differentiated human esophageal squamous cell carcinoma line in the mouse esophageal wall, delivering an anatomically faithful preclinical system for studying tumor progression and evaluating therapeutic strategies. Alfa Cytology offers this model as a complete, quality-controlled preclinical service: authenticated KYSE-510 banks, aseptic orthotopic microsurgery, structured in-life monitoring, randomized treatment arms, and thorough necropsy with histopathology—all executed and documented so that sponsors receive decision-ready data without standing up their own esophageal surgical program.

Overview of KYSE-510 Orthotopic Mouse Model for Esophageal Cancer

Most widely used ESCC cell lines are poorly differentiated, but KYSE-510 occupies a distinctive niche: it was established from a well-differentiated squamous carcinoma with moderate invasion of the esophageal adventitia, and it retains that differentiated character in culture and in xenografts. This matters scientifically. Well-differentiated tumors preserve more of the architectural organization, keratinization patterns, and growth restraint of clinical ESCC, so findings generated on this background can be argued to sit closer to the differentiated end of the disease spectrum—an advantage when studying early-stage biology, differentiation-targeting agents, or radiotherapy response. Its donor history adds another layer of relevance: the primary tumor was resected after cisplatin and radiotherapy, so the line carries the genomic imprint of treatment pressure and is a natural fit for resistance and radiosensitization research.

Comparative characterization of KYSE510 and other human esophageal squamous cell carcinoma cell lines, showing the high proliferative capacity of KYSE510 relevant to preclinical tumor model development. (Miyake, Takanori, et al., 2026)

Genomically, KYSE-510 is described as carrying a TP53 mutation together with amplifications of MYC, HST1, and cyclin D1, a combination that places it at the intersection of transcriptional addiction, cell-cycle dysregulation, and DNA-damage response. Whole-exome and transcriptome profiling of the common ESCC panel ranks it among the lowest in tumor mutational burden, with a mutation signature consistent with homologous-recombination deficiency—features that frame it as a comparatively “clean” background for pathway-specific interrogation, in contrast to the hypermutated KYSE-150. In practice, the KYSE-510 orthotopic model has been applied to molecular mechanism studies, including EMT and invasion research through PURα overexpression validated in vitro, and to compound evaluation tested in vivo. Because the model carries no optical reporter by default, disease course is followed through scheduled endoscopic inspection, ultrasound or micro-CT sizing where available, body weight trajectories, and terminal histology—with a luciferase-labeled variant available when sponsors prefer an imaging-centric design.

Cell Line Information: KYSE-510

KYSE-510 is a well-differentiated human ESCC line established by Shimada and colleagues from the cervical esophagus of a 67-year-old Japanese woman whose stage III tumor showed moderate invasion of the adventitia and was resected after treatment with cisplatin and radiotherapy. The line is distributed by DSMZ (ACC 374) and JCRB (JCRB1436), is part of the Cancer Cell Line Encyclopedia, and has been characterized in vitro through exome, RNA-seq, methylation, and pharmacogenomic datasets. In nude mice it regenerates well-differentiated squamous tumors, with tumorigenicity confirmed in vivo. Its principal characteristics are listed below.

Our Services

Executing an orthotopic study on a well-differentiated line demands particular care: slower, more organized tumors can mask sloppy technique, and only disciplined cell handling and surgical consistency keep cohorts comparable. Alfa Cytology brings exactly that discipline—STR-authenticated, mycoplasma-free KYSE-510 working banks; standardized low-passage expansion; aseptic esophageal wall implantation with defined inocula; scheduled endoscopic and imaging checks; weight-based humane monitoring; and terminal necropsy with H&E and immunohistochemistry—documented in a single, audit-ready report for each study.

Workflow of KYSE-510 Orthotopic Mouse Model Construction

Model construction proceeds from authenticated cell banks through surgical orthotopic delivery to longitudinal observation and terminal histology. Inoculum size—typically on the order of 1–5 × 106 cells in a 25–50 µl volume—monitoring modality, and endpoint criteria are fixed with the sponsor at study design, and an optional luciferase-labeled variant can substitute when imaging readouts are preferred.

  1. Cell bank release: KYSE-510 vials are revived from the working bank, tested negative for mycoplasma, and authenticated by STR profiling against the reference profile.
  2. Inoculum preparation: Log-phase cells are harvested by trypsinization, counted, and resuspended in a defined volume of buffer with or without a Matrigel-type carrier, according to the study design.
  3. Animal preparation: BALB/c nude mice complete quarantine and acclimatization and are randomized into cohorts balanced by body weight.
  4. Anesthesia and exposure: Under general anesthesia with perioperative analgesia, a small upper abdominal incision exteriorizes the stomach and distal esophagus.
  5. Orthotopic implantation: A fine-gauge needle injects the cell suspension into the esophageal wall; the raised intramural wheal confirms correct placement before layered closure.
  6. Postoperative monitoring: Animals recover on warming pads and are checked daily for body weight, feeding behavior, and incision healing.
  7. In-life disease follow-up: Tumor development is tracked by scheduled endoscopy and, where available, ultrasound or micro-CT, supplemented by body weight curves and condition scoring; luciferase-labeled variants are imaged by bioluminescence instead.
  8. Terminal analysis: At protocol endpoints, mice are euthanized; the esophagus, regional lymph nodes, and organs are harvested for gross pathology, tumor measurement, and H&E/immunohistochemical processing.

Case Study-KYSE-510 Orthotopic Mouse Model Development

The KYSE-510 orthotopic model is an active offering within Alfa Cytology's esophageal cancer model portfolio. In a recent preclinical engagement, our team expanded low-passage, STR-verified KYSE-510 cells, established orthotopic tumors in nude mice, and followed disease through scheduled endoscopic checks with terminal histopathology confirming well-differentiated squamous growth in the esophageal wall. Randomized treatment arms were supported with body weight tracking and defined humane endpoints. Further representative results from KYSE-510 orthotopic studies are available upon request.
