MM.1S / MM.1S-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: MM.1S / MM.1S-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The MM.1S / MM.1S-luc orthotopic mouse model of multiple myeloma engrafts the field’s standard glucocorticoid-sensitive human myeloma line into the bone-marrow compartment, where disseminated disease is monitored longitudinally by luciferase bioluminescence alongside serum light-chain tracking and its dexamethasone-resistant sibling MM.1R provides a built-in resistance control. Alfa Cytology delivers this platform as an integrated preclinical service: authenticated CRL-2974 banking, luciferase line qualification, intravenous or intra-tibial engraftment in NOD/SCID or NSG recipients, scheduled whole-body imaging, proteasome-inhibitor and steroid reference dosing, and endpoints spanning marrow chimerism, paraprotein quantification, survival analysis, and histopathology.

Overview of MM.1S / MM.1S-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

MM.1S is a human multiple myeloma cell line deposited with ATCC as CRL-2974, derived from the peripheral blood of a 42-year-old female patient whose IgA lambda myeloma had progressed despite steroid-based therapy. It belongs to the MM.1 family of lines that parallel disease progression from a single patient: MM.1S expresses the glucocorticoid receptor and is exquisitely sensitive to dexamethasone, while its sibling MM.1R (ATCC CRL-2975) was selected under months of continuous drug exposure and is stably resistant — having lost the GRα isoform and acquired alternatively spliced GRβ with low hormone-binding affinity. That S/R pairing has made the MM.1 pair the definitive system for glucocorticoid pharmacology in myeloma. Phenotypically, MM.1S is a B lymphoblast with mixed suspension and lightly adherent growth, a 72-80-hour doubling time, a notoriously slow post-thaw recovery that can take two weeks, and a documented surface profile — CD25, CD38, CD52, and CD59 positive, CD19 and CD20 negative, low CD10 and CD45 — that reads like a textbook plasma-cell immunophenotype and renders the line a standard target for CD38-directed, CD52-directed, and CAR-T/CAR-NK killing assays.

Fig 2: Reference figures for MM.1S / MM.1S-luc cell-related literature.Fig 1. Disseminated MM.1S-luc myeloma model in mice treated with PBS or anti-BCMA immunotoxin LMB-75, showing dorsal and ventral bioluminescence images at days 4, 10, 17, 28, 38 and 47 where PBS-group MM.1S-luc tumor burden progressively increased (luminescence rising to ~2×10⁶ units with intense skull/spine/limb signals) while LMB-75 treatment eliminated detectable MM.1S-luc signal from day 10 onward, demonstrating durable complete remission. (Shancer, Zoe, et al., 2019)

As an in vivo system, MM.1S-luc is among the most validated disseminated myeloma models in the literature. Early luciferase tagging — via the MM.1S-pMMP-LucNeo construct — produced reproducible in vivo growth in both SCID-beige and NSG hosts with clean bioluminescence tracking of disseminated disease. In the classic NOD/SCID implementation, 10 million eGFP-luciferase-fusion MM.1S cells injected intravenously established measurable burden, and therapeutic arms defined the model’s pharmacology: anti-IGF-1R antibody alone was inactive, while melphalan or bortezomib alone prolonged survival, and combinations produced significant tumor-burden reduction and the longest survival of any arm. Engraftment efficiency data position MM.1S at the top of the myeloma xenograft hierarchy — among lines profiled, MM.1S achieved roughly 75% bone-marrow chimerism six weeks after intravenous injection into preconditioned NSG mice, the highest of the lines tested. Imaging characterizations further show bioluminescence concentrating in marrow-rich skeletal sites with the spine carrying the highest burden, and modern constructs such as the CMV-driven luciferase-eGFP cassette cloned by limiting dilution serve as reference target cells for CAR-T and CAR-NK optimization. Together, these properties make the MM.1S-luc orthotopic platform the go-to disseminated myeloma system for proteasome-inhibitor benchmarking, glucocorticoid and IGF-pathway programs, ADC and antibody evaluation, and cellular-therapy development.

Our Services

MM.1S is a line that quietly tests a laboratory’s patience and rewards its discipline: the 72-80-hour doubling time, the two-week post-thaw recovery, and the rosette-forming over-confluence all punish rushed handling — while the S/R sibling pair punishes sloppy controls. Alfa Cytology manages both realities as routine: authenticated CRL-2974 stocks cultured with seed-density discipline, reporter lots released only after uniform in vitro luminescence, dexamethasone-sensitivity verified as a lot-release assay, and MM.1R arms available wherever resistance context matters. Engraftment follows written intravenous or intra-tibial protocols in NOD/SCID or NSG hosts, imaging concentrates on the spine-predominant pattern the model is known for, and terminal analyses resolve marrow chimerism, light chain, survival, and histology. From thaw to final report, Alfa Cytology keeps the MM.1S / MM.1S-luc orthotopic workflow under one accountable roof.

Workflow of MM.1S / MM.1S-luc Orthotopic Mouse Model Construction

The MM.1S-luc orthotopic workflow is built around the model’s two signatures: spine-predominant marrow dissemination on imaging, and the glucocorticoid-sensitive/resistant pharmacology anchored by MM.1R. The sequence below is the standard construction-to-analysis flow.

  1. Authentication and lot-release assays: MM.1S / MM.1S-luc stocks are expanded from authenticated CRL-2974 master banks at controlled seed density, mycoplasma-tested, STR-verified, and released only after uniform luciferase expression is confirmed in vitro and dexamethasone sensitivity is documented as a functional lot-release assay.
  2. Engraftment cell preparation: Suspension cultures maintained at 0.4-0.8×106 cells/mL are harvested in log phase, washed, counted, and resuspended in sterile PBS at the challenge dose (about 106 cells in 100 µL for preconditioned NSG hosts; up to 107 cells in published NOD/SCID designs; 2-5×106 cells for intra-tibial delivery).
  3. Recipient preparation: NOD/SCID or NSG mice are acclimatized, body-weight stratified, and where protocols call for it, preconditioned with 175 cGy total-body irradiation 24 hours before inoculation; baseline serum is banked for light-chain measurement.
  4. Systemic or marrow-seated engraftment: Cells are injected via the lateral tail vein for disseminated designs, or microinjected into the tibial marrow cavity for orthotopic seating; baseline bioluminescence after D-luciferin (75-150 mg/kg) confirms deposition, with spinal signal documented as the model’s signature pattern.
  5. Longitudinal monitoring: On a fixed schedule, in vivo whole-body imaging tracks burden across spine, pelvis, and long bones, with serial serum draws quantifying lambda light chain; body weight, posture, and hindlimb function are scored as myeloma-specific clinical signs.
  6. Randomized treatment with resistance context: Cohorts are randomized by matched photon flux and paraprotein level; bortezomib, melphalan, dexamethasone, or test agents are dosed per protocol, with MM.1R arms incorporated wherever glucocorticoid-resistance context is required.
  7. Endpoint and marrow analysis: At termination, spine, pelvis, and long bones are collected with spleen and viscera; ex vivo imaging localizes deposits; human CD138 flow cytometry quantifies marrow chimerism; serum light chain is quantified; survival data feed Kaplan-Meier analysis; and H&E with CD138 immunostaining documents plasma-cell infiltration.

Case Study-MM.1S / MM.1S-luc Orthotopic Mouse Model Development

Alfa Cytology has implemented the MM.1S orthotopic platform in preclinical programs spanning proteasome-inhibitor and glucocorticoid benchmarking, IGF-pathway antibody evaluation, and CAR-T/CAR-NK target-line workflows. In these programs, serial bioluminescence tracked the model’s characteristic spine-predominant dissemination alongside serum light-chain curves, randomized arms started from matched signal and paraprotein baselines, MM.1R sibling arms supplied dexamethasone-resistance context, and terminal analyses combined ex vivo skeletal imaging, human CD138 marrow quantification, and survival analysis. Where programs required it, CD49d/CD29 adhesion-marker status was assessed to contextualize homing behavior. Study-specific parameters and outcomes are retained as sponsor-confidential deliverables, and representative outlines can be furnished upon request.
