Meth A Syngeneic Model Service for Sarcoma

Fig 1: Meth A Syngeneic Model for Sarcoma preclinical research.

The Meth A syngeneic model is one of the longest-serving preclinical sarcoma platforms, pairing a methylcholanthrene-induced BALB/c fibrosarcoma with its syngeneic host so that tumor growth and host immunity can be studied together in vivo. Alfa Cytology runs complete Meth A programs—cell authentication, tumor implantation, treatment scheduling, and immune-focused endpoint analysis—giving sponsors a dependable route from candidate selection to decision-grade preclinical data.

Overview of Meth A Syngeneic Model for Sarcoma

Meth A is a fibrosarcoma that arose in a BALB/c mouse after induction with 3-methylcholanthrene, a polycyclic aromatic hydrocarbon carcinogen, and it was adapted to culture in the 1970s (DeLeo et al., 1977). Because carcinogen-induced sarcomas carry tumor-specific transplantation antigens that differ from tumor to tumor, Meth A quickly became a prototype for tumor immunology: its individually distinct surface antigen was described in detail in 1977, and the line later proved important in early studies of the p53 oncoprotein, when stabilized transformation-related p53 protein was reported in Meth A and related murine fibrosarcomas (Halevy et al., 1989).

Unlike human tumor xenografts, which must be grown in immunodeficient hosts, Meth A grows in syngeneic BALB/c mice whose immune compartments stay fully functional, so tumor rejection, immune escape, and therapy-induced immune activation can be observed directly. The cells carry an H-22d MHC class I profile with strong surface expression of H-2D2d, H-2K2d, and H-2L2d, divide with a ploidy-dependent doubling time of roughly 24 to 30 hours, and form reproducible flank tumors after subcutaneous injection of about 1 × 106 to 2 × 106 cells. Derivatives selected for enhanced metastatic behavior, such as the ML-01 and MH-01 clones, extend the system beyond the primary tumor setting, and the model remains in routine use for vaccine, cytokine-therapy, and combination immunotherapy studies.

Cell Line Information: Meth A

Meth A is generally maintained in RPMI 1640 medium supplemented with 10% serum and shows adherent, fibroblast-like growth with a ploidy-dependent doubling time of about 24 to 30 hours. Serial syngeneic passage preserves its antigenic character well enough for reproducible immunological work, which helps explain why the line has stayed in continuous service for close to fifty years—spanning tumor antigen discovery, biological response modifiers such as protein-bound polysaccharide K, and engineered vaccine constructs that secrete cytokines like tumor necrosis factor or interleukin-15.

Our Services

Beyond routine efficacy testing, Alfa Cytology uses the Meth A system to generate mechanism-rich datasets: NK-cell and T-cell infiltration, antigen-dependent responses, and cytokine signatures are captured alongside tumor volume, so a single study can support both go/no-go decisions and biological rationale. Engagements can begin with in vitro characterization or a pilot inoculation study and scale to full combination trials, with pathologist-reviewed histology and flow-based immune profiling included as standard deliverables.

Workflow of Meth A Syngeneic Model Construction

A Meth A study proceeds through defined stages from cell preparation to final report. The sequence below reflects the most common configuration; timing, group sizes, and endpoints are adjusted to each sponsor’s objectives.

  1. Cell recovery and qualification: Cryopreserved Meth A stocks are revived, identity-confirmed, tested for mycoplasma, and in vitro expanded to the scale required by the study plan.
  2. Protocol definition: Implantation site (most often the subcutaneous flank), enrollment targets, randomization rules, and humane endpoints are documented before any animal procedures begin.
  3. Tumor implantation: Single-cell suspensions of 1 × 106 to 2 × 106 viable Meth A cells are injected subcutaneously into BALB/c mice, with implantation day recorded as the study reference point.
  4. Establishment phase: Palpable masses typically appear within the first week; tumor dimensions are captured on a fixed caliper schedule to build growth curves.
  5. Cohort balancing and dosing: Animals are distributed across vehicle, benchmark, and test-article arms once tumors enter the preset size window, and dosing follows the approved regimen.
  6. Health surveillance: Body weight, condition scores, and cage-side observations are logged on a defined cadence, with interim blood sampling added where pharmacodynamic markers are requested.
  7. Necropsy and analysis: At termination, tumors and organs are harvested and weighed; histopathology, immunohistochemistry, flow cytometry, and serum cytokine measurements are completed before statistical summarization and report delivery.

Case Study-Meth A Syngeneic Model Development

Alfa Cytology has conducted Meth A-based preclinical work covering cytokine-armed tumor cell vaccines, checkpoint inhibitor benchmarking, combinations that pair targeted agents with immune stimulation, and natural-product immunomodulators. Endpoints across these programs have ranged from growth curves and survival to phenotypic analysis of T-cell and NK-cell populations within the tumor microenvironment, and study-specific datasets can be shared under confidentiality as discussions progress.

Why Choose Alfa Cytology?

Evaluating sarcoma candidates in immunocompetent systems calls for a partner that respects both the biology of a classic model and the discipline of modern preclinical operations. Alfa Cytology combines the two, so the data sponsors receive hold up under internal and external scrutiny.

  • Immunologically authentic: a chemically induced sarcoma with defined tumor antigens delivers immune readouts in vivo that xenograft systems cannot reproduce.
  • Flexible dosing designs: single agents, combinations, metronomic schedules, and systemic or local routes are all accommodated.
  • Mechanistic depth: histopathology, immunohistochemistry, and multiparameter flow cytometry are performed under one roof.
  • Regulatory-minded operations: IACUC-supervised, SPF-housed animals with enforced humane endpoints.
  • Responsive communication: milestone-based updates keep sponsors informed between report deliveries.
  • Complete transparency: authenticated cells, documented randomization, and full access to raw data at every stage.
