A549 / A549-luc Orthotopic Mouse Model Service for Lung Cancer

Fig 1: A549 / A549-luc Orthotopic Mouse Model for Lung Cancer preclinical research.

The A549 / A549-luc orthotopic mouse model establishes the most widely used human lung adenocarcinoma line directly within the lung parenchyma, where tumors invade through visceral pleura, seed the contralateral lung, and metastasize to brain and liver at rates that faithfully reproduce advanced non-small cell lung cancer. Alfa Cytology builds this model with the refined intercostal microinjection technique — a sub-one-minute procedure with under 5% operative mortality and 90% tumor take — and supports it with luciferase-based bioluminescence tracking in vivo, spiral CT volumetrics, and systematic brain-and-liver metastasis screening, giving your NSCLC program a rigorously benchmarked test system.

Overview of A549 / A549-luc Orthotopic Mouse Model for Lung Cancer

A549 is the reference human lung adenocarcinoma line: established by D.J. Giard in 1972 from explant culture of a lung tumor of a 58-year-old Caucasian man and deposited at ATCC (CCL-185), it is a hypotriploid alveolar basal epithelial line that doubles roughly once a day and carries the KRAS G12S and STK11 (LKB1) mutations on a TP53 wild-type background, with the CDKN2A locus inactivated — the genotype of a large, therapeutically stubborn subgroup of KRAS-driven NSCLC. Orthotopic implantation unlocks its full clinical behavior. In the landmark characterization of this model, 106 A549 cells in 20 µL of Matrigel-containing buffer injected 3 mm deep through the intercostal space produced solitary primary nodules in 100% of mice, and — distinctively — every animal developed multiple intrapulmonary metastases across both lungs (averaging 28.3 ± 16.9 surface nodules), while visible lymph-node metastasis remained rare; high c-MET expression was implicated in this intraorgan spread.

Independent validations have deepened the picture. An intrathoracic-inoculation protocol achieved a 90% tumor formation rate with a median survival of 30.7 days (range 20–41) and 100% intrathoracic metastasis involving the contralateral lung, mediastinum, and chest wall, with body-weight decline from about day 10 serving as a simple clinical sentinel; histopathologic series further documented brain metastases in roughly two-thirds of grafted mice and liver micrometastases in about 40%, alongside visceral-pleura invasion and lymphovascular emboli — a metastatic portfolio closer to patient autopsy series than to any flank xenograft. Method refinements continue: multicellular spheroids of fluorescent A549 cells inoculated intercostally progress through stages resembling clinical NSCLC and remain drug-responsive, as shown by cisplatin validation. The A549-luc variant enables quantitative whole-body and thoracic bioluminescence in vivo from the first week, while non-luc configurations are followed by spiral CT or ultrasound; together these options support chemotherapy benchmarks (cisplatin, paclitaxel), targeted and metabolic-vulnerability studies exploiting the STK11-deficient background, radiation strategies, and anti-metastatic evaluation.

Cell Line Information: A549 / A549-luc

A549's half-century of continuous use rests on its tractability in vitro and its sobering realism in vivo. The cells are adherent, spindle-shaped epithelial cells of 10–14 µm that grow readily in F-12K medium (Kaighn's modification) or DMEM with 10% fetal bovine serum, transfect efficiently, and retain a stable hypotriploid karyotype. Orthotopic tumors are poorly differentiated, PAS-positive for mucin secretion, and positive for the adenocarcinoma markers CK7 and TTF-1, with frequent mitoses, necrotic foci, and c-MET expression. The A549-luc derivative adds firefly luciferase for quantitative imaging, with monoclonal selection of high-signal clones in vitro recommended for uniform bioluminescence in vivo.

Our Services

Intrathoracic work is unforgiving of small errors — a millimeter of needle depth or a moment of hesitation decides whether a mouse carries a clean solitary nodule or a thoracic catastrophe. Alfa Cytology performs A549 orthotopic implantation with the published intercostal microinjection protocol, then carries each study through scheduled bioluminescence or spiral CT staging, body-weight sentinel monitoring, benchmark-validated dosing, and full necropsy with brain, liver, and mediastinal screening — a continuous chain of custody from cell bank to survival curve.

Workflow of A549 / A549-luc Orthotopic Mouse Model Construction

Construction of the A549 orthotopic model is a rapid, standardized microsurgical procedure whose parameters are tightly specified in the literature. The workflow below follows the validated intercostal technique, with monitoring and endpoint steps configured for luciferase or non-luciferase studies.

  1. Cell Expansion and Quality Control: A549 or A549-luc cells are expanded in vitro in F-12K or DMEM + 10% FBS to the logarithmic phase; batches are confirmed mycoplasma-negative, STR-authenticated, and precisely counted, with monoclonal luciferase expression verified for tagged lines before implantation.
  2. Host Selection and Acclimatization: BALB/c nude or NSG mice, typically 5–6 weeks of age, are acclimatized for approximately one week under SPF conditions and health-screened before entering the surgical cohort.
  3. Inoculum Preparation: Cells are washed in calcium- and magnesium-free PBS and resuspended at 5 × 107 cells/mL with 1 mg/mL Matrigel (20 µg per 20 µL aliquot), kept on ice so the suspension remains fluid for the brief procedure.
  4. Intercostal Implantation: Under isoflurane anesthesia, a ~5 mm incision is made in the left chest wall, and a fine needle is advanced approximately 3 mm through the intercostal space into the lung parenchyma, where 20 µL containing 106 cells is injected slowly; the needle is held briefly on withdrawal to limit back-leakage, and the whole procedure is completed in under one minute per animal, keeping operative mortality below 5%.
  5. Closure and Early Monitoring: The skin is closed with wound clips or sutures; animals recover on a warming platform and are weighed daily — a body-weight decline emerging around day 10 serves as the standard early sentinel of intrathoracic tumor growth.
  6. Tumor Staging and Randomization: A549-luc tumors are imaged weekly by bioluminescence in vivo, while non-luc configurations are staged by spiral CT or ultrasound; animals are randomized into treatment arms once tumors are established, typically between days 7 and 14.
  7. Treatment and Follow-Up: Test agents are dosed on study-defined schedules — with cisplatin or paclitaxel arms available as historical benchmarks — while body weight, respiratory signs, and imaging readouts are recorded until humane endpoints, at which untreated cohorts historically reach a median survival of approximately 31 days.
  8. Necropsy and Metastasis Screening: At termination, lungs are harvested with lobe-by-lobe surface nodule counts, and the contralateral lung, mediastinum, chest wall, brain, and liver are inspected macroscopically and microscopically; tissues are processed for H&E and CK7/TTF-1 immunohistochemistry confirming adenocarcinoma origin, ex vivo organ bioluminescence is recorded for luciferase studies, and survival, imaging, and pathology data are compiled into the final report.

Case Study-A549 / A549-luc Orthotopic Mouse Model Development

The A549 orthotopic system is among the most heavily benchmarked lung cancer models in the preclinical literature, and it has carried a wide range of Alfa Cytology client programs — platinum and taxane benchmarking, KRAS/STK11 metabolic-vulnerability studies, targeted and antibody-based agents, radiation combination strategies, and anti-metastatic designs evaluated against its documented brain and liver spread. Across these programs, readouts have combined longitudinal bioluminescence or CT kinetics in vivo, survival analysis, and systematic metastasis mapping at necropsy. Anonymized case summaries and representative deliverable formats are available on request.

Why Choose Alfa Cytology?

The A549 orthotopic model is deceptively simple on paper and genuinely demanding in the thorax. Alfa Cytology has operationalized the published microinjection standard and built its service around the reproducibility metrics that define this model.

  • A rigorously specified intercostal technique — 20 µL, 3 mm depth, under one minute per mouse — with operative mortality below 5% and take rates of 90–100% documented across independent laboratories.
  • A metastatic phenotype that includes bilateral intrapulmonary spread in 100% of animals, plus brain and liver lesions in a majority — a benchmark few orthotopic models can cite.
  • A therapeutically relevant genotype: KRAS G12S with homozygous STK11 loss on a TP53 wild-type background, mirroring a major NSCLC subgroup.
  • Flexible monitoring: quantitative bioluminescence in vivo for luciferase configurations, spiral CT volumetrics and ultrasound for non-luciferase studies, and body-weight sentinel tracking from day 10.
  • Benchmark-validated study designs, with cisplatin and paclitaxel reference arms available against published survival and metastasis data.
  • Systematic brain, liver, mediastinal, and chest-wall screening at necropsy, with CK7/TTF-1 histopathological confirmation.
