Hepa1-6 Syngeneic Model Service for Liver Cancer

Fig 1: Hepa1-6 Syngeneic Model for Liver Cancer preclinical research.

The Hepa1-6 syngeneic model is the most commonly used immunocompetent platform for hepatocellular carcinoma research, engrafting a hepatocyte-like BW7756-derived tumor line with authentic liver-function secretome into syngeneic hosts for immunotherapy, targeted-therapy, and gene-editing studies. Alfa Cytology delivers this benchmark model as a complete preclinical service — authenticated ATCC-line banking, dose-optimized subcutaneous and orthotopic left-lobe implantation, bioluminescence monitoring, and full hepatic histopathology — giving sponsors the field's standard-setting HCC system with the strain and dose discipline its literature demands.

Overview of Hepa1-6 Syngeneic Model for Liver Cancer

Hepa1-6 is the descendant of BW7756 — a hepatoma that arose spontaneously in a C57L mouse — and it was brought into modern laboratory use by Gladys Darlington, whose 1980 Journal of the National Cancer Institute paper documented liver-phenotype expression in cultured mouse hepatoma cells and whose 1987 Methods in Enzymology chapter established the line's canonical culture protocols. Deposited as ATCC CRL-1830, the line displays epithelial morphology, adherent growth, and — unusually among transplantable hepatomas — a living piece of liver physiology: it secretes albumin, alpha-fetoprotein, α1-antitrypsin, and amylase, expresses multiple cytochrome P450 enzymes, and retains an undifferentiated hepatocellular carcinoma phenotype that makes it a dual-purpose tool for oncology and hepatotoxicology alike. Tumorigenicity is thoroughly dose-mapped: in C57BL/6J hosts, subcutaneous inoculation at 5 × 105 cells produces only 10% takes, at 106 cells 20%, and at 2 × 106 cells 90%, with palpable tumors by day 7 at the highest dose and a tumor-bearing median survival of roughly 57 days — a steep dose–response that rewards careful inoculum standardization. Orthotopically, 5 × 106 luciferase-tagged cells in 20 μL implanted into the left liver lobe recreate the hepatic microenvironment, producing trabecular tumors with preserved sinusoidal structures.

Mechanistic and in vivo validation of the USP5-YTHDF1 axis in the Hepa1-6 syngeneic HCC model — USP5 stabilizes YTHDF1 through mTORC1-mediated phosphorylation (co-IP and ubiquitination assays), and in vivo panels show shUsp5 Hepa1-6 tumors with reduced growth, smaller volume/weight, suppressed USP5/YTHDF1 expression and improved anti-PD-L1 sensitivity with survival benefit. The model's immunology is as instructive as its biology. A landmark multiplex-imaging comparison of syngeneic HCC models found Hepa1-6 tumors to be the most immune-infiltrated of the panel — dense CD4+, CD8+, and FoxP3+ T-cell infiltrates, abundant F4/80+ macrophages of an M1-like character, low MDSC burden, and even tertiary lymphoid structures composed of B220+, CD4+, and CD8+ cells, the structures correlated with improved prognosis in human HCC. The functional consequence is striking: orthotopic Hepa1-6 tumors frequently clear even without checkpoint blockade, and anti-PD-1-treated and control cohorts alike can survive the full eight-week observation window — a robustness that is simultaneously the model's charm and its caution, since reviewers note its microenvironment is not fully representative of human HCC. Two design nuances deserve respect: the line is C57L/J-lineage, so C57BL/6 implantation — however common in the literature — carries a documented haplotype mismatch that can depress orthotopic take rates (3 of 7 in one careful study) and confound immune interpretation; and the standard flank dose of 2 × 106 cells should not be assumed interchangeable across protocols. Within those guardrails, Hepa1-6 has carried some of the decade's most cited liver-cancer studies — METTL3 loss driving sorafenib resistance through FOXO3-mediated autophagy, GDF15-targeting CRISPR nanocapsules reprogramming the immunosuppressive microenvironment of orthotopic disease, and the HDAC8-inhibitor-plus-anti-PD-L1 combination work that produced multi-month tumor-free survival — making it a versatile, deeply annotated workhorse for modern HCC research.

Our Services

Engaging Alfa Cytology for the Hepa1-6 syngeneic model places the field's reference HCC workflow under one accountable team: authenticated ATCC-provenance banks are maintained at low passage with the documented 1:2–1:6 split discipline, hosts are selected with deliberate strain logic (C57BL/6 by default, C57L/J where strict syngeneicity is required), subcutaneous inocula are standardized against the published dose–response curve, and orthotopic left-lobe implants — surgical or ultrasound-guided — are followed by scheduled bioluminescence imaging through treatment arms ranging from checkpoint inhibitors to gene-editing nanocarriers, with terminal hepatic histopathology and immune profiling delivered as an audit-ready data package.

Workflow of Hepa1-6 Syngeneic Model Construction

Hepa1-6 studies succeed or fail on two disciplines no other liver line demands quite so starkly: dose-standardized inocula and deliberate strain selection. Around those pillars, the subcutaneous and orthotopic formats follow the platform's published geometry.

  1. Cell preparation and quality control: Hepa1-6 or Hepa1-6-Luc cells are expanded at low passage in DMEM + 10% FBS with the documented split rhythm, confirmed mycoplasma-free, and luciferase activity validated in vitro; inocula are prepared against the published dose–response anchors (106 vs 2 × 106 flank).
  2. Host selection and acclimation: C57BL/6 mice (standard practice) or C57L/J mice (strict syngeneicity), typically 6–8 weeks of age, are sourced, quarantined, and acclimated in a specific-pathogen-free barrier facility, with the strain decision documented in the study record.
  3. Implantation: subcutaneous route. 2 × 106 cells in 0.1 mL sterile PBS are injected into the flank to exploit the 90% take tier; the site is compressed briefly on needle withdrawal, and palpable tumors are expected within about 7 days.
  4. Implantation: orthotopic liver route. Under isoflurane anesthesia with perioperative buprenorphine, a left subcostal laparotomy exposes the left lateral lobe, and 5 × 106 cells in 20 μL PBS are injected into the hepatic parenchyma; an ultrasound-guided variant reduces anesthesia and recovery time with comparable growth kinetics.
  5. Closure and postoperative care: The lobe is returned, the abdomen is closed in layers, and mice recover with 72-hour analgesic coverage and daily monitoring of weight and activity.
  6. Randomization and treatment: Flank studies are randomized at 150–250 mm3 (typically day 7–10) and orthotopic cohorts at a defined post-implantation interval; checkpoint inhibitors (e.g., anti-PD-1 at 200 μg i.p. three times weekly), targeted agents, or nanocarrier formats are dosed per protocol.
  7. Longitudinal monitoring: Tumors are measured by caliper on the protocol cadence; luciferase-tagged cohorts undergo bioluminescence imaging after intraperitoneal D-luciferin injection on IVIS-class systems, with ultrasound as an anatomical complement for orthotopic disease.
  8. Endpoint analysis and reporting: At termination, tumors and livers are resected and weighed; tissues undergo H&E with hepatic histology (trabecular pattern, sinusoidal preservation), immunohistochemistry (CD8, FoxP3, F4/80, B220, CD31, PD-L1 panels), flow-cytometric tumor-infiltrating-leukocyte profiling where specified, and metastatic organ screening, with all data consolidated into the final report.

Case Study-Hepa1-6 Syngeneic Model Development

Alfa Cytology has applied the Hepa1-6 syngeneic platform to preclinical programs spanning checkpoint inhibitor evaluation, epigenetic-drug combinations, targeted-therapy resistance mechanisms, and gene-editing nanocarrier studies in hepatocellular carcinoma. In these engagements, caliper and bioluminescence readouts tracked flank and orthotopic tumor kinetics, while terminal hepatic histopathology with immune-marker panels and flow-cytometric tumor-infiltrating-leukocyte profiling completed the pharmacological and immunological assessment. Study parameters — strain selection, inoculum tier, implantation route, and endpoint panels — are tailored to each program. Case materials can be shared under confidentiality agreements upon request.
