SK-Hep-1 / SK-Hep-1-luc Orthotopic Mouse Model Service for Endothelial Sarcoma

Fig 1: SK-Hep-1 / SK-Hep-1-luc Orthotopic Mouse Model for Endothelial Sarcoma preclinical research.

The SK-Hep-1 / SK-Hep-1-luc orthotopic mouse model for endothelial sarcoma transplants a human endothelial-origin tumor into its native hepatic environment, where it grows a vascularized, capillary-forming tumor that faithfully reports its burden through bioluminescence. Dedicated exclusively to preclinical oncology research, Alfa Cytology provides this model as a fully managed service, from authenticated SK-Hep-1 / SK-Hep-1-luc cell supply and donor-tumor expansion through orthotopic implantation into the liver, scheduled in vivo imaging, and endpoint vascular histopathology.

Overview of SK-Hep-1 / SK-Hep-1-luc Orthotopic Mouse Model for Endothelial Sarcoma

SK-Hep-1 is one of the most instructive case studies in cell line identity. Established in 1971 at Memorial Sloan Kettering from the ascitic fluid of a 52-year-old male patient with adenocarcinoma of the liver, the line was long used as a hepatocellular carcinoma model until ultrastructural and molecular evidence demonstrated an endothelial origin: cells carry Weibel-Palade bodies, form tubules on extracellular matrix, express endothelial nitric oxide synthase and von Willebrand factor, and display fenestrae lacking diaphragms—the signature of liver sinusoidal endothelium. Subsequent work revealed an additional layer of biology, showing that SK-Hep-1 cells bear a full mesenchymal stem cell marker set, can differentiate toward adipogenic and osteogenic lineages, and behave as oncogenic stem-like cells capable of tumor initiation from a single cell.

Fig 2: Reference figures for SK-Hep-1 / SK-Hep-1-luc cell-related literature.Fig 1. Orthotopic xenograft experiment in nude mice showing that luciferase-labeled SK-HEP-1 tumors with HOMER3-AS1 overexpression grew significantly larger (higher liver flux by bioluminescence imaging) while HOMER3-AS1 knockdown suppressed SK-HEP-1 orthotopic tumor growth, accompanied by corresponding Ki-67, cleaved caspase-3 and F4/80 immunostaining changes. (Pu, Jian, et al., 2021)

This unusual biology makes SK-Hep-1 a natural model for endothelial malignancies such as angiosarcoma and endothelial sarcoma, and it has been widely adopted for that purpose. In nude mice the cells form tumors with remarkable reliability—reported take rates of 100% after subcutaneous injection of 5 × 106 cells—and the engrafted tissue recapitulates the line’s vascular nature, assembling capillary-like structures that stain positive for CD31. For orthotopic work, luciferase-tagged SK-Hep-1 (SK-Hep-1-luc) cells or fragments expanded as subcutaneous donor tumors are transplanted into the liver, the tissue of the line’s original isolation, where primary tumor growth and spread can be tracked by whole-animal bioluminescence; the same labeling strategy has proven powerful in metastasis research, in which SK-Hep-1-luc dissemination to lung, kidney, liver, muscle, and skin has been documented. Figure 1 summarizes how this orthotopic endothelial sarcoma platform is assembled and monitored.

Cell Line Information: SK-Hep-1 / SK-Hep-1-luc

The SK-Hep-1 line has been scrutinized by multiple laboratories over five decades, leaving it unusually well characterized for an older line. The defining attributes of the parent line and its luciferase-tagged derivative are summarized below.

Our Services

An SK-Hep-1 / SK-Hep-1-luc engagement with Alfa Cytology is handled as one continuous program: STR-verified cell banking, donor tumor expansion, meticulous fragment-based orthotopic implantation into the liver, imaging visits on a pre-agreed cadence, and a terminal package that links photon-flux curves to CD31-based vascular histopathology.

Workflow of SK-Hep-1 / SK-Hep-1-luc Orthotopic Mouse Model Construction

Building an SK-Hep-1 or SK-Hep-1-luc orthotopic endothelial sarcoma model typically follows a fragment-based strategy in which a luciferase-labeled subcutaneous donor tumor is expanded first and then grafted into the liver. The workflow below describes this standard execution path from cell bank to data package.

  1. Cell Bank Verification and Expansion: SK-Hep-1 or SK-Hep-1-luc stocks are authenticated by STR profiling and mycoplasma testing, then expanded in EMEM-based medium to the required scale.
  2. Luciferase Calibration (SK-Hep-1-luc Only): In vitro signal output per cell is verified so that in vivo photon flux can be converted into relative tumor burden.
  3. Donor Tumor Expansion: A pilot cohort of nude mice receives a subcutaneous inoculum (commonly 5 × 106 cells in a minimal-volume carrier) to grow donor tumors of defined size; 100% engraftment is expected within about two weeks.
  4. Donor Tumor Excision and Fragment Preparation: Donor tumors are aseptically resected, necrotic tissue is removed, and uniform tumor fragments or single-cell suspensions are prepared for orthotopic transfer.
  5. Laparotomy and Liver Exposure: Under aseptic conditions the abdominal cavity is opened and the target hepatic lobe is gently exteriorized for graft placement.
  6. Orthotopic Implantation: Tumor fragments are secured to the liver surface with sutures or absorbable adhesive, or a cell suspension is injected slowly into the parenchyma; the graft site is inspected to confirm secure placement before closure.
  7. Closure and Longitudinal Bioluminescence Monitoring: The abdominal wall and skin are sutured, animals recover under warming with daily health checks, and whole-animal imaging after intraperitoneal D-luciferin is performed on a scheduled basis to chart primary tumor growth and flag dissemination.
  8. Endpoint Necropsy and Vascular Histopathology: At humane endpoint the liver and distant organs are resected for ex vivo imaging and weighing; tissues are processed for H&E and immunohistochemistry including CD31 and vWF to document capillary-like structures, and metastatic foci are mapped.
