Colon 38 Syngeneic Model Service for Colon Cancer

Fig 1: Colon 38 Syngeneic Model for Colon Cancer preclinical research.

The Colon 38 syngeneic model service for colon cancer is an immune-competent C57BL/6 platform built on a differentiated, NMU-induced colon adenocarcinoma that uniquely combines slow, treatment-window-friendly primary growth with a high-incidence, cachexia-linked hepatic metastatic cascade after orthotopic grafting. Alfa Cytology provides this model as a complete preclinical capability — covering authenticated Colon 38 / luciferase-engineered Colon 38 expansion and banking, subcutaneous or orthotopic cecal implantation, scheduled tumor and metastasis monitoring, and full histopathology with survival-based endpoints — all configured around each sponsor’s regimen, schedule, and decision timeline.

Overview of Colon 38 Syngeneic Model for Colon Cancer

Colon 38 is the deliberate counterpart to Colon 26 within the landmark NCI panel of transplantable colon tumors created by Corbett and colleagues in 1975 through rectal application of the carcinogen N-nitroso-N-methylurethane (NMU) — and the pairing of the two lines remains one of the most instructive contrasts in preclinical oncology. Where Colon 26 is a grade IV, undifferentiated, blisteringly fast BALB/c tumor, Colon 38 is a C57BL/6 adenocarcinoma of differentiated glandular histology that grows considerably more slowly, with in vivo tumor-volume doubling times measured in days rather than hours. That slower tempo is a feature, not a bug: it opens treatment windows long enough to model adjuvant-style and modulator-based regimens that the racing Colon 26 cannot accommodate, and it made Colon 38 the workhorse of the 5-FU modulation era — the line in which the intratumoral “trapping” of 5-FU was visualized by in vivo 19F nuclear magnetic resonance, in which dihydropyrimidine dehydrogenase inhibition by 5-ethynuracil and DHODH inhibition by brequinar were shown to potentiate 5-FU, and in which interferon–fluoropyrimidine synergy was first characterized at the cytokinetic level.

Temporal evolution of the Colon 38 (MC38) syngeneic tumor microenvironment from day 7 to day 21 — T-cell infiltration gives way to T-cell exclusion by dense desmoplastic stroma, with progressive changes in vascularity, necrosis, T-cell exhaustion and regulatory T cells. (Shields, Nicholas J., et al., 2023)

The model’s most distinctive capability emerges with orthotopic placement. When small intact fragments of Colon 38 tumor are transplanted to the cecum of syngeneic C57BL/6 mice, hepatic metastasis arises with high incidence within about one month, and untreated animals succumb to cachexia at roughly two months — a reproducible timeline that has served anti-metastatic screening for decades. In the benchmark E7010 study, delayed administration reduced hepatic metastases from 17–41 per animal in controls to 2–3 in the treated group and extended median survival from approximately 50 to 63 days, demonstrating the model’s power to separate anti-metastatic from merely cytostatic activity. Contemporary practice has extended the line with luciferase engineering for bioluminescent burden tracking in radiation-response studies. Together, Colon 38’s differentiated histology, generous treatment window, C57BL/6 immune competence, and liver-metastatic orthotopic variant make it a complementary pillar to faster colorectal lines for chemotherapy modulation, interferon-era combination logic, and modern anti-metastatic evaluation in vivo.

Cell Line Information: Colon 38

The table below consolidates the key biological, cultural, and modeling characteristics of the Colon 38 line, including its luciferase-engineered derivatives.

Parameter Details
Cell line designation Colon 38 (also written Colon-38, Colon38); luciferase-engineered derivatives generated by stable transfection with clonal selection are documented in contemporary radiation-response studies
Species / genetic background Mouse (Mus musculus), C57BL/6 inbred background; syngeneic in C57BL/6 immunocompetent hosts — the classic C57BL/6 counterpart to the BALB/c Colon 26 line
Origin Induced by N-nitroso-N-methylurethane (NMU) administered rectally to C57BL/6 mice within the transplantable colon tumor panel developed by Corbett et al. (Cancer Research 35:2434–2439, 1975)
Depository / catalogue Distributed by ATCC (murine Colon-38, C57BL/6); carried in the classical NCI-DCTD transplantable tumor panel
Tumor type Murine colon adenocarcinoma with differentiated glandular histology (in contrast to the undifferentiated grade IV Colon 26)
Growth properties Adherent epithelial cells; markedly slower growth than Colon 26, with in vivo tumor-volume doubling times in the multi-day range, providing extended treatment windows for modulator and adjuvant-style regimens
Recommended culture conditions RPMI-1640 supplemented with fetal bovine serum at 37 °C in a humidified 5% CO₂ atmosphere (as used across the classical Colon 38 literature)
Standard implantation practice Subcutaneous implantation of approximately 1 × 106 cells for flank efficacy studies; orthotopic transplantation of small intact tumor fragments to the cecal wall for the hepatic-metastatic variant (metastases arise with high incidence within about one month, with cachectic death near two months in untreated controls)
Documented in vivo behavior 5-FU accumulates in Colon 38 tumors with a concentration-dependent “trapping effect” visualized by in vivo 19F NMR; EU-mediated DPD inhibition increases intratumoral 5-FU nucleotide formation 2–3-fold; delayed E7010 dosing reduced hepatic metastases from 17–41 to 2–3 per animal and extended median survival from ≈50 to ≈63 days
Documented in vitro behavior 5-FU inhibits growth by 28–40% at standard concentrations; interferon shows antiproliferative activity with synergistic interactions against halogenated pyrimidines; adipokine (leptin) exposure modulates proliferation and fluorouracil response
Host strains C57BL/6 immunocompetent mice (syngeneic)
Key research applications 5-FU modulator and biochemical-potentiation studies (DPD/DHODH inhibition, uridine and leucovorin modulation, interferon combinations); orthotopic anti-metastatic screening with hepatic endpoint; adjuvant-style and fractionated regimens benefiting from the long treatment window; radiation-response and responder/non-responder biology using luciferase-engineered derivatives

Our Services

Colon 38 rewards patience and precision in equal measure, and Alfa Cytology runs the model accordingly: authenticated banks of the differentiated C57BL/6 line, orthotopic fragment grafting with high-incidence hepatic metastasis, scheduled primary and metastatic monitoring, and survival-based endpoint analysis — all delivered as one accountable preclinical program.

Workflow of Colon 38 Syngeneic Model Construction

Construction follows the classical methodology for this line’s two deployment formats: Colon 38 cells are expanded under authenticated conditions for flank studies, while orthotopic studies graft small intact tumor fragments to the cecal wall; both are followed through treatment with primary-tumor, metastatic, and survival endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: Colon 38 cells are identity-verified, confirmed mycoplasma-negative, and expanded into low-passage working banks under standardized RPMI-1640 culture conditions.
  2. Cell harvest or fragment preparation: For flank studies, healthy cultures are harvested, counted and viability-checked, then resuspended at approximately 1 × 106 cells in 100 μL PBS; for orthotopic studies, donor tumors are aseptically excised and minced into standardized fragments for cecal-wall grafting.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane; cells are injected subcutaneously, or fragments are grafted into the cecal wall through a midline abdominal approach with the cecum briefly exteriorized.
  4. Establishment monitoring: Flank tumors are measured by caliper from first palpability; orthotopic burden is monitored by scheduled palpation, weight tracking, and in vivo imaging where luciferase-engineered cells are used.
  5. Treatment administration: Experimental arms — 5-FU alone and with modulators (DPD or DHODH inhibitors, leucovorin), interferon combinations, targeted agents such as E7010-class sulfonamides, and modern immunotherapy — proceed per study design with body-weight and toxicity monitoring.
  6. Metastasis and survival follow-up: Orthotopic cohorts are followed for hepatic metastatic emergence (high incidence within about one month) and survival, with interim imaging where available; necropsy documents metastatic distribution at endpoint.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth curves, metastatic counts in liver and other organs, and survival with histopathology (H&E confirming differentiated adenocarcinoma histology, plus Ki67 and marker-specific immunohistochemistry) and body-composition/cachesia assessment where relevant.
