LP-1 / LP-1-luc Orthotopic Mouse Model Service for Multiple Myeloma (MM)

Fig 1: LP-1 / LP-1-luc Orthotopic Mouse Model for Multiple Myeloma (MM) preclinical research.

The LP-1 / LP-1-luc orthotopic mouse model of multiple myeloma engrafts an IgG-secreting, TP53-mutant human myeloma line into the bone marrow compartment, where tumor growth in the marrow microenvironment is quantified by luciferase bioluminescence and corroborated by serum paraprotein tracking. Alfa Cytology delivers this platform as an integrated preclinical service: authenticated ACC 41 banking, luciferase line qualification, intra-tibial or intravenous engraftment in NOD/SCID or NSG recipients, scheduled whole-body imaging with serum IgG quantification, proteasome-inhibitor and alkylator reference dosing, and endpoints spanning bone-disease imaging, CD138-based marrow analysis, and histopathology.

Overview of LP-1 / LP-1-luc Orthotopic Mouse Model for Multiple Myeloma (MM)

LP-1 is a human multiple myeloma cell line established in 1986 from the peripheral blood of a 56-year-old woman whose IgG-secreting, EBV-negative myeloma had transformed into a refractory, terminal leukemic phase — a leukemic origin that makes LP-1 one of the few myeloma lines genuinely comfortable in suspension and in circulation. Genomically it carries a well-documented TP53 mutation (cataloged in COSMIC and the IARC p53 database) and notably lacks the t(4;14) translocation and FGFR3 overexpression that define lines like KMS-11, OPM-2, and NCI-H929, placing LP-1 in the non-FGFR3 myeloma molecular compartment. The German DSMZ distributes the line as ACC 41 with matched exome and RNA-sequencing data publicly available, and its cells grow in Iscove’s MDM with a 35-80 hour doubling time that varies with culture density — a quirk that rewards careful density management and makes LP-1 a practical teaching line for marrow-mimicking suspension culture.

Integrated study showing that the RBM39 molecular glue degrader indisulam induced apoptosis in H929, MM.1S and LP-1 myeloma cells in vitro (panel A, LP-1 apoptotic fraction rising dose-dependently), and established two in vivo models—a subcutaneous H929 xenograft and a disseminated model by injecting luciferase-labeled LP-1 cells into NOG mice via tail vein—demonstrating that RBM39-MEK5 axis inhibition synergizes with bortezomib. (Liu, Jia, et al., 2025)

In vivo, LP-1 joins the established human myeloma xenograft repertoire, and the orthotopic reading of myeloma is literal: the bone marrow is the disease’s home. The modern luciferase-orthotopic paradigm was validated with intra-tibial implantation of luciferase-tagged myeloma cells into NOD/SCIDγcnull mice — engraftment detectable by week 3, a significant bioluminescence rise between weeks 5 and 7, concordant paraprotein elevation, marrow-confined disease confirmed by CD138 flow cytometry and histology, and spinal progression ending in hindlimb paralysis, with bortezomib and melphalan validating the system as a preclinical assay. Luciferase-tagged LP-1 adapts this architecture directly: intra-tibial delivery seats the tumor in the marrow niche, intravenous delivery models systemic leukemic-phase disease, and the line’s IgG secretion supplies a natural serum biomarker running in parallel with imaging. For microenvironment purists, the SCID-hu tradition — implanting human bone subcutaneously and injecting tumor into it — and scaffold-based variants extend the same question. The LP-1-luc orthotopic platform thus serves proteasome-inhibitor and alkylator benchmarking, marrow-interaction and bone-disease studies, immunomodulatory-agent evaluation, and imaging-method development, with a TP53-mutant, non-FGFR3 genotype that broadens the molecular coverage of any myeloma program.

Our Services

Bone-marrow orthotopic work is a craft: the surgery is micro-scale, the disease is marrow-confined, and the difference between a clean cohort and a lost one is often a single well-handled tibia. Alfa Cytology brings that craft to LP-1-luc studies — authenticated ACC 41 stocks grown with the density discipline the line demands, reporter lots released only after uniform in vitro luminescence, and intra-tibial or intravenous engraftment executed under written technique with serum IgG baselines recorded in parallel. Imaging calendars follow the published engraftment tempo, bortezomib-class reference arms anchor efficacy claims, and CD138 flow cytometry closes every study with marrow-level truth. From first baseline scan to final histology, Alfa Cytology keeps the LP-1 / LP-1-luc orthotopic workflow under single-point control.

Workflow of LP-1 / LP-1-luc Orthotopic Mouse Model Construction

The LP-1-luc orthotopic workflow literalizes “orthotopic” for myeloma: the marrow cavity is the disease site, and the study spine combines imaging, paraprotein, and marrow histology. The sequence below is the standard construction-to-analysis flow.

  1. Authentication and reporter release: LP-1 / LP-1-luc stocks are expanded from authenticated ACC 41 master banks at controlled density, mycoplasma-tested, and released only after uniform firefly luciferase expression is confirmed by serial in vitro luminescence assay.
  2. Engraftment cell preparation: Suspension cultures maintained at 0.5-1.0×106 cells/mL are harvested in log phase, washed, counted, and resuspended in sterile PBS at the challenge dose (commonly 2.5×106 to 5×106 cells in 20-50 µL for intra-tibial delivery; about 5×106 cells in 100 µL for intravenous designs, calibrated during pilot runs).
  3. Recipient preparation: NOD/SCID or NSG mice are acclimatized, body-weight stratified, and where protocols require, preconditioned with sublethal irradiation; baseline serum is collected for IgG paraprotein measurement.
  4. Marrow-cavity engraftment: Under anesthesia with aseptic technique, the cell suspension is microinjected through the cortical surface into the tibial (or femoral) marrow cavity, or delivered via the lateral tail vein for systemic designs; baseline bioluminescence after D-luciferin confirms deposition.
  5. Longitudinal monitoring: On a fixed schedule, in vivo whole-body imaging tracks marrow signal and spread to pelvis, contralateral limbs, and spine, while serial serum draws quantify IgG paraprotein; body weight and clinical signs are scored, with hindlimb function watched as a myeloma-specific endpoint.
  6. Randomized treatment: Once signals establish, cohorts are randomized by matched photon flux and paraprotein level; bortezomib, melphalan, or test agents are dosed per protocol, with reference arms aligned to the published intra-tibial paradigm.
  7. Endpoint and marrow analysis: At termination (or humane endpoint), injected and contralateral tibiae, femora, spine, spleen, and viscera are collected; ex vivo imaging localizes deposits; human CD138 flow cytometry quantifies marrow chimerism; micro-CT or histomorphometry assesses bone lesions; and H&E with CD138 immunostaining documents plasma-cell infiltration and loss of marrow architecture.

Case Study-LP-1 / LP-1-luc Orthotopic Mouse Model Development

Alfa Cytology has implemented the LP-1 orthotopic platform in preclinical programs spanning proteasome-inhibitor benchmarking, alkylator combination strategies, and marrow-interaction studies. In these programs, scheduled bioluminescence sessions tracked marrow-confined tumor growth alongside serum IgG paraprotein curves, randomized arms started from matched signal and paraprotein baselines, and terminal analyses combined ex vivo bone imaging, human CD138 flow-cytometric marrow quantification, and histological assessment of bone lesions. Where designs required microenvironment context, SCID-hu-style bone-implant or scaffold variants were incorporated. Program-specific parameters and outcomes are retained as sponsor-confidential deliverables, and representative outlines can be furnished upon request.
