KPC (derived) Syngeneic Model Service for Pancreas Cancer

Fig 1: KPC (derived) Syngeneic Model for Pancreas Cancer preclinical research.

The KPC (derived) syngeneic model of pancreas cancer transplants tumor cells carrying the endogenous KrasG12D and Trp53R172H drivers into immunocompetent C57BL/6 hosts, combining human-relevant PDAC genetics with the control of an implantable system. Alfa Cytology delivers the complete KPC-derived workflow—genotype-verified cell banking, subcutaneous, orthotopic, fragment or intrasplenic implantation, balanced randomization, scheduled dosing and stroma- and metastasis-inclusive endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of KPC (derived) Syngeneic Model for Pancreas Cancer

The KPC lineage begins with a genetically engineered mouse that changed pancreatic cancer research: Pdx1-Cre; LSL-KrasG12D; LSL-Trp53R172H animals (Hingorani and colleagues, 2005) develop pancreatic intraepithelial neoplasia that progresses to invasive, metastatic PDAC with dense desmoplasia and effector-T-cell exclusion—a genetic pairing that mirrors human disease, where KRAS is mutated in the overwhelming majority and TP53 in roughly seventy percent of cases. The murine Trp53 R172H allele corresponds to the human R175H hotspot. Because spontaneous disease in the GEMM is powerful but variable in latency, the field distilled it into transplantable derivatives—cell lines such as FC1245 and FC1199 (backcrossed to C57BL/6 over ten generations), 7940B, MT-5 and luciferase-tagged variants—whose Kras and Trp53 recombined alleles are confirmed by genotyping PCR. The cells grow as loosely adherent, polygonal epithelial monolayers in DMEM with 10% serum, with published practice favoring early passages (six to twelve) and mycoplasma-negative certification.

Orthotopic bioluminescence imaging and tumor growth comparison of parental KPC versus KPC-Luc cells, revealing that firefly luciferase expression elicits an anti-tumor immune response—smaller or absent tumors with NK cell accumulation and increased CD8+ T-cell infiltration—an important caveat for luciferase-engineered syngeneic PDAC models. (Ferrari, Daniele Pereira, et al., 2024)

What distinguishes KPC-derived tumors is their microenvironment: abundant F4/80-positive macrophages, sparse CD3-positive T cells, and collagen-rich desmoplasia—though direct comparisons show that cell-line implants carry substantially less stroma than the parent GEMM, and that three-millimeter tumor-fragment orthotopic implants preserve the desmoplastic, leukocyte-rich architecture most faithfully. Format options map to questions: subcutaneous implants for routine efficacy; orthotopic parenchymal injection (one hundred thousand cells) where organ context matters, with aggressive courses reaching median survival near twenty-three days (KPC-Luc) versus thirty-six days (MT-5); fragment implants for stroma-question work; and intrasplenic delivery of half a million luciferase-tagged cells for liver-metastasis imaging at three weeks. The model has carried mechanistic breakthroughs—CD40 agonism reprogramming macrophages to degrade fibrosis and enhance gemcitabine efficacy, PIK3CA loss rendering tumors immunogenic through MHC-I and CD80 upregulation, and DHODH-inhibitor sensitivity mapping. Caveats: match the strain to the line’s documented background (some derivatives such as PDA530Met sit on mixed 129/C57BL stock)—and accept that cell-line formats trade away some desmoplasia, so stroma-centric hypotheses may demand fragment or GEMM-context designs.

Cell Line Information: KPC (derived)

Key characteristics of KPC-derived lines and their formats are summarized below.

Item Details
Cell line name KPC-derived PDAC lines; documented examples: FC1245 and FC1199 (C57BL/6, backcrossed, David Tuveson laboratory), 7940B (C57BL/6, Gregory Beatty laboratory), MT-5 (Trp53-null variant), KPC-Luc (luciferase), UN-KPC-960/961, PDA530Met (liver-metastasis derived)
Species / strain background Mus musculus; syngeneic host: C57BL/6J for backcrossed lines (F10 documented); mixed 129/SvJae × C57BL/6 backgrounds exist for some derivatives — strain matching per line is essential
GEMM origin Pdx1-Cre; LSL-KrasG12D; LSL-Trp53R172H genetically engineered model (Hingorani et al., Cancer Cell, 2005); PanIN → invasive, metastatic PDAC; GEMM metastases in ~80% of animals, predominantly liver and lung
Human relevance KRAS mutated in ~80–95% and TP53 in ~70% of human PDAC; murine Trp53 R172H corresponds to the human TP53 R175H hotspot; recombined alleles verified by genotyping PCR
Morphology / culture Loosely adherent, polygonal epithelial monolayer; DMEM + 10% FBS + penicillin/streptomycin (some stocks with 83 µg/mL gentamicin); early passages (6–12) favored; split 1:2 every 2 days; 37 °C, 5% CO2
Tumor microenvironment Dense desmoplastic stroma, abundant F4/80+ macrophages, scarce CD3+ T cells with effector exclusion; cell-line implants carry less stroma than fragment implants or the GEMM — 3 mm fragment orthotopic implants preserve architecture most faithfully
Model formats Subcutaneous (1 × 106 cells); orthotopic pancreas parenchyma (1 × 105 cells; median survival ~23 days for KPC-Luc, ~36 days for MT-5); 3 mm tumor-fragment orthotopic (stroma-richest); intrasplenic 5 × 105 cells for liver-metastasis imaging at 3 weeks
Mechanistic precedents CD40 agonist reprograms macrophages to degrade fibrosis and enhance gemcitabine efficacy (7940B lineage, Cancer Discovery 2016); PIK3CA loss upregulates MHC-I and CD80, rendering orthotopic tumors susceptible to immunosurveillance (FC1245 CRISPR); DHODH inhibitor brequinar sensitivity mapped across FC1245/FC1199
Considerations Match host strain to the line’s documented background; cell-line formats under-represent desmoplasia (use fragments for stroma questions); GEMM-level variability is traded for implant control; orthotopic courses are aggressive and compress windows
Representative applications KRAS-G12D dependency and targeted-agent studies, desmoplasia and stroma-targeting programs, CD40/checkpoint agonist combinations, liver-metastasis formats, biomarker and TME immunophenotyping

Workflow of KPC (derived) Syngeneic Model Construction

Construction of the KPC-derived syngeneic model is an implantation workflow whose quality rests on genotype-verified, early-passage cells, strain-matched recipients, format-matched technique and a disciplined measurement cadence. A standard engagement proceeds as follows:

  1. Source and qualify cells: KPC-derived stocks are cleared by mycoplasma testing, genotyped by PCR for the recombined KrasG12D and Trp53R172H alleles, kept within early passages (6–12 per published practice), and luciferase expression is titrated in vitro where imaging readouts are required.
  2. Prepare recipients: C57BL/6J mice (typically 6–8 weeks old; strain matched to the line’s documented background) complete acclimation and health screening and are individually marked before implantation.
  3. Prepare the inoculum: log-phase cells are harvested, washed, counted and resuspended in PBS (1 × 106 cells in 100 µL for flank implants; 1 × 105 cells in a small volume for orthotopic parenchymal injection; 5 × 105 cells for intrasplenic delivery); 3 mm fragments are trimmed fresh for stroma-preserving formats.
  4. Implant: cells are injected subcutaneously into the flank, into the pancreatic parenchyma under anesthesia with perioperative analgesia, into the spleen for the liver-metastasis format, or as fragments into the pancreas, with aseptic technique throughout.
  5. Randomize: flank-format animals are grouped when tumors reach palpability, balanced by tumor volume and body weight; orthotopic, fragment and intrasplenic cohorts are randomized at implantation or at the first staging scan.
  6. Dose and follow: test articles are administered on schedule; flank tumors are measured with calipers two to three times weekly, while orthotopic and metastatic cohorts are followed by body weight and weekly bioluminescence imaging; survival is recorded for orthotopic designs.
  7. Terminate and analyze: tumors are resected and weighed; liver, lung and peritoneum are inspected for metastasis; desmoplasia is scored by trichrome staining; CD3, CD8 and F4/80 immunohistochemistry and flow cytometry profile the infiltrate; tissues enter histopathology and molecular analysis.
