PLC/PRF/5 / PLC/PRF/5-luc Orthotopic Mouse Model Service for Liver Cancer

Fig 1: PLC/PRF/5 / PLC/PRF/5-luc Orthotopic Mouse Model for Liver Cancer preclinical research.

The PLC/PRF/5 / PLC/PRF/5-luc orthotopic mouse model grows the Alexander hepatoma — a human HCC line that permanently carries integrated hepatitis B virus DNA — directly within the liver parenchyma of immunodeficient mice, coupling authentic human tumor biology with the viral oncogenesis signature that drives most liver cancers worldwide. Alfa Cytology delivers this virologically unique platform with standardized intrahepatic implantation, luciferase-based tumor tracking in vivo, quantitative serum HBsAg monitoring as a built-in disease biomarker, and metastasis-oriented endpoint analysis, giving HBV-related HCC programs a model that no syngeneic line can replicate.

Overview of PLC/PRF/5 / PLC/PRF/5-luc Orthotopic Mouse Model for Liver Cancer

PLC/PRF/5, universally known as the Alexander cell line, was established in South Africa in 1976 from the primary tumor of a 24-year-old man with hepatocellular carcinoma. Its defining feature is virological: the genome harbors multiple integrated copies of hepatitis B virus DNA, and the cells secrete hepatitis B surface antigen (HBsAg) — at roughly 500 ng per day per 106 cells in vitro — without producing Dane particles, recapitulating the integrated-HBV state of virus-associated human HCC. The line is epithelial and adherent, AFP-positive but albumin-negative, retains functional p53, and carries a modal chromosome number near 55. In immunodeficient hosts it is reliably tumorigenic: after subcutaneous injection into nude mice, 96% of animals develop tumors within twelve weeks, and spontaneous lung metastases have been documented and even exploited to derive the more aggressively metastatic NL/PLC subline.

Placed orthotopically into the liver, PLC/PRF/5 forms intrahepatic tumors with moderate invasiveness, making it a well-regarded system for modeling intrahepatic metastasis — the pattern that most often dictates prognosis in HCC patients. Subcapsular inoculation of the left lobe reproducibly establishes tumors in a high fraction of recipients, and the resulting disease can be followed non-invasively in two independent ways: by bioluminescence imaging in vivo with the PLC/PRF/5-luc variant, and by serial serum HBsAg quantification, since the tumor itself manufactures a circulating antigen whose levels track disease burden. This dual-readout design, combined with the line's documented use in drug-resistance, cytokine-biology, and targeted-therapy research, makes the orthotopic PLC/PRF/5 platform a distinctive choice for programs that need human HCC biology with an authentic HBV-integration footprint.

Cell Line Information: PLC/PRF/5 / PLC/PRF/5-luc

The Alexander line remains one of the most cited human hepatoma models, prized for pairing straightforward culture in vitro with authentic HBV biology. Cells grow as an adherent epithelial monolayer in DMEM with 10% fetal bovine serum, expand quickly, and preserve their HBsAg-secreting phenotype across passages. Orthotopically, the line shows moderate invasiveness with a documented capacity for spontaneous pulmonary spread in vivo — an unusual combination among widely available HCC lines. The PLC/PRF/5-luc derivative adds firefly luciferase for quantitative imaging, with monoclonal selection of high-signal clones in vitro recommended to keep bioluminescence uniform in vivo.

Our Services

What distinguishes an Alfa Cytology PLC/PRF/5 study is the pairing of orthogonal readouts: bioluminescence curves from the luciferase-tagged tumor and serum HBsAg titers from the same animals, validated against caliper and histology at endpoint. Our team handles authenticated cell banking, subcapsular liver implantation, scheduled imaging and blood sampling, and full necropsy with lung-metastasis screening — a coherent workflow that keeps your virology and oncology questions answered from a single cohort.

Workflow of PLC/PRF/5 / PLC/PRF/5-luc Orthotopic Mouse Model Construction

Orthotopic implantation of PLC/PRF/5 follows the established subcapsular technique for human HCC xenografts, with monitoring strengthened by the line's dual readouts. The workflow below details construction and study execution from authenticated cell preparation through metastasis screening.

  1. Cell Expansion and Quality Control: PLC/PRF/5 or PLC/PRF/5-luc cells are expanded in vitro in DMEM + 10% FBS to the logarithmic phase; batches are confirmed mycoplasma-negative, STR-authenticated, and precisely counted, with HBsAg secretion and monoclonal luciferase expression verified for the tagged variant before implantation.
  2. Host Selection and Acclimatization: BALB/c nude, NOD-SCID, or NSG mice, typically 5–7 weeks of age, are acclimatized for approximately one week under SPF conditions and health-screened before entering the surgical cohort.
  3. Anesthesia and Liver Exposure: Animals are anesthetized with isoflurane or ketamine-based regimens; a subxiphoid or left subcostal incision exposes the left hepatic lobe, which is gently exteriorized onto moistened gauze.
  4. Subcapsular Implantation: A suspension of 1–3 × 106 cells in 20–50 µL PBS or PBS:Matrigel (1:1) is injected slowly beneath the hepatic capsule with the needle held parallel to the surface and withdrawn after a brief pause, the entry site compressed to prevent bleeding and leakage; where uniform solitary nodules are preferred, ~1 mm3 fragments prepared from donor subcutaneous tumors are implanted into a subcapsular pocket and secured with tissue adhesive.
  5. Closure and Recovery: The abdominal wall and skin are closed with 4–0 absorbable sutures; animals recover on a warming platform with analgesia per IACUC protocol and are monitored daily until fully ambulatory.
  6. Longitudinal Monitoring with Dual Readouts: Tumor growth is tracked weekly by bioluminescence imaging in vivo after D-luciferin administration, complemented by small-volume retro-orbital or tail-vein blood collection for serum HBsAg quantification at defined time points, with body weight and clinical condition recorded throughout.
  7. Randomization and Treatment: Once tumors reach the predefined bioluminescence or antigen threshold, animals are randomized into vehicle and treatment arms, and test agents — cytotoxics, targeted inhibitors, anti-angiogenics, or virologically active compounds — are dosed on study-defined schedules.
  8. Endpoint Analysis and Reporting: At termination, the primary liver tumor is excised, measured, and weighed; lungs are inflated, sectioned, and screened macroscopically and microscopically for metastatic nodules; tissues are processed for H&E, AFP, and HBsAg immunohistochemistry, serum antigen titers are finalized, ex vivo organ bioluminescence is recorded, and all imaging, biomarker, and pathology data are compiled into the final report.
