9464D Syngeneic Model Service for Neuroblastoma

Fig 1: 9464D Syngeneic Model for Neuroblastoma preclinical research.

The 9464D syngeneic model service for neuroblastoma is an immune-competent C57BL/6 platform built on the transplantable TH-MYCN tumor line — a MYCN-driven, GD2-expressing, MHC-I-low neuroblastoma that reproduces the immunologically cold phenotype and NK-cell-dependent surveillance of human high-risk disease. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated 9464D / 9464D-GD2 / reporter-engineered expansion and banking, subcutaneous, intra-adrenal, or experimental-metastatic implantation, scheduled caliper and in vivo bioluminescence monitoring, and full neuroblastoma histopathology with GD2 and checkpoint immunophenotyping — all configured around each sponsor’s anti-GD2 construct, checkpoint combination, radio-immunotherapy, or microenvironment hypothesis.

Overview of 9464D Syngeneic Model for Neuroblastoma

9464D descends from the most genetically faithful mouse model of human neuroblastoma: the TH-MYCN transgenic mouse developed by William Weiss’s laboratory at UCSF, in which tyrosine-hydroxylase-driven MYCN overexpression drives tumors that closely mirror the histology, gene expression, and molecular signature of human high-risk disease. From a spontaneous tumor of this model, Hansford and colleagues established the transplantable line 9464D in 2004, and its defining biology maps almost point-for-point onto the human disease: it is MYCN-driven, carries a low tumor mutation burden, endogenously expresses the neuroblastoma antigen GD2 (though at lower surface levels than many human lines), and displays low MHC class I expression with only about a quarter of cells capable of upregulating it under interferon-γ — the molecular signature of the “cold” high-risk tumors that resist T-cell attack in patients. The immunological characterization published in 2014 completed the picture: the adaptive immune system exerts little influence on 9464D growth out of the box, yet depletion of NK1.1+ cells accelerates tumor outgrowth even in Rag1-deficient animals, formally demonstrating that NK cells — not T cells — are the principal natural effectors against this tumor; and the tumor-infiltrating leukocyte landscape, rich in myeloid suppressor cells and regulatory T cells, reproduces the immunosuppressive microenvironment of human neuroblastoma.

What has made 9464D the default neuroblastoma immunotherapy model is how productively its coldness has been engineered around. To study anti-GD2 antibody therapy — the standard of care built around dinutuximab — investigators overexpressed GD3 synthase and GM2/GD2 synthase in the parental line to generate 9464D-GD2, whose dinutuximab suppression of established flank tumors in vivo was complete until resistance was deliberately induced by tumor-derived small extracellular vesicles — an experiment that identified farnesyltransferase inhibitor tipifarnib as an adjunct to rescue anti-GD2 efficacy. A complementary IFN-γ-inducible MHC-I subclone, 9464D-GD2-I, enabled the mechanistic demonstration that local radiation converts cold tumors into T-cell-inflamed ones whose checkpoint blockade then succeeds — the radio-immunotherapy paradigm now central to neuroblastoma immunotherapy development. Orthotopic intra-adrenal implantation has been shown to grow faster than flank grafts with a denser, more M2-like macrophage infiltrate, and combinations of cyclophosphamide with anti-GD2 or anti-4-1BB have extended survival in that setting. The parent line’s honest limitation is metastasis: 9464D itself forms spontaneous metastases poorly — two liver metastases emerged from thirty intravenously infused mice — and those rare metastases have been captured as the M1 and M2 derivative lines, which metastasize with 100% penetrance to liver and bone respectively and model the organotropism of human high-risk disease. Reporter engineering with luciferase and tdTomato completes a platform in which an immunologically faithful primary tumor, engineered antigen variants, and true metastatic derivatives coexist in a single C57BL/6 system.

Cell line Information: 9464D

The table below consolidates the key biological, genetic, and modeling characteristics of the 9464D line and its engineered derivatives.

Parameter Details
Cell line designation 9464D (parental); engineered variants 9464D-GD2 (high GD2) and 9464D-GD2-I (IFN-γ-inducible MHC-I plus GD2); reporter-engineered 9464D-Luc-tdT; metastatic derivatives M1, M2, and M1-variant lines (organ-tropic)
Species / genetic background Mouse (Mus mus musculus), C57BL/6 inbred background; syngeneic in C57BL/6 and C57BL/6J immunocompetent hosts
Origin Immortalized cell line derived from a spontaneous neuroblastoma arising in the TH-MYCN transgenic mouse developed by the Weiss laboratory (University of California, San Francisco); established by Hansford et al. (Molecular Cancer Therapeutics 3:323–332, 2004) with the sibling line 975A2
Depository / catalogue Distributed through the originating laboratories (Weiss lineage, obtained via the NCI/Jon Wigginton channel in published studies); not an ATCC listing — authentication is by documented provenance, MYCN-expression confirmation, and morphology
Tumor type Murine neuroblastoma, MYCN-driven; reflects human high-risk neuroblastoma in histology, gene-expression profile, and low tumor mutation burden
Molecular and antigen features MYCN overexpression; endogenous GD2 expression (lower than many human neuroblastoma lines); low MHC class I with approximately 25% of cells capable of IFN-γ-inducible upregulation — the immunologically cold, NK-surveilled phenotype of human high-risk disease
Immune biology Adaptive immunity exerts little influence on outgrowth at baseline; NK1.1⁺ depletion accelerates growth even in Rag1⁻/⁻ mice, demonstrating NK-cell dominance of natural surveillance; tumor-infiltrating leukocytes include tumor-associated myeloid cells and regulatory T cells mimicking human tumors
Growth properties Adherent/partially adherent neuroblastoma cells of rapid proliferation in standard complete medium; reliable syngeneic take at standard challenge doses
Recommended culture conditions DMEM supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 1 mM sodium pyruvate, 1× MEM non-essential amino acids, and penicillin/streptomycin at 37 °C in 5% CO₂; 9464D-GD2 cultures maintained under puromycin (6 μg/mL) plus blasticidin (7.5 μg/mL) selection; mycoplasma PCR-negative documented prior to in vivo use
Implantation formats and doses Subcutaneous: 0.5 × 106 to 1 × 106 cells per mouse with palpable tumors within 1–2 weeks; intra-adrenal orthotopic: faster growth with TAM-dense, M2-like infiltrate; intravenous experimental metastasis: 0.25–2 × 106 cells with in vivo bioluminescence tracking — parental 9464D metastasizes poorly (2/30 mice), while M1/M2 derivatives achieve 100% metastasis penetrance with liver and bone organotropism
Documented in vivo responses Anti-GD2 monoclonal antibody (dinutuximab-class) dramatically suppresses established 9464D-GD2 flank tumors; tumor-derived small extracellular vesicles confer near-complete resistance to anti-GD2 therapy, rescued by tipifarnib adjunct; local radiation plus checkpoint blockade succeeds in the 9464D-GD2-I setting through T-cell inflame conversion; cyclophosphamide plus anti-GD2 or anti-4-1BB extended survival in intra-adrenal models; NK-cell depletion accelerates parental tumor outgrowth
Key research applications Anti-GD2 antibody, bispecific, and CAR construct evaluation with ADCC- and NK-dependent readouts; checkpoint and radio-immunotherapy mechanism studies using the inducible-MHC-I subclone; macrophage/myeloid and Treg microenvironment targeting; organ-tropic metastasis biology with M1 (liver) and M2 (bone) derivatives; minimal-residual-disease and vaccination studies under immune competence

Our Services

An immunologically cold tumor is won or lost on engineering and analytics, and Alfa Cytology runs 9464D accordingly: authenticated banks of the parental, GD2-engineered, inducible-MHC-I, and reporter lines, validated flank, intra-adrenal, and intravenous formats with the platform’s metastasis caveats respected, scheduled caliper and in vivo bioluminescence monitoring, and endpoint pathology pairing GD2 expression with NK- and T-cell immunophenotyping — all under one accountable preclinical operation.

Workflow of 9464D Syngeneic Model Construction

Construction follows the published methodologies for each deployment format: 9464D or its engineered derivatives are expanded under authenticated conditions, implanted subcutaneously, intra-adrenally, or intravenously into C57BL/6 mice, grown to a standardized baseline, randomized, treated, and analyzed with neuroblastoma-appropriate efficacy and immune endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: 9464D or derivative lines are provenance-verified, confirmed mycoplasma-negative by PCR, checked for MYCN expression and GD2/MHC-I status by flow cytometry, and expanded into low-passage working banks under the validated DMEM formulation with selection maintained where engineered.
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at the study-specified dose of 0.5 × 106 to 1 × 106 cells (flank) or 0.25–2 × 106 cells (metastatic arms) in 100 μL of PBS.
  3. Anesthesia and implantation: C57BL/6 mice are anesthetized with isoflurane; cells are implanted subcutaneously into the flank, injected into the adrenal gland through a small abdominal approach for orthotopic arms, or infused intravenously via tail vein for experimental-metastasis designs.
  4. Establishment monitoring and randomization: Flank and orthotopic tumors are measured by caliper from first palpability; metastatic arms are tracked by in vivo bioluminescence after D-luciferin administration; animals are randomized once tumors reach protocol-specified baselines.
  5. Treatment administration: Experimental arms — anti-GD2 monoclonals and next-generation constructs, checkpoint inhibitors, local radiation fractions, farnesyltransferase and myeloid-targeting adjuncts, and combinations — proceed per study design with body-weight and toxicity monitoring.
  6. Immune monitoring: Tumor-infiltrating leukocytes are profiled ex vivo for NK-cell, CD8⁺ T-cell, myeloid-suppressor, and Treg composition by multi-parametric flow cytometry; GD2 and MHC-I surface expression are re-quantified at endpoint; NK-dependence depletion arms are included where the design requires.
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth inhibition, metastatic burden by imaging and ex vivo organ mapping, and survival with histopathology (H&E confirming neuroblastoma morphology, plus GD2, synaptophysin, Ki67, CD8, NK1.1, and PD-L1 immunohistochemistry) and terminal mechanistic sampling for resistance studies.
