PyMT (derived) Syngeneic Model Service for Breast Cancer

Fig 1: PyMT (derived) Syngeneic Model for Breast Cancer preclinical research.

The PyMT (derived) syngeneic model service for breast cancer is an immune-competent platform built on cell lines from the MMTV-PyMT transgenic mouse — the gold-standard model of human breast cancer progression — offering defined luminal epithelial, triple-negative-like mesenchymal, and TGFβ-plasticity variants across FVB/N and C57BL/6 syngeneic hosts. Alfa Cytology delivers this model as a complete preclinical capability — covering authenticated PyMT-derived line expansion and banking (PyMT-1099, Py8119, Py230, and reporter derivatives), mammary fat-pad orthotopic or subcutaneous implantation, scheduled caliper and in vivo bioluminescence monitoring, and full mammary histopathology with epithelial-mesenchymal immunophenotyping — all configured around each sponsor’s EMT, metastasis, checkpoint, or combination hypothesis.

Overview of PyMT (derived) Syngeneic Model for Breast Cancer

The MMTV-PyMT transgenic mouse, created by Guy, Cardiff, and Muller in 1992, is the single most influential model in breast cancer research: mammary-specific expression of the polyomavirus middle T antigen — which transforms epithelium through c-Src and PI3K/Akt signaling — drives tumors that recapitulate the human progression from ductal carcinoma in situ to invasive carcinoma with predictable, strain-dependent kinetics (mean latency around 53 days on the FVB background). From the tumors of this model, a family of transplantable lines has been derived that preserves the model’s biology while removing its long latency. On the FVB/N background, PyMT-1099 — isolated from the second/third mammary gland tumor of mouse 1099 — displays a well-differentiated cobblestone epithelial morphology, carries the PyMT transgene verifiable by genotyping PCR, and has become a flagship of epithelial plasticity research: treatment with 2 ng/mL TGF-β induces a complete epithelial-to-mesenchymal transition — total loss of E-cadherin and ZO-1 with induction of fibronectin and N-cadherin — and withdrawal reverses it through a mesenchymal-to-epithelial transition, with the cells notably resistant to TGF-β growth arrest while using non-canonical signaling to drive EMT; long-term exposure yields the PyMT LT mesenchymal subline, which metastasizes to the lung in 100% of orthotopically implanted animals.

Intratibial bone tumor imaging workflow in breast cancer bone models: bioluminescence imaging (BALB/c vs nu/nu), fluorescent imaging of whole leg, μCT longitudinal/cross sections, X-ray, and Goldner's/H&E histology — the imaging paradigm applied to PyMT-CK(OB). (Flatt, Courtney L., et al., 2026)

On the C57BL/6 background — the strain preferred for immunology because of its deep toolbox of knockout and transgenic hosts — the paired lines Py8119 and Py230 provide a deliberately staged contrast. Py8119 is a mesenchymal-like, triple-negative-phenotype (ER⁻/PR⁻/HER2⁻) line expressing N-cadherin, vimentin, Slug, and high cytokeratin 14, palpable in the mammary fat pad within five days at half of injection sites and far more aggressive than its sibling; Py230 is the luminal-like epithelial counterpart (cytokeratin 8⁻ and E-cadherin⁻ positive) with slower, well-differentiated growth. Py8119’s documented behavior carries a caution that belongs in every study design: attenuating its TGF-β signaling — by TβRII knockdown or systemic receptor-I kinase inhibition — enhances orthotopic growth in syngeneic C57BL/6 mice and increases metastatic colonization after intracardiac inoculation to lung, brain, jaw, and tibia, the opposite of what simpler models predict, making it the ideal substrate for precision testing of TGF-β-pathway inhibitors. Luciferase-GFP and luciferase-engineered derivatives support in vivo burden tracking, and the bone-metastasis-derived PyMT-BO1 line extends the platform to skeletal disease. Together, the PyMT-derived family spans the full epithelial-mesenchymal spectrum of breast cancer in two fully immunocompetent strain contexts — a combination no other single syngeneic platform offers.

Cell Line Information: PyMT (derived)

The table below consolidates the key biological, genetic, and modeling characteristics of the principal PyMT-derived lines and their reporter derivatives.

Transformant biology All lines carry the polyomavirus middle T antigen transgene, transforming cells via c-Src and PI3K/Akt signaling; PyMT transgene confirmed by genotyping PCR
PyMT-1099 (FVB/N) Well-differentiated cobblestone epithelial morphology; TGF-β (2 ng/mL) induces complete EMT (loss of E-cadherin/ZO-1, induction of fibronectin/N-cadherin) with MET on withdrawal; resistant to TGF-β growth arrest via non-canonical signaling; long-term TGF-β yields PyMT LT mesenchymal subline with high migration; orthotopic palpable tumors at ≈14 weeks in immunocompromised hosts with ≈50% lung metastasis, and 100% lung metastasis with PyMT LT
Py8119 (C57BL/6) Mesenchymal-like, triple-negative-phenotype (ER⁻/PR⁻/HER2⁻) line: spindle-shaped, N-cadherin⁻/vimentin⁻/Slug⁻ with high cytokeratin 14; highly aggressive — mammary fat-pad tumors palpable in 50% of sites within 5 days (versus >20 days for Py230); TGF-β signaling functional but non-growth-inhibitory; TβRII knockdown or systemic TβRI-kinase inhibition enhances orthotopic growth and metastatic colonization (lung, brain, jaw, tibia) — a documented caution for TGF-β-inhibitor studies
Py230 and PyMT-BO1 (C57BL/6) Py230: luminal-like epithelial counterpart with cuboidal morphology, cytokeratin 8⁻/E-cadherin⁻ positive, forming well-differentiated colonies, slower tumorigenicity; PyMT-BO1: bone-metastasis-derived subline characterized as luminal-like, used in 3D organotypic and in vivo bone studies

Key research applications EMT/MET plasticity and TGF-β-pathway precision studies; metastasis and organotropism research (lung, bone) with PyMT LT and PyMT-BO1 arms; checkpoint and combination-therapy evaluation in two strain backgrounds; tumor-microenvironment and MDSC biology; comparative luminal-versus-mesenchymal experimental design using the Py230/Py8119 pair

Our Services

A platform spanning two strain backgrounds and the full epithelial-mesenchymal spectrum rewards exactly the matched-design discipline Alfa Cytology brings: provenance-authenticated banking of each PyMT-derived line, genotype-confirmed expansion with PyMT transgene PCR verification, fat-pad orthotopic technique with contralateral matched-pair designs, in vivo bioluminescence tracking, and endpoint pathology pairing E-cadherin/vimentin/cytokeratin immunophenotyping with TIL profiling — all under one accountable preclinical operation.

Workflow of PyMT (derived) Syngeneic Model Construction

Construction follows the published derivation-and-implantation methodology for each line: PyMT-derived cells are expanded under authenticated conditions, implanted into the mammary fat pad or subcutaneously of syngeneic hosts, grown to a standardized baseline, randomized, treated, and analyzed with breast-cancer-appropriate efficacy and EMT endpoints. The workflow comprises the following stages.

  1. Cell authentication and banking: PyMT-derived lines are identity-verified (STR where on file), confirmed mycoplasma-negative, and verified for PyMT transgene presence by genotyping PCR, then expanded into low-passage working banks under line-specific formulations (F-12K low-serum for Py8119; standard complete medium for PyMT-1099).
  2. Cell harvest and formulation: Healthy cultures are trypsinized, washed, counted and viability-checked, then resuspended at the study-specified dose (e.g., 0.1 × 106 Py8119-Luc cells per fat pad in 1:1 Matrigel for matched-pair designs) per validated protocol.
  3. Anesthesia and implantation: Syngeneic hosts are anesthetized with isoflurane; cells are injected into the cleared #2/#4 mammary fat pads through a small skin approach, with contralateral matched-pair designs (e.g., control versus TβRII-knockdown Py8119) where designs require.
  4. Establishment monitoring and randomization: Tumors are monitored by palpation and caliper (Py8119 palpable within ≈5 days at 50% of sites) or by in vivo bioluminescence for reporter arms; animals are randomized once tumors reach protocol-specified baselines.
  5. Treatment administration: Experimental arms — TGF-β-pathway inhibitors, checkpoint inhibitors, EMT-targeting agents, chemotherapeutics, and combinations — proceed per study design, with the documented Py8119 pro-growth caution informing TGF-β-arm interpretation from the outset.
  6. Plasticity and immune monitoring: Paired in vitro EMT/MET assays (TGF-β 2 ng/mL induction and withdrawal timecourses with E-cadherin/ZO-1/fibronectin readouts) run alongside in vivo tumor-infiltrating-lymphocyte profiling by flow cytometry (CD8⁺, Treg, MDSC, macrophage compartments).
  7. Endpoint and histopathological analysis: Endpoints integrate tumor-growth kinetics, metastatic burden by imaging and ex vivo organ mapping, and survival with histopathology (H&E plus E-cadherin, vimentin, cytokeratins 8/14, Ki67, CD8, and marker-specific immunohistochemistry) and terminal EMT-marker quantification in primary tumors.
