B16-F10-OVA Syngeneic Model Service for Melanoma

Fig 1: B16-F10-OVA Syngeneic Model for Melanoma preclinical research.

The B16-F10-OVA syngeneic model of melanoma adds a defined ovalbumin antigen to the aggressive, poorly immunogenic B16-F10 melanoma, turning the field’s coldest solid tumor into a traceable platform for vaccine and combination-immunotherapy research in C57BL/6 hosts. Alfa Cytology delivers the complete B16-F10-OVA workflow—construct-verified cell banking, flank implantation, OT-I/OT-II-compatible designs, intervention scheduling and tetramer-based endpoint analysis—giving sponsors decision-ready preclinical datasets shaped to their program, strictly within a preclinical scope.

Overview of B16-F10-OVA Syngeneic Model for Melanoma

B16-F10-OVA answers a pointed question: what happens when a defined antigen is bolted onto a tumor famous for ignoring immunity? The parental B16-F10—selected through ten cycles of Fidler’s lung-colonization protocol from a 1954 spontaneous C57BL/6 melanoma—is poorly immunogenic, expresses MHC-I at low levels, grows with a 17-hour doubling time and metastasizes relentlessly. Engineering it to express chicken ovalbumin produced a line with dual antigen biology: the foreign model antigen, whose SIINFEKL epitope (OVA257–264) is displayed on H-2Kb and OVA323–339 on I-Ab, sits alongside the melanoma’s native differentiation antigens such as gp100—allowing model-antigen and tumor-antigen responses to be compared in the same tumor. Importantly, the construct landscape is plural: documented versions include lentiviral overexpression clones (Biocytogen’s 1-G12 and 1-G04), an ATCC pLX305 puromycin-resistance version (CRL-3686), the classic B16-OVA MO4 clone created for Falo and Rock’s 1995 Nature Medicine study, and transposon-based OVA-eGFP-neo integrations selected with G418. OVA expression demonstrably warms the tumor: subcutaneous implants show increased TIL infiltration with a significantly higher CD8+ fraction than wild-type B16-F10, and harvested tumors retain surface OVA—proof the antigen survives the in vivo passage.

In practice, two hundred thousand to half a million cells in the flank grow progressively—tracked by the 0.5 × long × short2 formula—while the heavier MO4 dosing tradition (5–10 million cells, greater than 60% take) models long-established, invariably progressive disease. The line’s combination record is remarkable: adoptively transferred OT-I cells synergize with agonistic anti-CD137 (clone 1D8) in a CD8-dependent fashion; an antigen-specific bacterial vaccine combined with anti-PD-L1 rescued dysfunctional endogenous T cells and rejected long-established tumors—one of the cleanest demonstrations of checkpoint-enabled T-cell revival; the dsRNA mimic BO-110 restored antitumor immunity with OVA/OT-I readouts; and prior OVA vaccination protected against B16-OVA challenge months later. Caveats are specific: construct heterogeneity across sources demands explicit verification before use; antibiotic selection must be maintained to prevent antigen loss; parental fast kinetics compress windows; low baseline MHC-I tempers expectations; and the MO4 subline carries documented BXV-1 provirus—a biosafety flag for that stock.

Parental line B16-F10 (ATCC CRL-6475); ten-cycle Fidler selection from B16, a spontaneous C57BL/6 melanoma (1954); poorly immunogenic, low MHC-I, ~17-hour doubling
Species / strain background Mus musculus; syngeneic host: C57BL/6; OT-I / OT-II transgenic compatibility
Antigen biology Full-length chicken OVA; SIINFEKL (OVA257–264) on H-2Kb to OT-I; OVA323–339 on I-Ab to OT-II; dual-antigen landscape alongside native gp100/pmel melanoma antigens; surface display confirmed by 25-D1.16 staining
Immunogenicity OVA expression increases immunogenicity versus parental B16-F10; subcutaneous implants show increased TIL infiltration with a significantly higher CD8+ fraction; harvested tumors retain surface OVA expression
Standard model 2 × 105–5 × 105 cells subcutaneously per flank (female C57BL/6, ~7 weeks in published designs); volume = 0.5 × long diameter × (short diameter)2; MO4 tradition: 5–10 × 106 cells, >60% take, invariably progressive and lethal course; sacrifice at >1.5 cm3
Combination precedents OT-I (2 × 106, day 3/7) plus anti-CD137 (clone 1D8), CD8-dependent; antigen-specific bacterial vaccine plus anti-PD-L1 rescues dysfunctional endogenous T cells to reject long-established tumors; BO-110 (dsRNA mimic) restores antitumor immunity with OVA/OT-I readouts; OVA vaccination protects from later B16-OVA challenge (documented at ~65 days post-vaccination)
Detection toolkit H-2Kb–SIINFEKL tetramers; anti-SIINFEKL/H-2Kb clone 25-D1.16; RF33.70 hybridoma (MO4 history); OVA-specific ELISPOT and intracellular cytokine staining; melanin retained as a parental identity feature
Culture conditions DMEM high glucose (with pyruvate) + 10% FBS (RPMI-1640 also reported); 37 °C, 5% CO2; selection antibiotic maintained per construct (G418 ~400 µg/mL for transposon versions; puromycin for pLX305)
Considerations Construct heterogeneity across sources demands explicit verification; antibiotic selection required to maintain OVA expression; parental fast kinetics compress treatment windows; low baseline MHC-I tempers single-agent immunotherapy; MO4 subline carries documented BXV-1 provirus (biosafety flag for that stock); model-antigen translation gap applies
Representative applications Checkpoint-inhibitor screening in a defined-antigen setting, cancer vaccine discovery (peptide, protein, mRNA, DC and bacterial platforms), adoptive T-cell plus costimulatory combinations, T-cell dysfunction rescue studies, dsRNA immunomodulator testing, melanoma immunogenicity engineering

Workflow of B16-F10-OVA Syngeneic Model Construction

Construction of the B16-F10-OVA syngeneic model is an implantation workflow whose quality rests on construct-verified cells, vigorous C57BL/6 recipients and disciplined monitoring. A standard engagement proceeds as follows:

  1. Source and qualify cells: B16-F10-OVA stocks are cleared by mycoplasma PCR and STR-authenticated, with the construct explicitly documented; OVA expression is confirmed by Western blot and SIINFEKL–H-2Kb staining (25-D1.16), selection is maintained through expansion, and luciferase labeling is titrated in vitro where imaging readouts are required.
  2. Prepare recipients: C57BL/6 mice (typically 6–8 weeks old; OT-I/OT-II transgenic or congenically marked hosts where antigen-specific designs call for them) complete acclimation and health screening.
  3. Prepare the inoculum: log-phase cells are harvested, washed, counted and resuspended in PBS (2 × 105–5 × 105 cells in 100 µL for standard flank implants; higher doses per the MO4 tradition where long-established disease is the design).
  4. Implant: cells are injected subcutaneously into the flank with consistent anatomical positioning for reproducible caliper measurement.
  5. Randomize: when tumors reach palpability, animals are assigned to groups balanced by tumor volume and body weight.
  6. Dose and follow: test articles (checkpoint agents, vaccines, costimulatory antibodies, dsRNA immunomodulators) are administered on schedule; tumors are measured two to three times weekly using the 0.5 × long × short2 formula, with body weight and survival logged; OT-I/OT-II transfer arms follow the documented day 3–7 window where designed.
  7. Terminate and analyze: tumors are resected and weighed; tumor-infiltrating lymphocytes are analyzed by flow cytometry with H-2Kb–SIINFEKL tetramers and activation/exhaustion panels; sections are stained with 25-D1.16 and CD3 alongside melanin assessment; draining lymph nodes, spleen and lungs are examined; tissues enter histopathology and molecular analysis.
