K7M3 / K7M3-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: K7M3 / K7M3-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The K7M3 / K7M3-luc orthotopic mouse model delivers one of the most aggressive syngeneic osteosarcoma systems available, implanting a lung-metastasis-selected murine line into the tibial marrow cavity of immunocompetent BALB/c mice where it drives rapid bone destruction and pulmonary spread. Alfa Cytology provides this model as a fully managed preclinical service — authenticated cell banking, precision intratibial surgery with X-ray and bioluminescence monitoring, dosed intervention including NK-cell, cytokine, and bisphosphonate strategies, and complete terminal pathology — giving sponsors a demanding bone-sarcoma platform without standing up specialized surgical and imaging capabilities internally.

Overview of K7M3 / K7M3-luc Orthotopic Mouse Model for Sarcoma

K7M3 belongs to the celebrated K7 family of murine osteosarcoma lines, all tracing back to a tumor that arose spontaneously in the femur of a BALB/c mouse nearly 900 days of age. Whereas its parent K7M2 was selected through repeated cycles of intraosseous implantation and lung-metastasis harvest, K7M3 was taken further: after intravenous challenge of the parental line, the lung metastases that escaped the pulmonary first-pass defense were recovered and expanded, locking in a phenotype of exceptional metastatic aggression. In orthotopic comparisons, K7M3 outruns its relatives — among the clonally related K7M2, K12, and K7M3 lines, K7M3 is classified as the highly aggressive member, with K7M2 and K12 substantially milder. Its aggression is molecularly legible: the line expresses RANKL (unlike the milder K12), secretes abundant osteoprotegerin, and actively drives osteoclast differentiation and bone resorption, making it the natural substrate for studying osteosarcoma-mediated osteolysis and the bone-tumor vicious cycle; bisphosphonates have been shown to blunt K7M3-driven osteolysis by attenuating tumor MCP-1 and RANKL expression.

A second distinctive thread is immune evasion. Work on the K7 family demonstrated that Fas-positive osteosarcoma cells are efficiently cleared from the lung within about 48 hours of intravenous arrival, while K7M3 cells — selected precisely for surviving that gauntlet — are characteristically Fas-negative or Fas-low, a corruption of the Fas pathway that delays pulmonary clearance and boosts metastatic potential; this same mechanism has been shown to blunt the efficacy of inhaled chemotherapeutics such as aerosol gemcitabine. The model has consequently become a preferred arena for pulmonary-delivery and innate-immunity strategies, hosting studies of aerosol interleukin-2, natural killer cell therapy, β-glucan plus CD40-agonist combinations, and therapeutic vaccination with tumor-lysate-loaded dendritic cells. Orthotopically, K7M3 is implanted by intratibial injection of a compact inoculum — published protocols use two 10 μL injections of 1 × 105 cells given 24 hours apart — with weekly X-ray or caliper assessment, amputation at a defined tumor threshold, and histological scoring of femoral relapse and lung metastases at termination. The K7M3-luc derivative, engineered with firefly luciferase, adds quantitative whole-body tracking of primary and pulmonary tumor burden, sharpening response kinetics for every one of these study styles.

Cell Line Information: K7M3 / K7M3-luc

K7M3 (Cellosaurus CVCL_XG68; available from Hysigen as TCM-C791) is a murine osteosarcoma subline of K7M2, established by intravenous injection of the parent line into mice followed by harvest of the resulting lung metastases, as reported by Gordon and colleagues in 2007. The luciferase-tagged K7M3-luc derivative was originally generated at the NCI and is widely distributed through the osteosarcoma research community for optical tracking studies. Principal characteristics are compiled below.

Cellosaurus accession CVCL_XG68 (RRID: CVCL_XG68); commercially available as Hysigen TCM-C791
Species / strain Mus musculus, BALB/c
Lineage Sublime of K7M2 (CVCL_V455), itself derived from the spontaneous BALB/c osteosarcoma line K7
Disease Osteosarcoma
Donor sex / age Female / 895 days
Derivation Established by intravenous injection of the parent K7M2 line into mice, followed by harvest and expansion of lung metastases (Gordon et al., 2007); derived site: metastatic lung
Aggressiveness Classified as the highly aggressive member among the clonally related K7M2, K12, and K7M3 lines; rapid orthotopic primary growth with early osteolysis
Bone biology Expresses RANKL (unlike K12) and secretes roughly twofold more osteoprotegerin (OPG) than non-neoplastic MC3T3-E1 osteoblasts; drives osteoclast differentiation and bone resorption; bisphosphonates attenuate K7M3-mediated osteolysis via reduced MCP-1 and RANKL
Fas pathway Characteristically Fas-negative / Fas-low; corruption of the Fas pathway delays pulmonary clearance of tumor cells, enhances metastatic potential, and reduces the effect of aerosol gemcitabine

Workflow of K7M3 / K7M3-luc Orthotopic Mouse Model Construction

Orthotopic K7M3 work rewards discipline in three places: compact-inoculum tibial surgery, radiographic plus optical monitoring, and rigorous metastatic histopathology. A standard program unfolds as follows.

  1. Cell preparation and quality control: K7M3 or K7M3-luc cells are expanded at low passage, confirmed mycoplasma-free, and luciferase activity is validated in vitro; the working inoculum is prepared at 1 × 105 cells in 10 μL PBS per injection shortly before surgery.
  2. Host selection and acclimation: Male or female BALB/c mice, typically 6–8 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility prior to the procedure.
  3. Anesthesia and tibial access: Under isoflurane anesthesia, the proximal tibia is exposed through a small incision, the knee is flexed, and a 27-gauge needle is advanced through the tibial plateau into the marrow cavity.
  4. Intratibial inoculation: The 10 μL cell suspension is injected slowly into the marrow cavity; the procedure is repeated 24 hours later into the same limb, a two-hit inoculation format reported in published K7M3 protocols to standardize take rates.
  5. Closure and postoperative care: The skin is closed, analgesia is administered, and mice are monitored daily for weight, gait, and wound integrity, with humane endpoints defined in advance.
  6. Primary tumor monitoring: Tibial tumors are assessed weekly by caliper measurement and X-ray radiography, which captures the osteolytic signature of K7M3 growth; luciferase-tagged cohorts additionally undergo scheduled bioluminescence imaging after intraperitoneal D-luciferin injection.
  7. Randomization, treatment, and amputation: Mice are randomized into treatment arms at a defined tumor volume, test articles are dosed according to protocol, and the affected limb is amputated once the primary tumor reaches the preset threshold (for example ~1.5 cm3), so that anti-metastatic efficacy is measured against established pulmonary disease.
  8. Endpoint analysis and reporting: At termination, femora are examined histologically for local relapse and lungs for metastatic burden, with necrotic versus viable tumor areas scored per published criteria; imaging signals are reconciled with histology, and all data are consolidated into the final report.
