RM-1 / RM-1-luc Orthotopic Mouse Model Service for Prostate Cancer

Fig 1: RM-1 / RM-1-luc Orthotopic Mouse Model for Prostate Cancer preclinical research.

The RM-1 / RM-1-luc orthotopic mouse model of prostate cancer places murine prostate tumor cells directly into the prostatic gland of immunocompetent C57BL/6 hosts, recreating the anatomical niche, stromal crosstalk and vascular support that subcutaneous implants cannot provide. From cell line authentication, survival surgery and longitudinal luciferase-based monitoring to necropsy, histopathology and tumor-immune profiling, Alfa Cytology delivers complete, decision-ready RM-1 / RM-1-luc orthotopic programs configured around each sponsor’s mechanism, dosing schedule and endpoint requirements—strictly within a preclinical scope.

Overview of RM-1 / RM-1-luc Orthotopic Mouse Model for Prostate Cancer

RM-1 is a murine prostate carcinoma cell line syngeneic in C57BL/6 mice, first described in the mid-1990s. It was derived from urogenital sinus epithelial cells of a Trp53-deficient C57BL/6 mouse and transformed ex vivo with a recombinant retrovirus (Zipras/myc9) carrying v-Ha-Ras and v-Myc. The cells show a luminal epithelial identity (cytokeratin 18 positive) and express androgen receptor, yet behave aggressively and grow androgen-independently in vivo. Because donor and host share the same genetic background, RM-1 tumors arise in animals with a fully intact immune system—a fundamental advantage over human xenografts propagated in immunodeficient mice.

In the orthotopic format, RM-1 or RM-1-luc cells are implanted into the prostatic lobe, where the tumor recruits organ-resident stroma, acquires physiological vasculature and develops within an authentic tumor microenvironment. The RM-1-luc derivative constitutively expresses firefly luciferase, permitting non-invasive, serial bioluminescence imaging from the earliest stages of tumor establishment and sensitive detection of occult lesions. These tumors grow remarkably fast—often measurable within days of implantation—and display a characteristically “cold” immune profile with sparse T-cell infiltration and abundant myeloid-derived suppressor cells, so single-agent checkpoint blockade typically yields limited activity while radiotherapy-based combinations can re-sensitize the tumor to immune attack. This biology makes the model a workhorse for preclinical evaluation of immunotherapy combinations, radiation, nanoparticle delivery, androgen-pathway agents and metastatic spread, although its rapid kinetics make it less suitable for long-horizon studies.

Cell Line Information: RM-1 / RM-1-luc

RM-1 and its luciferase-labeled derivative RM-1-luc share the same C57BL/6 genetic background and tumorigenic behavior; the reporter line additionally carries a constitutively expressed firefly luciferase cassette for bioluminescence imaging. Key characteristics are summarized below.

Our Services

Supported by a dedicated in vivo pharmacology team and integrated surgical, imaging and pathology capabilities, Alfa Cytology builds each RM-1 / RM-1-luc orthotopic program around the sponsor’s hypothesis—adjusting inoculum size, randomization, dosing windows and endpoint panels to produce clean, interpretable efficacy data under a strictly preclinical framework.

Workflow of RM-1 / RM-1-luc Orthotopic Mouse Model Construction

Orthotopic construction of the RM-1 / RM-1-luc model is a standardized survival-surgery procedure performed under inhalation anesthesia, with aseptic technique, scheduled analgesia and daily post-operative observation aligned to institutional animal-care standards. A typical engagement proceeds through the following stages:

  1. Cell line qualification: RM-1 or RM-1-luc cells are mycoplasma-tested, identity-confirmed, and—for the reporter line—screened for stable, uniform luciferase expression before scale-up.
  2. Animal preparation: male C57BL/6 mice (typically 6–8 weeks of age) are acclimatized, health-checked and randomized by body weight prior to surgery.
  3. Anesthesia and asepsis: mice are anesthetized with isoflurane, the lower abdomen is disinfected, and sterile instruments and gloves are maintained throughout.
  4. Surgical exposure: a small midline abdominal incision is made and the bladder is gently retracted to expose the prostate gland under magnification.
  5. Orthotopic inoculation: RM-1 / RM-1-luc cells (commonly 2 × 106 cells in 50 µL of 50% Matrigel) are injected into the dorsal prostatic lobe with a fine-gauge needle to form a confined bleb at the site.
  6. Closure and analgesia: the abdominal wall is sutured with absorbable material, the skin is closed with wound clips, and buprenorphine is administered with scheduled follow-up doses until full recovery.
  7. Longitudinal monitoring: tumor burden is tracked by serial bioluminescence imaging (D-luciferin, IVIS) for RM-1-luc, supplemented by caliper, ultrasound or CT measurements, together with body weight and clinical scoring.
  8. Endpoint and analysis: at termination, tumors are excised and weighed, organs are inspected for metastasis, and tissues are processed for histopathology, immunohistochemistry, flow-cytometric immune profiling and downstream molecular assays.
