K7M2 / K7M2-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: K7M2 / K7M2-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The K7M2 / K7M2-luc orthotopic mouse model is the benchmark syngeneic osteosarcoma platform, implanting a highly metastatic murine bone-tumor line directly into the tibial marrow cavity of immunocompetent BALB/c mice to recreate primary bone sarcoma growth and its near-obligatory spread to the lungs. Alfa Cytology provides this model as a complete preclinical package — authenticated cell banking, precision intratibial surgery, serial luciferase imaging of primary tumors and pulmonary metastases, therapeutic intervention including immunotherapies, and terminal histopathology — so sponsors gain a demanding metastasis model without developing microsurgical and imaging capabilities in-house.

Overview of K7M2 / K7M2-luc Orthotopic Mouse Model for Sarcoma

K7M2 is a murine osteosarcoma cell line with one of the most instructive pedigrees in preclinical bone cancer research. Its ancestor, K7, was among the first metastatic osteosarcoma lines ever established, derived from a tumor that arose spontaneously in an aged female BALB/c mouse. The K7M2 (wild-type) line was created by Khanna and colleagues in 2000 through deliberate in vivo selection: K7 cells were injected into the proximal tibia, the resulting lung metastases were harvested and re-implanted into tibial sites, and this cycle was repeated until a ferociously aggressive derivative emerged. The result is a bone-lineage osteoblast-type cell (expressing bone sialoprotein, osteopontin, decorin, biglycan, and ezrin, with CD31 positivity) that forms primary tumors with better than 95% take rates in syngeneic BALB/c hosts and seeds pulmonary metastases in over 90% of animals — against 33% for its intermediate sister line K12 and essentially zero for the parental K7. Histology of both primary tumors and lung lesions mirrors human osteosarcoma, and the model's high angiogenic index and ezrin-driven metastatic biology have made it a reference system for dissecting the spread of bone sarcoma.

Orthotopically, K7M2 is most often implanted by intratibial injection of a concentrated cell suspension — classically 1 × 106 cells in 25 μL — into the marrow cavity of the proximal tibia, a technique that places tumor cells in contact with bone, marrow stroma, and the remodeling microenvironment from which osteosarcoma naturally arises. Primary tibial tumors become palpable within roughly two weeks, after which pulmonary dissemination accelerates, and a widely used study design couples orthotopic growth with palliative limb amputation once tumors reach a defined volume so that anti-metastatic therapies can be tested against established lung disease. The luciferase-tagged derivative, K7M2-luc, enables quantitative bioluminescence tracking of both the primary lesion and lung burden on IVIS-class imagers, sharpening the detection of metastatic foci and response kinetics. Because the model runs in immunocompetent BALB/c mice with an intact immune compartment — and shows high immune-cell infiltration and responsiveness to combination immunotherapy while being largely refractory to PD-1 blockade alone — it has become a preferred proving ground for checkpoint combinations, cytokine therapies, and anti-metastatic strategies in sarcoma research.

Cell Line Information: K7M2 / K7M2-luc

K7M2 wt is distributed by the American Type Culture Collection (ATCC CRL-2836); ATCC additionally banks the plasmid-bearing derivative K7M2-pCI Neo (CRL-2837). The luciferase-tagged variant used for optical tracking, K7M2-luc, is generated by stable transfection of a luciferase expression construct and retains the parental take rates and metastatic behavior. Core characteristics are compiled below.

Parameter Description
Cell line K7M2 / K7M2-luc (wild-type K7M2 with luciferase-tagged derivative)
ATCC catalog No. CRL-2836 (K7M2 wt); related derivative K7M2-pCI Neo = CRL-2837; K7M2-luc generated by stable luciferase transfection

Key markers & products CD31 positive; produces factor VIII, bone sialoprotein (BSP), biglycan, decorin, osteopontin (SPP1), and ezrin (villin 2); expresses complement C3 and high-affinity IgG Fc receptor (Fcgr1)
Metastasis-associated biology Ezrin expression linked to metastatic behavior; metastatic cells and lesions are Fas-negative, with low Fas correlating to high metastatic potential; Wnt/β-catenin and PI3K/AKT pathways implicated in growth
Complete growth medium DMEM (4.5 g/L glucose, 4 mM L-glutamine) with 10% fetal bovine serum; 1% penicillin–streptomycin commonly added

Reporter system (K7M2-luc) Firefly luciferase; quantified in vivo after intraperitoneal D-luciferin injection on an IVIS-class imager, with histological confirmation of metastatic lesions
Immunotherapy relevance Immunogenic phenotype with high immune infiltrates in syngeneic hosts; refractory to anti-PD-1/PD-L1 monotherapy but responsive to combination strategies — suited to checkpoint, cytokine, and vaccine studies
Primary applications Osteosarcoma biology, metastasis and anti-metastatic drug screening, immunotherapy evaluation, angiogenesis studies, radiation and combination regimens

Workflow of K7M2 / K7M2-luc Orthotopic Mouse Model Construction

Orthotopic osteosarcoma modeling hinges on three skills: disciplined culture of the metastatic line, precise intratibial microsurgery, and quantitative imaging of disseminated disease. A standard engagement proceeds as follows.

  1. Cell preparation and quality control: K7M2 or K7M2-luc cells are expanded at low passage, confirmed mycoplasma-free, and luciferase activity validated in vitro; the inoculum is concentrated to the standard 1 × 106 cells in 25 μL PBS shortly before surgery.
  2. Host selection and acclimation: Male or female BALB/c mice, typically 6–10 weeks of age, are quarantined and acclimated in a specific-pathogen-free barrier facility before the procedure.
  3. Anesthesia and surgical access to the tibia: Under isoflurane anesthesia, the proximal tibia is exposed through a small longitudinal skin incision, and the knee joint is flexed to position the tibial plateau for marrow-cavity access.
  4. Intratibial inoculation: A fine-gauge needle is advanced through the tibial plateau into the marrow cavity, and the 25 μL cell suspension is injected slowly; the injected limb is typically the hind limb, with care taken to prevent reflux along the needle track.
  5. Closure and postoperative management: The skin is closed with sutures or wound clips, analgesia is administered, and mice recover with daily monitoring of weight, gait, and incision integrity.
  6. Primary tumor monitoring and imaging: Palpation and caliper measurement track the tibial tumor from approximately two weeks post-implantation, while bioluminescence imaging after intraperitoneal D-luciferin injection provides quantitative readouts of primary tumor and early pulmonary signal.
  7. Randomization, intervention, and amputation option: Mice are randomized into treatment arms at a defined tumor volume; where the study targets metastatic disease, the affected limb is amputated at the preset threshold (for example near 450 mm3) so that therapies are tested against established lung metastases.
  8. Endpoint analysis and reporting: At termination, primary tumors, lungs, and other organs are resected; metastatic nodules are enumerated and weighed, lungs undergo H&E staining and immunohistochemistry (including CD31/angiogenesis and ezrin panels), imaging signals are cross-referenced with histology, and all data are consolidated into the final report.
