MG-63 / MG-63-luc Orthotopic Mouse Model Service for Sarcoma

Fig 1: MG-63 / MG-63-luc Orthotopic Mouse Model for Sarcoma preclinical research.

The MG-63 / MG-63-luc orthotopic mouse model transplants one of the most widely studied human osteosarcoma lines directly into the femur or tibia of immunodeficient mice, where it recreates bone-confined tumor growth with an osteoblastic, mineralizing character distinct from more aggressive sarcoma platforms. Alfa Cytology delivers this model as a complete preclinical service — authenticated cell banking, precision intraosseous implantation in either cell-suspension or tumor-fragment format, X-ray and luciferase-based monitoring, therapeutic intervention, and terminal histopathology — giving sponsors a well-characterized human bone-tumor platform without building microsurgical and imaging capability from scratch.

Overview of MG-63 / MG-63-luc Orthotopic Mouse Model for Sarcoma

MG-63 is among the oldest and most cited human osteosarcoma cell lines in existence, deposited with the American Type Culture Collection in 1977 by A. Billiau after isolation from the distal femur of a 14-year-old Caucasian male with osteosarcoma. The line shows fibroblast, spindle-shaped morphology, adherent growth, and a hypotriploid karyotype with a modal chromosome number of 66 and 18–19 marker chromosomes common to all cells. Biologically it behaves as an immature, pre-osteoblastic cell: it responds to parathyroid hormone and vitamin D3 much like primary human osteoblasts, displays alkaline phosphatase activity, yet mineralizes poorly and is generally arrested before full maturation. Two features gave the line its early fame — it produces exceptionally high yields of interferon following superinduction with polyinosinic–polycytidylic acid, cycloheximide, and actinomycin D, and it expresses both type I and type II transforming growth factor-β receptors — while modern genomic annotation records a homozygous CDKN2A (p16) deletion. This combination has made MG-63 a reference line not only for osteosarcoma pharmacology but also for bone biology, biomaterial cytocompatibility, and three-dimensional scaffold research.

In vivo, MG-63 forms xenografts in immunodeficient mice, though its orthotopic personality differs instructively from highly aggressive lines such as 143B: primary intraosseous tumors grow more indolently, and published orthotopic studies report a cell-suspension take rate of about 70% at the distal femur with tumor tissue transplantation achieving essentially 100%, radiographic evidence by roughly four weeks, and histology faithful to human osteosarcoma. Far from a drawback, this slower tempo suits long-window efficacy studies, neoadjuvant-style treatment schedules, and bone-formation questions, and the line's osteoblastic phenotype provides a contrasting comparator to osteolytic platforms within the same skeletal site. The luciferase-tagged derivative, MG-63-luc — generated with high-efficiency lentiviral transduction and validated in Swiss nude and NSG hosts — enables quantitative bioluminescence tracking of intraosseous tumor burden from early time points, while serial X-ray captures the mineralizing signature of the disease. Together, the MG-63 / MG-63-luc orthotopic model offers a well-behaved, extensively characterized human osteosarcoma system for studies where controlled growth kinetics and bone-specific biology matter.

Our Services

Commissioning Alfa Cytology for the MG-63 / MG-63-luc orthotopic model places a complete preclinical workflow under one roof: authenticated, low-passage human cell banks are expanded and luciferase-verified, immunodeficient hosts are prepared and acclimated, the intraosseous inoculum — cell suspension or serially passaged tumor tissue — is delivered into the femur or tibia by microsurgeons, tumor take and growth are followed by scheduled X-ray and bioluminescence imaging, and your treatment arms run through amputation criteria, terminal bone histology, and full reporting under IACUC-compliant conditions.

Workflow of MG-63 / MG-63-luc Orthotopic Mouse Model Construction

Orthotopic MG-63 work centers on three disciplines: pristine culture of a well-characterized human line, intraosseous microsurgery in either suspension or fragment format, and dual-mode radiographic-plus-optical monitoring. A standard program unfolds as follows.

  1. Cell preparation and quality control: MG-63 or MG-63-luc cells are expanded at low passage, confirmed mycoplasma-free and STR-authenticated, and luciferase activity validated in vitro before implantation.
  2. Host selection and acclimation. Male or female immunodeficient mice: athymic nude or NSG strains, typically 4–6 weeks of age — are quarantined and acclimated in a specific-pathogen-free barrier facility.
  3. Anesthesia and bone access: Under isoflurane anesthesia, a small incision exposes the distal femur or proximal tibia, and the knee is flexed to position the growth plate for marrow-cavity entry.
  4. Intraosseous inoculation: A fine-gauge needle is advanced through the cortical entry point into the marrow cavity, and a concentrated cell suspension — on the order of 106 cells in 10–20 μL, often with Matrigel support — is injected slowly; alternatively, a small fragment of serially passaged MG-63 tumor tissue is inserted to exploit its higher take rate.
  5. Closure and postoperative management: The entry site is sealed, the incision is closed in layers, analgesia is administered, and mice recover with daily monitoring of weight, gait, and wound integrity.
  6. Tumor monitoring with dual modalities: Caliper and scheduled X-ray radiography follow the mineralizing tumor from approximately two weeks post-implantation, while luciferase-tagged cohorts undergo bioluminescence imaging after intraperitoneal D-luciferin injection to quantify intraosseous burden and detect dissemination.
  7. Randomization and treatment: Mice are randomized into arms on the basis of imaging signal, and test articles are dosed according to protocol — a tempo well suited to MG-63's longer, more controllable growth window.
  8. Endpoint analysis and reporting: At termination, tumor-bearing bones are resected, imaged, and processed for H&E and immunohistochemistry (including osteogenic markers), metastatic organs are examined, imaging signals are reconciled with histology, and all data are consolidated into the final report.

Case Study-MG-63 / MG-63-luc Orthotopic Mouse Model Development

Alfa Cytology has applied the MG-63 / MG-63-luc orthotopic platform to preclinical programs favoring controlled, longer-window efficacy readouts — including targeted agent evaluation, bone-directed therapies, and biomaterial-adjacent oncology studies where an osteoblastic human tumor is essential. In these engagements, serial bioluminescence imaging quantified intraosseous tumor burden over time while X-ray captured the mineralizing disease phenotype, and terminal bone histology with osteogenic-marker panels completed the assessment. Study parameters such as implantation site, inoculum format, imaging cadence, and endpoint panels are tailored to each program. Case materials can be shared under confidentiality agreements upon request.
