L1210 / L1210-luc Orthotopic Mouse Model Service for Leukemia

Fig 1: L1210 / L1210-luc Orthotopic Mouse Model for Leukemia preclinical research.

The L1210 / L1210-luc orthotopic mouse model for leukemia reproduces rapidly disseminating lymphocytic leukemia in immunocompetent DBA/2 hosts, combining a one-century pedigree in cancer drug screening with modern bioluminescence readouts. With a preclinical-only service portfolio, Alfa Cytology provides this historic syngeneic platform end to end: authenticated L1210 or L1210-luc cell supply, systemic inoculation, scheduled in vivo imaging and survival monitoring, and terminal organ analysis.

Overview of L1210 / L1210-luc Orthotopic Mouse Model for Leukemia

L1210 is a mouse lymphocytic leukemia cell line with a distinguished history: it arose in 1948 when DBA/2 mice were exposed to the chemical carcinogen 3-methylcholanthrene, and lymphoid cells from the resulting leukemia were propagated as the L1210 line. The cells display lymphoblast morphology, grow in suspension, and divide roughly every 10 to 12 hours in vitro, giving the population an exceptional proliferative capacity. Because the line is of mouse origin, it can be engrafted into syngeneic DBA/2 mice or related F1 hybrids without rejection, keeping the immune system fully intact. Historically, L1210 became the first leukemia model adopted by the National Cancer Institute’s national drug development program in the 1950s and served for decades as the primary in vivo screen for antitumor agents; numerous drug-resistant sublines, such as cytarabine-resistant L1210/Ara-C, were derived from it and remain in use for resistance research.

(Kun, Li, et al., 2026)

In contemporary orthotopic use, L1210 cells are introduced into the natural disease compartment of the host, most commonly by intraperitoneal or intravenous injection of a dilute ascitic suspension containing on the order of 105 cells per animal. The cells seed the spleen, bone marrow, lymphoid tissues, and peritoneal cavity with a take rate approaching 100%, producing uniform, aggressive disease in which the in vivo tumor burden doubles in under half a day and untreated animals typically survive only about one week. Luciferase-tagged derivatives (L1210-luc) bring this classic model into the imaging era: whole-animal bioluminescence after D-luciferin administration allows disease burden to be quantified longitudinally, responses to treatment to be read out days before clinical decline, and terminal organ involvement to be mapped by ex vivo imaging. The combination of speed, reproducibility, and quantitative imaging makes the L1210 / L1210-luc orthotopic model a practical choice for first-pass in vivo pharmacology of antileukemic agents. Figure 1 summarizes how this model is deployed.

Cell Line Information: L1210 / L1210-luc

Few cell lines have contributed more to cancer pharmacology than L1210. The essential characteristics of the parent line and its luciferase-tagged derivative are compiled below.

Cell Line Name L1210 (synonyms: L-1210, L1210D); luciferase-labeled derivative: L1210-luc
Catalog / Accession ATCC CCL-219; Cellosaurus CVCL_0382
Species / Strain of Origin Mouse (Mus musculus) / DBA/2
Derivation Chemically induced by skin painting with 3-methylcholanthrene in 1948; established from the lymphocytic leukemia that developed; propagated by serial transplantation in DBA/2 mice, with in vitro suspension culture first reported in 1966
Morphology / Growth Lymphoblast morphology; grows in suspension without attachment
Doubling Time Approximately 10–12 hours in vitro; in vivo tumor doubling time reported at 0.34–0.46 days
Culture Conditions RPMI-1640 with 10% fetal bovine serum (37 °C, 5% CO₂); historical formulations use DMEM with horse serum; start cultures at about 5 × 104 viable cells/mL and feed every 2–4 days
Biosafety Level BSL-1
Engraftment Profile Near-100% take rate in DBA/2 or F1 hybrid recipients after intraperitoneal or intravenous inoculation of ~105 cells; median survival of untreated animals approximately 6–10 days
Tumor Distribution Disseminates to spleen, bone marrow, lymph nodes, and peritoneal cavity; measurable as ascites or by bioluminescence
Historical Significance First leukemia model used in the National Cancer Institute’s national drug screening program from the 1950s; benchmark system for antitumor agent evaluation and a parent of numerous drug-resistant sublines (e.g., L1210/Ara-C)
Immunology Notes Expresses tumor-associated antigens recognized by syngeneic monoclonal antibodies; reported insensitive to anti-PD-1 / anti-PD-L1 monotherapy in the ascites format, a useful baseline for immunotherapy combination studies
L1210-luc Derivative Stably expresses firefly luciferase, enabling non-invasive in vivo bioluminescence imaging of disseminated disease and ex vivo organ-level mapping at endpoint
Research Applications First-pass in vivo antileukemic efficacy screening, drug resistance and pharmacology studies, pharmacokinetic/pharmacodynamic modeling, immunotherapy combination evaluation, biomarker and minimal residual disease research

Our Services

An L1210 / L1210-luc engagement with Alfa Cytology covers the full arc of the study: verified cell banking, standardized inoculation of syngeneic recipients, caliper-free imaging or body-weight-based disease monitoring, dosing per the client’s regimen, and a terminal package pairing survival statistics with organ-level bioluminescence.

Workflow of L1210 / L1210-luc Orthotopic Mouse Model Construction

Deploying the L1210 / L1210-luc orthotopic leukemia model is a rapid, highly standardized process in which a small inoculum reliably generates disseminated disease within days. The workflow below reflects how this platform is executed from cell bank to data package.

  1. Cell Bank Verification and Expansion: L1210 or L1210-luc stocks are authenticated by STR profiling and mycoplasma testing, then expanded in suspension culture with fresh medium replenished every two to four days.
  2. Luciferase Calibration (L1210-luc Only): Signal output per cell is verified in vitro so that in vivo photon flux can be converted into relative disease burden.
  3. Mouse Randomization and Grouping: DBA/2 or F1 hybrid mice are acclimatized and distributed into treatment and control groups by body weight under approved animal protocols.
  4. Orthotopic Systemic Inoculation: A dilute single-cell suspension, on the order of 105 cells in a small volume of sterile buffer or ascitic fluid, is delivered intraperitoneally or via the tail vein so that leukemia cells home to their natural target organs.
  5. Disease Monitoring: In L1210-luc studies, whole-animal bioluminescence is captured after intraperitoneal D-luciferin injection on a defined schedule; in unlabeled studies, body weight, abdominal distension, and clinical scores serve as the readouts, with disease typically declared by day 5–7.
  6. Treatment Administration: Compounds are dosed according to the client’s schedule by intraperitoneal, intravenous, oral, or subcutaneous routes, with body weight and general condition recorded throughout.
  7. Surveillance and Survival Endpoint: Animals are monitored daily; moribund mice are euthanized and the date recorded for Kaplan–Meier analysis.
  8. Terminal Organ Analysis: At endpoint, spleen, bone marrow, lymphoid organs, and peritoneal fluid are collected for ex vivo bioluminescence, flow cytometry, and histopathology to profile residual disease.
