TE-7-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: TE-7-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

TE-7-luc Orthotopic Mouse Model Service for Esophageal Cancer pairs one of the most storied esophageal cancer cell lines with luciferase-based tracking, placing tumors in the esophageal wall where they can be monitored non-invasively throughout a preclinical study. Alfa Cytology supports this model with provenance-verified cell sourcing, precision orthotopic surgery, scheduled bioluminescence imaging, and full histopathological endpoints—an authentication-first workflow designed for teams that treat cell identity as seriously as tumor biology.

Overview of TE-7-luc Orthotopic Mouse Model for Esophageal Cancer

TE-7 originates from the TE series, one of the earliest collections of esophageal cancer cell lines, established in Japan by Nishihira and colleagues in the late 1970s and early 1980s. The line occupies a special place in cell culture history: although long cataloged and studied as an esophageal adenocarcinoma model, STR profiling, mutation analysis, array-CGH, and xenograft histology demonstrated in 2007 that TE-7 is in fact a squamous cell carcinoma line genetically identical to TE-2, TE-3, TE-12, and TE-13. TE-7-luc is the firefly luciferase–expressing derivative of this squamous line, enabling tumor burden to be quantified by bioluminescence after D-luciferin injection.

Deployed as an orthotopic model, TE-7-luc cells are implanted into the esophageal wall of immunodeficient mice, where the tumor develops amid the organ’s native layers and drainage routes rather than in a subcutaneous pocket. Serial imaging then turns each animal into its own time course: photon output rises and falls with tumor cellularity, so response to a candidate agent can be read week by week instead of inferred from endpoint size alone. Decades of published work on the parental line—spanning EGFR and c-erbB signaling, p53 status, and LRP1B silencing—further supply a mechanistic context that helps investigators interpret orthotopic results against a well-mapped biological background.

Cell Line Information: TE-7-luc

Because TE-7’s history includes a widely cited misidentification, provenance documentation is central to any credible use of the line. Cellosaurus registers TE-7 as CVCL_9972 (ICLAC-00277), a male, Japanese-origin human esophageal squamous cell carcinoma; the original supplier entries at RCB and TKG have been discontinued, making verified stocks and STR confirmation especially important. The luciferase derivative carries the same squamous identity with an added imaging reporter. Documented features are summarized below.

Field Description
Cell Line Name TE-7-luc (parental line: TE-7; TE series member)
Cellosaurus Accession CVCL_9972; registered with the International Cell Line Authentication Committee as ICLAC-00277
True Histotype Esophageal squamous cell carcinoma (NCIt C4024); long misclassified as adenocarcinoma until Boonstra et al. (Cancer Research, 2007) established squamous identity
Identity Notes Genetically identical to TE-2, TE-3, TE-12, and TE-13; xenograft histology shows squamous differentiation; original RCB (RCB1893) and TKG distribution discontinued
Series Origin TE series established in Japan by Nishihira and colleagues (late 1970s–early 1980s); extensively described in J. Cancer Res. Clin. Oncol. (1993)
Donor Information Human (Homo sapiens); male; Japanese
Morphology & Growth Epithelial-like, adherent monolayer; grows as anchorage-dependent cultures with typical ESCC polygonal morphology
Culture Conditions RPMI 1640 with 10% fetal bovine serum; 37 °C, 5% CO2, humidified atmosphere
Biosafety Level BSL-1
Signaling Biology Frequently profiled for EGFR and c-erbB family expression; p53 status examined in multi-line functional assays; LRP1B silencing documented in ESCC studies including TE-series lines
Omics Resources Transcriptomic microarray and SNP-array data available; entries in CGH-DB, Progenetix, COSMIC, and the Cell Model Passport (SIDM00066)
Tumorigenicity Forms xenograft tumors in immunodeficient mice with squamous histology, as shown in the identity investigation of the TE series
Reporter Derivative TE-7-luc stably expresses firefly luciferase for quantitative bioluminescence imaging of tumor establishment, growth, and response
Common Applications Longitudinal preclinical efficacy studies, EGFR-pathway and differentiation research, metastasis observation, and studies requiring an authenticated ESCC background
Authentication Note Verify STR profile against the CVCL_9972 reference before study start; the line’s documented identity history makes supplier documentation essential

Our Services

Alfa Cytology built its TE-7-luc orthotopic service around a simple principle: a model is only as strong as the certainty behind its cells. Every project begins with STR confirmation against the registered TE-7 reference profile and an explicit provenance record, followed by luciferase verification, surgical implantation by an experienced microsurgery team, imaging sessions run on a fixed calendar, and endpoint pathology that re-confirms squamous identity. Clients receive the documentation trail alongside the data, which makes results easier to defend in internal reviews and manuscript submissions alike.

Workflow of TE-7-luc Orthotopic Mouse Model Construction

The TE-7-luc workflow is deliberately audit-friendly: identity, reporter function, surgery, and imaging each generate their own records before the next phase begins. A standard construction sequence looks like this:

  1. Provenance review and STR confirmation: Incoming TE-7-luc stocks are checked against supplier documentation, then STR-profiled against the CVCL_9972 reference before acceptance into the project.
  2. Reporter and quality testing: Luciferase output is measured in vitro, cultures are mycoplasma-tested, and passage number is capped to limit drift between the cell bank and the surgical cohort.
  3. Cohort-ready cell preparation: On the day of surgery, cells are harvested from log-phase cultures, counted, viability-checked, and concentrated into a small inoculum volume (typically around 1 × 105 cells in a 10 μL carrier such as basement membrane matrix).
  4. Anesthesia and orthotopic implantation: Immunodeficient mice are anesthetized with perioperative analgesia; the esophagus is exposed and the cell suspension is injected into the wall under magnification using a fine-gauge needle, avoiding the lumen.
  5. Closure and recovery: The abdominal layers are sutured, animals recover under warming with daily welfare scoring until feeding and weight trajectories normalize.
  6. Baseline and serial imaging: D-luciferin is administered and an initial bioluminescence scan documents take; subsequent scans on a fixed schedule generate longitudinal photon-flux growth curves per animal.
  7. Randomization and treatment execution: Animals are allocated to vehicle or treatment arms once signals fall within the pre-specified window, and dosing proceeds according to the approved design.
  8. Endpoint pathology and identity re-check: Tumors are excised for size recording, H&E staining, and IHC or molecular assays, with squamous morphology documented in the final report alongside the imaging dataset.

Why Choose Alfa Cytology?

A luciferase orthotopic study generates many data points, but only a few decide whether the result can be trusted. Alfa Cytology’s TE-7-luc service concentrates on those decisive points:

  • Registered-reference STR confirmation is performed before any animal work begins, directly addressing the line’s documented identity history.
  • Luciferase expression is verified in parallel with mycoplasma and passage controls, protecting signal quality across the study.
  • Magnification-guided wall implantation by practiced microsurgeons supports consistent tumor placement and take rates.
  • Fixed-interval imaging with standardized luciferin administration produces growth curves that are comparable across arms and time points.
  • Endpoint pathology explicitly documents squamous morphology, closing the loop between cell identity and in vivo observation.
  • Every record—from cell bank to final report—is assembled into a traceable documentation package suitable for internal audit.
