TE-1-luc Orthotopic Mouse Model Service for Esophageal Cancer

Fig 1: TE-1-luc Orthotopic Mouse Model for Esophageal Cancer preclinical research.

The TE-1-luc orthotopic mouse model transplants firefly luciferase-tagged esophageal squamous carcinoma cells into the esophageal wall of immunodeficient mice, creating a surgically precise, optically quantifiable tumor system that mirrors the anatomical reality of human esophageal cancer. Alfa Cytology provides this model as a comprehensive preclinical package: authenticated TE-1-luc reporter banks, aseptic orthotopic microsurgery, serial bioluminescence imaging sessions, randomized dosing cohorts, and terminal necropsy with full histopathology—so sponsors obtain longitudinally quantified, decision-grade datasets without assembling their own surgical and imaging capability.

Overview of TE-1-luc Orthotopic Mouse Model for Esophageal Cancer

TE-1 holds a special place in esophageal cancer research: established in 1979 by Nishihira and colleagues, it was among the very first human ESCC lines brought into continuous culture, and it has served the field ever since. The line is genetically distinctive—it carries a missense TP53 mutation at codon 272 (p.Val272Met), one of the classic contact-site mutants studied for gain-of-function effects, and it is microsatellite-stable with a relatively low mutation burden. Two practical characteristics shape how TE-1 behaves in orthotopic work. First, its reported doubling time of about 60 hours is considerably slower than most modern ESCC lines, which produces a more deliberate, clinically plausible tumor growth tempo and is well suited to studies where realistic disease kinetics matter. Second, early biochemical work characterized TE-1 as expressing fewer but higher-affinity epidermal growth factor receptors—an unusual receptor configuration that continues to make the line a reference point in EGFR-pathway studies.

Adding stable firefly luciferase converts TE-1 into an orthotopic platform suited to modern imaging-driven study designs. After implantation into the esophageal wall, photon flux measured following D-luciferin administration tracks viable tumor burden noninvasively across the entire study, revealing treatment-induced growth delays, partial regressions, and relapses that caliper-based models could never capture at this anatomical site. The TE-1 background is further supported by decades of literature covering cytogenetics, microRNA regulation, EphA2-angiogenesis biology, and radiation response—including the existence of a characterized radioresistant derivative, TE-1R—giving sponsors an unusually deep comparator base for both in vitro mechanism work and in vivo efficacy testing. Orthotopic TE-1-luc studies are therefore applied to radiosensitizer screening, targeted agent evaluation, and combination regimen optimization in preclinical pipelines.

Cell Line Information: TE-1-luc

TE-1-luc is the firefly luciferase reporter derivative of TE-1, a human ESCC line established in 1979 from a primary esophageal tumor of a 58-year-old Japanese man. The parental line is deposited in major repositories (including RIKEN RCB1894, BCRJ 0371, and CLS 305060), is represented in the Cancer Cell Line Encyclopedia, COSMIC, and DepMap compendia, and carries an extensive public omics footprint including whole-exome, RNA-seq, methylation, and quantitative proteomics data. Reporter labeling does not alter the parental growth characteristics while adding a D-luciferin-dependent optical signal usable in vivo. Principal characteristics are summarized below.

Our Services

Orthotopic esophageal work with a slow-growing heritage line like TE-1 rewards patience and punishes inconsistency—every unlogged passage, every imprecise implantation, compounds over a long study. Alfa Cytology absorbs that complexity for sponsors: we maintain low-passage, STR-authenticated, mycoplasma-free TE-1-luc banks with documented passage bookkeeping; perform the esophageal wall implantation under aseptic conditions with defined inocula; execute scheduled bioluminescence reads on calibrated instrumentation; run randomized dosing arms exactly as specified; and close each study with ex vivo organ imaging, tumor metrics, and H&E/immunohistochemistry—assembled into a single audit-ready report.

Workflow of TE-1-luc Orthotopic Mouse Model Construction

Construction follows a validated sequence from reporter cell qualification through surgical orthotopic delivery to longitudinal optical follow-up and terminal histology. Because TE-1 grows more slowly than most ESCC lines, imaging cadence and study duration are planned accordingly; inoculum size—commonly 2 × 106 cells in a small Matrigel-containing volume—and endpoint criteria are fixed with each sponsor at study design.

  1. Reporter cell bank release: TE-1-luc vials are revived, tested negative for mycoplasma, authenticated by STR profiling, and validated for luciferase signal linearity across the working cell range.
  2. Inoculum preparation: Log-phase cells are harvested, counted, and mixed with Matrigel or an equivalent carrier to anchor the bolus at the intended injection site.
  3. Animal preparation: BALB/c nude mice complete quarantine and acclimatization and are randomized into cohorts balanced for body weight.
  4. Anesthesia and surgical exposure: Under general anesthesia with perioperative analgesia, a small upper abdominal incision exteriorizes the stomach and exposes the distal esophagus.
  5. Orthotopic implantation: A fine-gauge needle deposits the cell suspension into the esophageal wall; the transient intramural bleb confirms correct placement before the incision is closed in layers.
  6. Postoperative care: Animals recover on warming pads and are monitored daily for body weight, feeding, and incision integrity until full recovery.
  7. Serial bioluminescence imaging: Following intraperitoneal D-luciferin injection, whole-body photon flux is quantified at scheduled intervals, charting engraftment, the slow steady growth characteristic of TE-1, and response to treatment.
  8. Terminal analysis: At protocol endpoints, mice are euthanized; the esophagus, regional lymph nodes, and organs are imaged ex vivo, then processed for tumor measurement, H&E, and immunohistochemistry.

Case Study-TE-1-luc Orthotopic Mouse Model Development

TE-1-luc orthotopic studies are an established part of Alfa Cytology’s esophageal cancer model portfolio. In a representative preclinical engagement, our team prepared authenticated reporter cells, completed orthotopic implantation in immunodeficient mice, and confirmed reproducible engraftment through serial bioluminescence imaging across the extended growth window characteristic of this line. Randomized treatment arms were supported with scheduled imaging reads, and terminal histopathology verified orthotopic tumor location and preserved esophageal wall architecture. Representative imaging and histology datasets from this program are available to sponsors upon request.
